    Primate Primordial Germ Cells Acquire Transplantation Potential by Carnegie Stage 23.

    Primordial germ cells (PGCs) are the earliest embryonic progenitors in the germline. Correct formation of PGCs is critical to reproductive health as an adult. Recent work has shown that primate PGCs can be differentiated from pluripotent stem cells; however, a bioassay that supports their identity as transplantable germ cells has not been reported. Here, we adopted a xenotransplantation assay by transplanting single-cell suspensions of human and nonhuman primate embryonic Macaca mulatta (rhesus macaque) testes containing PGCs into the seminiferous tubules of adult busulfan-treated nude mice. We discovered that both human and nonhuman primate embryonic testis are xenotransplantable, generating colonies while not generating tumors. Taken together, this work provides two critical references (molecular and functional) for defining transplantable primate PGCs. These results provide a blueprint for differentiating pluripotent stem cells to transplantable PGC-like cells in a species that is amenable to transplantation and fertility studies

    Now-a- day\u27s wireless control of appliance is in practice, because of its advantage like no wire is needed, multiple controlling is less complicated. The controlling range of wireless doorbell is 100M. As the wireless doorbell is wireless based project, it is divided into two major section i.e. transmitter and receiver. Transmitter: This transmitter section is designed around oscillator transistor (BF194B) T2 followed by two transistor (BC148) T1 and T3. Transistor T2 generate special radio frequency determined by trimmer (variable capacitor) and coil. Transistor T1 is used as pulse generator. The output from transistor T3 is given to emitter of transistor T2 in order to get radio frequency from its collector. Trimmer VC1 is used to adjust the transmitter frequency. Receiver: The receiver section is further divided into two main sections i.e. RF amplifier section and bell trigger section. An aerial is used to receive the transmitted frequency from remote which is further amplified by amplifier and trigger circuit. The whole receiver circuit utilizes seven transistors

    Lineage dynamics of murine pancreatic development at single-cell resolution.

    Organogenesis requires the complex interactions of multiple cell lineages that coordinate their expansion, differentiation, and maturation over time. Here, we profile the cell types within the epithelial and mesenchymal compartments of the murine pancreas across developmental time using a combination of single-cell RNA sequencing, immunofluorescence, in situ hybridization, and genetic lineage tracing. We identify previously underappreciated cellular heterogeneity of the developing mesenchyme and reconstruct potential lineage relationships among the pancreatic mesothelium and mesenchymal cell types. Within the epithelium, we find a previously undescribed endocrine progenitor population, as well as an analogous population in both human fetal tissue and human embryonic stem cells differentiating toward a pancreatic beta cell fate. Further, we identify candidate transcriptional regulators along the differentiation trajectory of this population toward the alpha or beta cell lineages. This work establishes a roadmap of pancreatic development and demonstrates the broad utility of this approach for understanding lineage dynamics in developing organs

    Influence of wind speed on surface layer stability and turbulent fluxes over southern Indian peninsula station

    Surface to atmosphere exchange has received much attention in numerical weather prediction models. This exchange is defined by turbulent parameters such as frictional velocity, drag coefficient and heat fluxes, which have to be derived experimentally from high-frequency observations. High-frequency measurements of wind speed, air temperature and water vapour mixing ratio (eddy covariance measurements), were made during the Integrated Ground Observation Campaign (IGOC) of Cloud Aerosol Interaction and Precipitation Enhancement Experiment (CAIPEEX) at Mahabubnagar, India (16∘44′N, 77∘59′E) in the south-west monsoon season. Using these observations, an attempt was made to investigate the behaviour of the turbulent parameters, mentioned above, with respect to wind speed. We found that the surface layer stability derived from the Monin–Obukhov length scale, is well depicted by the magnitude of wind speed, i.e., the atmospheric boundary layer was under unstable regime for wind speeds >4 m s−1; under stable regime for wind speeds <2 m s−1 and under neutral regime for wind speeds in the range of 2–3 m s−1. All the three stability regimes were mixed for wind speeds 3–4 m s−1. The drag coefficient shows scatter variation with wind speed in stable as well as unstable conditions

    Effect of rice husk biochar, carpet waste, farm yard manure and plant growth promoting rhizobium on the growth and yield of rice (Oryza sativa)

