    STABILITY-INDICATING RP-HPLC METHOD FOR SIMULTANEOUS ESTIMATION OF SOFOSBUVIR, VELPATASVIR, AND VOXILAPREVIR IN BULK AND TABLET DOSAGE FORMS

    Objective: The present study was aimed to develop a novel, simple, rapid accurate and precise, stability-indicating reversed-phase high-performance liquid chromatography method for the simultaneous estimation of sofosbuvir, velpatasvir, and voxilaprevir in bulk and tablet dosage forms. Methods: The chromatographic elution was achieved in isocratic mode using the combination of sofosbuvir, velpatasvir, and voxilaprevir in the ratio of acetonitrile and water (65:35%v/v) using a Phenomenex C18 column which has specification (150 × 4.6 mm, 5 μ particle size) and the flow rate of 1.0 ml/min and wavelength (ultraviolet) detection at 220 nm. Results: The retention time obtained for sofosbuvir, velpatasvir, and voxilaprevir was 2.213 min, 2.568 min, and 2.917 min, respectively. Sofosbuvir, velpatasvir, and voxilaprevir and their combination drug product were exposed to acidic, alkali, thermal, photolytic, and oxidative stress conditions. The current method was validated according to the ICH guidelines for accuracy, precision, linearity, specificity, and sensitivity. Conclusion: The method developed is more sensitive, accurate, precise, and robust then the methods reported earlier. Retention time and run time were decreased; hence, the method is economical simple and precise. Forced degradation studies indicated for the suitability of the method for stability studies of sofosbuvir, velpatasvir, and voxilaprevir. The proposed method can be used for routine quality control analysis test in pharmaceutical industries

    Supramolecular chemistry, other way called as intermolecular chemistry disclose the relationship of molecules with environment. It exploits while exposing the physicochemical phenomina that happens when two like or unlike molecules/ions/systems contact each other. Drug action involve the target recognition process and response triggered by the intermolecular complex of drug and target. Drug design therefore require in depth study of intermolecular forces that exist between drug and target. Formulation of the drug or Active Pharmaceutical Ingredient (API) is also regulated by these forces. Compatibility and incompatibility in formulations are nothing but of the effect of the intermolecular forces on physical behavior of systems.     Ant species richness and diversity in some selected localities of Western Ghats

    We have sampled ants from twelve different localities in the Uttara Kannada district of Karnataka, India using both a package of standardized trapping methods as well as by intensive all-out-search. The relative efficacies of trapping versus all-out-search are compared. Different methods of trapping are also compared among themselves. A check list of all ants seen by all methods put together, identified up to the genus level is provided. Different localities are compared by the similarity and diversity 'of their ant fauna. A weak but statistically significant positive correlation between ant species diversity. and plant species diversity is demonstrated. This report provides the first estimates of the diversity and abundance of ants for any forest habitat of India

    Large number of population suffers from low back pain. Chronic low back pain (CLBP) prevalence increases linearly from the third decade of life on, until the 60 years of age, being more prevalent in women. Sciatica is the most common debilitating condition causes CLBP. Radiating leg pain and related disabilities are the observed in sciatica. Nearly 40% people experience sciatic pain at some point in their life. The diagnosis of sciatica and its management varies considerably within and between countries. Conventional Medicine and surgery are widely used in the management of sciatica. There is radical rise in the use of Complementary and Alternative Medicine (CAM) in patients with sciatica. Ayurveda is one of the widely used CAM in the recent past. Sciatica resembles the disease Gridharsi of Ayurveda. Gridharsi is one among the Vataja nanatmaja vyadhi, where dysfunction of Vata affect gridharsi nadi characterized by low back pain radiating to lower limbs, stiffness and pricking type of pain. It starts from kati- prishta (pelvic region and Lumbosacral) radiating to jangha paada (Thigh, Feets) with impairment of lifting the leg. The gait of the person is very similar to vulture (Gridhra) hence the name is given as Gridharsi. In this article, attempt has been made to review the Ayurvedic classics text and related literatures to understand the disease Gridharsi with emphasis on its samprapti on the basis of Kriyakala and role of Shodhana and Shamana chikitsa in the management of Gridharsi

    Doctors on duty: the real-life heroes during pandemic: Your opinion matters!!!

    Background: Data for front-line health-care workers, risk of disease is limited so as the resources available. Although inspiring stories on doctors fighting against COVID-19-19 were covered we are still unaware of the challenges at ground level. The present study throws light on it and provides way through towards resolving the issues. Objectives:1. To assess the perceptions of the doctors towards the process of COVID-19-19 crisis management. 2.To identify the challenges and recommend solutions based on their views, suggestions. Methods: A cross sectional study on perception of doctors (94) involved in COVID-19 duty was conducted during July to September 2020, with the help of pre-validated semi-structured questionnaire through online google survey form. The questionnaire had demographic details, views on before, during and after the COVID-19 duty along with 4 open ended questions on their good and bad experiences, problems & suggestions. Descriptive statistics for quantitative & thematic analysis for qualitative data was used. Results: Nonresponse rate was 26%, mean age 27.55 year. Majority were from clinical departments (78%). 42% felt that training was proper, 60% adequate PPE kit, 52.6% were aware of management strategy, 86% stressed,82% found difficulties in communication. 42% were provided proper quarantine facility yet, 58% faced discrimination. Few themes emerged under challenges like lack of resources & training, stress & burnout, for which patient sensitization, refresher training, psychological & administrative support was suggested. Conclusion: Although COVID-19 pandemic has impacted life of doctors in either way, the challenges could easily be tackled by the recommended simple solutions

    This article presents the highlights of the ERS Lung Science Conference 2022, including a session organised by the Early Career Member Committee (ECMC) dedicated to career development https://bit.ly/3tarCXc Every year, the European Respiratory Society (ERS) organises the Lung Science Conference (LSC), in Estoril, to discuss basic and translational science. The topic of the 20th LSC was “Mucosal immunology of the lung: balancing protective immunity and chronic inflammation”. This was the first time that the LSC was organised as a hybrid congress with both in person and online attendance. In addition to an outstanding scientific programme, the LSC provides excellent opportunities for career development and inclusion of early career members (ECMs). All scientific and poster sessions are chaired by an ECM who is paired with a senior faculty to allow ECMs to become acquainted with session chairing, and there is a session organised by the Early Career Member Committee (ECMC) dedicated to career development. Moreover, travel bursaries are made available to abstract authors, and all bursary recipients and first-time attendees are invited to take part in a mentorship lunch.     Early alveolar macrophage response and IL-1R-dependent T cell priming determine transmissibility of Mycobacterium tuberculosis strains

Publisher Copyright: © 2022, The Author(s).Mechanisms underlying variability in transmission of Mycobacterium tuberculosis strains remain undefined. By characterizing high and low transmission strains of M.tuberculosis in mice, we show here that high transmission M.tuberculosis strain induce rapid IL-1R-dependent alveolar macrophage migration from the alveolar space into the interstitium and that this action is key to subsequent temporal events of early dissemination of bacteria to the lymph nodes, Th1 priming, granulomatous response and bacterial control. In contrast, IL-1R-dependent alveolar macrophage migration and early dissemination of bacteria to lymph nodes is significantly impeded in infection with low transmission M.tuberculosis strain; these events promote the development of Th17 immunity, fostering neutrophilic inflammation and increased bacterial replication. Our results suggest that by inducing granulomas with the potential to develop into cavitary lesions that aids bacterial escape into the airways, high transmission M.tuberculosis strain is poised for greater transmissibility. These findings implicate bacterial heterogeneity as an important modifier of TB disease manifestations and transmission.publishersversionpublishe
