    A Link Between Plant Stress and Hydrodynamics? Indications From a Freshwater Macrophyte

    Live plants are increasingly used in hydraulic laboratories to investigate flow-vegetation interactions. In such experiments, they are often exposed to stressful handling and storage that can cause strong physiological responses and modifications in plant biomechanics. Little is known about the potential effect of these impacts on the performance of plants during hydraulic experiments. In this multidisciplinary study with a freshwater macrophyte (Potamogeton natans) we assess whether the duration and the conditions in which plants are stored in a laboratory prior to testing can impact plant stress, biomechanics and hydrodynamics, and quantify this impact. Plant stress was evaluated using chlorophyll fluorescence analysis (and the maximum quantum yield of photosystem II as specific indicator). Plant hydrodynamics were assessed using the drag coefficient calculated from drag force measurements at two flow scenarios. The results show that different plant handling/storage procedures can have a significant impact on plant hydrodynamics even within a short time frame, with a variation of the mean drag coefficient of approximately 30% across groups, which is comparable to the variation found across different species of freshwater macrophytes in previous studies. Plants with the highest level of stress were also characterized by the lowest drag coefficient across the groups considered, suggesting a potential link between plant stress and hydrodynamics

    Für die starke Zunahme von Infektionen durch opportunistische Pilze sind vor allem prädisponierende Faktoren verantwortlich,welche die natürliche Abwehr des Wirtes abschwächen.Trotzdem müssen pathogene Pilze, wie die medizinisch bedeutende Hefe Candida albicans, über Virulenzfaktoren verfügen, die dem Mikroorganismus das Überleben auf und in einem Wirt sichern oder das Vordringen zu tieferen Geweben und Organen ermöglichen.Das Ziel der Forschungsprojekte der Nachwuchsgruppe „Pathogenitätsfaktoren bei Pilzinfektionen“ am Robert Koch-Institut (RKI) ist, solche Faktoren von Pilzen zu identifizieren und zu analysieren, von denen vermutet wird, dass sie bei einer Infektion eine wichtige Rolle spielen.Damit sollen nicht nur die Pathogenitätsmechanismen besser verstanden werden, sondern auch Ansatzpunkte für neue Medikamente gefunden werden.Schließlich gilt es,aufgrund der zunehmenden Resistenzen gegenüber den wenigen zur Verfügung stehenden Antimykotika, die Wirkungsweise existierender Medikamente zu verstehen

    Overcoming bortezomib resistance in human B cells by anti-CD20/rituximab-mediated complement-dependent cytotoxicity and epoxyketone-based irreversible proteasome inhibitors

    UNLABELLED: BACKGROUND: In clinical and experimental settings, antibody-based anti-CD20/rituximab and small molecule proteasome inhibitor (PI) bortezomib (BTZ) treatment proved effective modalities for B cell depletion in lymphoproliferative disorders as well as autoimmune diseases. However, the chronic nature of these diseases requires either prolonged or re-treatment, often with acquired resistance as a consequence. METHODS: Here we studied the molecular basis of acquired resistance to BTZ in JY human B lymphoblastic cells following prolonged exposure to this drug and examined possibilities to overcome resistance by next generation PIs and anti-CD20/rituximab-mediated complement-dependent cytotoxicity (CDC). RESULTS: Characterization of BTZ-resistant JY/BTZ cells compared to parental JY/WT cells revealed the following features: (a) 10-12 fold resistance to BTZ associated with the acquisition of a mutation in the PSMB5 gene (encoding the constitutive ββ5 proteasome subunit) introducing an amino acid substitution (Met45Ile) in the BTZ-binding pocket, (b) a significant 2-4 fold increase in the mRNA and protein levels of the constitutive ββ5 proteasome subunit along with unaltered immunoproteasome expression, (c) full sensitivity to the irreversible epoxyketone-based PIs carfilzomib and (to a lesser extent) the immunoproteasome inhibitor ONX 0914. Finally, in association with impaired ubiquitination and attenuated breakdown of CD20, JY/BTZ cells harbored a net 3-fold increase in CD20 cell surface expression, which was functionally implicated in conferring a significantly increased anti-CD20/rituximab-mediated CDC. CONCLUSIONS: These results demonstrate that acquired resistance to BTZ in B cells can be overcome by next generation PIs and by anti-CD20/rituximab-induced CDC, thereby paving the way for salvage therapy in BTZ-resistant disease

    Role of Pseudomonas aeruginosa Peptidoglycan-Associated Outer Membrane Proteins in Vesicle Formation

    Gram-negative bacteria produce outer membrane vesicles (OMVs) that package and deliver proteins, small molecules, and DNA to prokaryotic and eukaryotic cells. The molecular details of OMV biogenesis have not been fully elucidated, but peptidoglycan-associated outer membrane proteins that tether the outer membrane to the underlying peptidoglycan have been shown to be critical for OMV formation in multiple Enterobacteriaceae. In this study, we demonstrate that the peptidoglycan-associated outer membrane proteins OprF and OprI, but not OprL, impact production of OMVs by the opportunistic pathogen Pseudomonas aeruginosa. Interestingly, OprF does not appear to be important for tethering the outer membrane to peptidoglycan but instead impacts OMV formation through modulation of the levels of the Pseudomonas quinolone signal (PQS), a quorum signal previously shown by our laboratory to be critical for OMV formation. Thus, the mechanism by which OprF impacts OMV formation is distinct from that for other peptidoglycan-associated outer membrane proteins, including OprIclose17181

    Ageing exacerbates damage of systemic and salivary neutrophils from patients presenting Candida-related denture stomatitis

    <p>Abstract</p> <p>Background</p> <p>Ageing leads to a decline in the function of the immune system, increasing the body's susceptibility to infections through the impairment of T-cells, macrophages, neutrophils and dendritic cells Denture stomatitis is a primary oral disease affecting elderly denture wearers. The major etiologic factor involved in this pathology is the infection by <it>Candida albicans</it>, an opportunistic pathogen that causes local and disseminated diseases in immunosuppressed humans. Neutrophils play a critical role in the immune response against <it>C. albicans </it>and are continually present in the salivary fluid and in the blood. The aim of this study was to determine ageing-related changes in salivary and blood neutrophils and their potential implications in <it>Candida</it>-related denture stomatitis.</p> <p>Results</p> <p>Our results showed a lower number of neutrophils in the saliva from patients presenting <it>Candida</it>-related denture stomatitis in comparison to their matched controls. Furthermore, fewer neutrophils were isolated from the saliva of aged control individuals in comparison to matched younger subjects. CXCR1, CD62L and CD11b expression were significantly greater on systemic neutrophils from younger control individuals. Elderly individuals showed more apoptotic salivary neutrophils and lower GM-CSF levels than younger ones, regardless of the occurrence of <it>Candida </it>infection. On the other hand, CXCL-8 concentrations were higher in the saliva from elderly individuals. Besides, TNF-α was detected at elevated levels in the saliva from infected elderly subjects. Salivary neutrophils from elderly and young patients presented impaired phagocytic activity against <it>C. albicans</it>. However, just systemic neutrophils from elderly showed decreased phagocytosis when compared to the younger ones, regardless of the occurrence of infection. In addition, neutrophils from aged individuals and young patients presented low fungicidal activity.</p> <p>Conclusion</p> <p>The data suggests that the <it>Candida </it>related-denture stomatitis is associated to neutrophils function deficiency, and ageing drastically appears to alter important characteristics of such cells, facilitating the establishment of this infection.</p
