    Die Positionierung der Nationalratskandidierenden 2007 : eine explorative Analyse der Antworten auf die Fragen der Online-Wahlhilfe smartvote

    Die Studie prĂ€sentiert erste explorative Auswertungen zu den Antworten von ĂŒber 2600 Kandidierenden im Rahmen der Nationalratswahlen 2007 auf den umfangreichen Fragenkatalog der Online-Wahlhilfeplattform « smartvote ». Untersucht wird, wie die Fragen von der Gesamtheit der Kandidierenden beantwortet wurden, welche Themen umstritten waren und wie sich die Kandidierenden von den GewĂ€hlten unterscheiden. Schwergewichtig beschĂ€ftigt sich die Analyse mit den Kandidierenden der vier grössten Parteien SVP, FDP, CVP und SP sowie der GrĂŒnen. Welche Positionen nehmen sie ein ? Wie geschlossen prĂ€sentieren sie sich in den einzelnen Fragen ? Wo bestehen grosse Gemeinsamkeiten, wo erheblicheUnterscheide und wie sind die Möglichkeiten der einzelnen Parteien einzuschĂ€tzen, sich politisch zu profilieren ? Schliesslich wird ĂŒberprĂŒft, wie gut die Kandidierenden zu ihrer eigenen Partei passen und welche Aussagen sich ĂŒber die ZusammenhĂ€nge zwischen der Abweichung von der Parteilinie und der Herkunft von erhofften Panaschierstimmen machen lassen. Lors des Ă©lections au Conseil national en octobre 2007, plus de 2600 candidats ont rĂ©pondu au questionnaire concernant l'outil de soutien au vote en ligne « smartvote ». Ce cahier de l'IDHEAP prĂ©sente les rĂ©sultats d'une premiĂšre Ă©tude exploratoire, basĂ©e sur les rĂ©ponses des candidats au questionnaire de « smartvote ». Les auteurs analysent les questions et les thĂšmes controversĂ©s. Ils se sont Ă©galement intĂ©ressĂ©s aux diffĂ©rences de prises de position entre les Ă©lus et les candidats.     School choice is on the rise in many states. Since the start of the new millennium, many states have launched or expanded private school choice options, permitted and expanded independently operated public charter schools, eased restrictions on homeschooling, and enacted policies that allow and encourage various forms of public school choice. One thing that is not on the rise, unfortunately, is average student scores on the National Assessment of Education Progress (NAEP). Student performance on the assessments, typically called “The Nation’s Report Card,” were flat from 2001 until 2015 and have dropped slightly in both 2017 and 2019

    Many hydrographic and oceanographic agencies have moved or are moving towards gridded bathymetric products. However, there is no accepted format to allow these grids to be exchanged while maintaining data and metadata integrity. This paper describes the Open Navigation Surface (ONS) Project, which aims to fill this gap. The ONS Project is an open-source software project designed to provide a freely available, portable source-code library to encapsulate gridded bathymetric surfaces with associated uncertainty values. The data file format is called a Bathymetric Attributed Grid (BAG). The BAG is developed and maintained by the ONS Working Group (ONSWG), and the source code is available via the ONS websit

    Deoxyuridine triphosphatase (dUTPase) expression and sensitivity to the thymidylate synthase (TS) inhibitorD9331

    Uracil DNA misincorporation and misrepair of DNA have been recognized as important events accompanying thymidylate synthase (TS) inhibition. dUTPase catalyses the hydrolysis of dUTP to dUMP, thereby maintaining low intracellular dUTP. We have addressed the relationship between dUTPase expression and cellular sensitivity to TS inhibition in four human lung tumour cell lines. Sensitivity (5-day MTT assay) to the growth inhibitory effects of the non-polyglutamatable, specific quinazoline TS inhibitor ZD9331, varied up to 20-fold (IC 50 3–70 nM). TS protein expression correlated with TS activity (r2= 0.88 P= 0.05). Intracellular concentrations of drug following exposure to ZD9331 (1 ÎŒM, 24 h) varied by ~2-fold and dTTP pools decreased by > 80% in all cell lines. No clear associations across the cell lines between intracellular drug concentrations, TS activity/expression, or TTP depletion could be made. dUTPase activity varied 17-fold and correlated with dUTPase protein expression (r2= 0.94 P= 0.03). There was a striking variation in the amount of dUTP formed following exposure to ZD9331 (between 1.3 and 57 pmole 10–6cells) and was in general inversely associated with dUTPase activity. A large expansion in the dUTP pool was associated with increased sensitivity to a 24-h exposure to ZD9331 in A549 cells that have low dUTPase activity/expression. dUTPase expression and activity were elevated (approximately 3-fold) in two variants of a human lymphoblastoid cell line with acquired resistance to TS inhibitors, further suggesting an important role for this enzyme in TS inhibited cells. © 2000 Cancer Research Campaig

    Detection and characterization of hydraulically active fractures in a carbonate aquifer: results from self-potential, temperature and fluid electrical conductivity logging in the Combioula hydrothermal system in the southwestern Swiss Alps