    The present investigation was aimed for improving growth and yield of crop using waste products of differ-ent activities and also useful in ecological stability of soil environment. This objective is not only an economic option for poor farmer but also an effective strategy for increasing yield. The experiment was conducted in the organic farm-ing plot of the Institute of Agricultural Sciences, BHU, Varanasi during kharif season of rice crop in 2014. The field experiment was laid out in randomized block design (RBD) with 10 treatments and three replications. Application of graded level of biochar, carpet waste farm yard manure (FYM) and plant growth promoting rhizobium (PGPR) was found to significantly enhance the grain and straw yield of rice by 57.70% and 56.08% over control, respectively

    In vitro screening of white Jute (Corchorus capsularis L) against salinity stress

    The environmental stress such as salinity of soil or water is serious threat for field crops in the world, especially in arid and semiarid regions. To study salinity stress on jute (Corchorus capsularis), an experiment was laid out in factorial randomized block design keeping nine capsularis varieties (JRC-698, JRC-321, JRC-517, JRC-7447, JBC-5, JRC-212, JRC-80, JRC-532 and UPC-94) as one factor and four levels of salt concentrations i.e. control (Distilled H2O), 100, 160, 240 and 300 mM NaCl with three replications. Adverse effect of salinity increased with increasing concentration of sodium chloride. Among nine jute varieties the highest germination percentage (89.5%), root length (0.6 cm), shoot length (1.2 cm), fresh weight (93 mg) and dry weight (9.5 mg) of seedling were observed in ‘JRC-698’ at salt concentration at 160 mM NaCl as compared with other salt concentration. All growth parameters were recorded higher in control than other treatments. All growth parameters were decreased up to 240 mM NaCl concentration and it was completely inhibited at 300 mM NaCl. Variety ‘UPC-94’ was the most susceptible to salinity stress. Hence, white jute varieties, JRC-698 and JRC-517 can be grown at 160 mM NaCl concentration

    Inhaled PGE1 in neonates with hypoxemic respiratory failure: two pilot feasibility randomized clinical trials.

    BackgroundInhaled nitric oxide (INO), a selective pulmonary vasodilator, has revolutionized the treatment of neonatal hypoxemic respiratory failure (NHRF). However, there is lack of sustained improvement in 30 to 46% of infants. Aerosolized prostaglandins I2 (PGI2) and E1 (PGE1) have been reported to be effective selective pulmonary vasodilators. The objective of this study was to evaluate the feasibility of a randomized controlled trial (RCT) of inhaled PGE1 (IPGE1) in NHRF.MethodsTwo pilot multicenter phase II RCTs are included in this report. In the first pilot, late preterm and term neonates with NHRF, who had an oxygenation index (OI) of ≥15 and &lt;25 on two arterial blood gases and had not previously received INO, were randomly assigned to receive two doses of IPGE1 (300 and 150 ng/kg/min) or placebo. The primary outcome was the enrollment of 50 infants in six to nine months at 10 sites. The first pilot was halted after four months for failure to enroll a single infant. The most common cause for non-enrollment was prior initiation of INO. In a re-designed second pilot, co-administration of IPGE1 and INO was permitted. Infants with suboptimal response to INO received either aerosolized saline or IPGE1 at a low (150 ng/kg/min) or high dose (300 ng/kg/min) for a maximum duration of 72 hours. The primary outcome was the recruitment of an adequate number of patients (n = 50) in a nine-month-period, with fewer than 20% protocol violations.ResultsNo infants were enrolled in the first pilot. Seven patients were enrolled in the second pilot; three in the control, two in the low-dose IPGE1, and two in the high-dose IPGE1 groups. The study was halted for recruitment futility after approximately six months as enrollment targets were not met. No serious adverse events, one minor protocol deviation and one pharmacy protocol violation were reported.ConclusionsThese two pilot RCTs failed to recruit adequate eligible newborns with NHRF. Complex management RCTs of novel therapies for persistent pulmonary hypertension of the newborn (PPHN) may require novel study designs and a longer period of time from study approval to commencement of enrollment.Trial registrationCLINICALTRIALS.GOV: Pilot one: NCT number: 00598429 registered on 10 January 2008.