    A geophysical and geochemical study has been conducted in a fractured carbonate aquifer located at Combioula in the southwestern Swiss Alps with the objective to detect and characterize hydraulically active fractures along a 260-m-deep borehole. Hydrochemical analyses, borehole diameter, temperature and fluid electrical conductivity logging data were integrated in order to relate electrokinetic self-potential signals to groundwater flow inside the fracture network. The results show a generally good, albeit locally variable correlation of variations of the self-potential signals with variations in temperature, fluid electrical conductivity and borehole diameter. Together with the hydrochemical evidence, which was found to be critical for the interpretation of the self-potential data, these measurements not only made it possible to detect the hydraulically active fractures but also to characterize them as zones of fluid gain or fluid loss. The results complement the available information from the corresponding litholog and illustrate the potential of electrokinetic self-potential signals in conjunction with temperature, fluid electrical conductivity and hydrochemical analyses for the characterization of fractured aquifers, and thus may offer a perspective for an effective quantitative characterization of this increasingly important class of aquifers and geothermal reservoir

    Photodynamic diagnosis of breast tumours after oral application of aminolevulinic acid

    Photodynamic diagnosis is of increasing interest for diagnosis in oncology. It is based on a more intense incorporation of a fluorescent dye in tumours compared to normal tissue. As a feasibility study we investigated the effectiveness of oral application of 5-aminolevulinic acid for photodynamic diagnosis of human primary mammary tumours. The study included 16 patients with palpable breast tumours. Aminolevulinic acid was administered at a concentration of 40 mg kg−1bodyweight 150–420 min prior to tumourectomy. Intraoperatively blue light (405 nm) was applied to the operation site. Sections of the excised tumour and some lymph nodes were prepared and analysed with a fluorescent microscope. All primary mammary tumour tissues showed significantly higher fluorescence intensity than surrounding normal mammary tissue. Fluorescence of the mammary tumours could also be discriminated macroscopically and intraoperatively. Fluorescence intensity in nonmetastatic lymph node tissue was higher in 2 out of 3 patients than in primary tumour tissue. By photodynamic diagnosis using aminolevulinic acid we were able to reliably distinguish primary mammary tumours from normal mammary tissue microscopically and macroscopically in all our patients. We suggest that photodynamic diagnosis with aminolevulinic acid for breast tumours should be further investigated and developed for intraoperative use and may well be a simple tool for better intraoperative diagnosis and recognition of tumour margins.     Evidenzbasierte Therapie des Raynaud-Syndroms

    Zusammenfassung: Das Raynaud-Syndrom ist mit einer PrĂ€valenz von 3-5% ein hĂ€ufiges klinisches Problem. Dennoch ist die Wirkung der meisten Therapiemöglichkeiten nur unzureichend durch kontrollierte Studien belegt. Zu den Therapien mit höherem Evidenzgrad gehört der Kalziumantagonist Nifedipin, fĂŒr den in Metaanalysen sowohl bei primĂ€rem als auch bei sekundĂ€rem Raynaud-Syndrom eine verbesserte periphere Durchblutung sowie eine Abnahme der Frequenz und des Schweregrades der Raynaud-Attacken nachgewiesen werden konnte. Ähnliches gilt fĂŒr intravenös appliziertes Iloprost in der Therapie des sekundĂ€ren Raynaud-Syndroms bei systemischer Sklerose. Intravenös verabreichtes Iloprost verbessert darĂŒber hinaus das Abheilen von Fingerkuppenulzera bei Patienten mit systemischer Sklerose. Vielversprechende TherapieansĂ€tze stellen Angiotensin-II-Rezeptor-1-Antagonisten (Losartan), die Kalziumantagonisten Felodipin und Amlodipin, Serotonin-Reuptake-Hemmer (Fluoxetin) und Phosphodiesterase-V-Hemmer (Sildenafil, Vardenafil) dar, die sich in kontrollierten Einzelstudien als wirksam erwiesen haben. Jedoch fehlen Erfahrungen mit grĂ¶ĂŸeren Patientenzahlen und lĂ€ngeren Anwendungszeiten, um diese Therapiemöglichkeiten abschließend zu beurteile

    Optimization of loading protocols for tissue engineering experiments

    Tissue engineering (TE) combines cells and biomaterials to treat orthopedic pathologies. Maturation of de novo tissue is highly dependent on local mechanical environments. Mechanical stimulation influences stem cell differentiation, however, the role of various mechanical loads remains unclear. While bioreactors simplify the complexity of the human body, the potential combination of mechanical loads that can be applied make it difficult to assess how different factors interact. Human bone marrow-derived mesenchymal stromal cells were seeded within a fibrin-polyurethane scaffold and exposed to joint-mimicking motion. We applied a full factorial design of experiment to investigate the effect that the interaction between different mechanical loading parameters has on biological markers. Additionally, we employed planned contrasts to analyze differences between loading protocols and a linear mixed model with donor as random effect. Our approach enables screening of multiple mechanical loading combinations and identification of significant interactions that could not have been studied using classical mechanobiology studies. This is useful to screen the effect of various loading protocols and could also be used for TE experiments with small sample sizes and further combinatorial medication studies
