    Manual and automatic flight control during severe turbulence penetration

    An analytical and experimental investigation of possible contributing factors in jet aircraft turbulence upsets was conducted. Major contributing factors identified included autopilot and display deficiencies, the large aircraft inertia and associated long response time, and excessive pilot workload. An integrated flight and thrust energy management director system was synthesized. The system was incorporated in a moving-base simulation and evaluated using highly experienced airline pilots. The evaluation included comparison of pilot workload and flight performance during severe turbulence penetration utilizing four control/display concepts: manual control with conventional full panel display, conventional autopilot (A/P-A) with conventional full panel display, improved autopilot (A/P-B) with conventional full panel display plus thrust director display, and longitudinal flight director with conventional full panel display plus thrust director display. Simulation results show improved performance, reduced pilot workload, and a pilot preference for the autopilot system controlling to the flight director command and manual control of thrust following the trim thrust director

    Development of automatic and manual flight director landing systems for the XV-15 tilt rotor aircraft in helicopter mode

    The objective of this effort is to determine IFR approach path and touchdown dispersions for manual and automatic XV-15 tilt rotor landings, and to develop missed approach criteria. Only helicopter mode XV-15 operation is considered. The analysis and design sections develop the automatic and flight director guidance equations for decelerating curved and straight-in approaches into a typical VTOL landing site equipped with an MLS navigation aid. These system designs satisfy all known pilot-centered, guidance and control requirements for this flying task. Performance data, obtained from nonstationary covariance propagation dispersion analysis for the system, are used to develop the approach monitoring criteria. The autoland and flight director guidance equations are programmed for the VSTOLAND 1819B digital computer. The system design dispersion data developed through analysis and the 1819B digital computer program are verified and refined using the fixed-base, man-in-the-loop XV-15 VSTOLAND simulation

    The structure of a new flying and ground handling qualities specification for military rotorcraft is presented. This preliminary specification structure is intended to evolve into a replacement for specification MIL-H-8501A. The new structure is designed to accommodate a variety of rotorcraft types, mission flight phases, flight envelopes, and flight environmental characteristics and to provide criteria for three levels of flying qualities, a systematic treatment of failures and reliability, both conventional and multiaxis controllers, and external vision aids which may also incorporate synthetic display content. Existing and new criteria were incorporated into the new structure wherever they could be substantiated

    Handling properties of diverse automobiles and correlation with full scale response data

    Driver/vehicle response and performance of a variety of vehicles in the presence of aerodynamic disturbances are discussed. Steering control is emphasized. The vehicles include full size station wagon, sedan, compact sedan, van, pickup truck/camper, and wagon towing trailer. Driver/vehicle analyses are used to estimate response and performance. These estimates are correlated with full scale data with test drivers and the results are used to refine the driver/vehicle models, control structure, and loop closure criteria. The analyses and data indicate that the driver adjusts his steering control properties (when he can) to achieve roughly the same level of performance despite vehicle variations. For the more disturbance susceptible vehicles, such as the van, the driver tightens up his control. Other vehicles have handling dynamics which cause him to loosen his control response, even though performance degrades

    background: Uveal melanoma (UM) is the most common primary intraocular tumour of adults, frequently metastasising to the liver. Hepatic metastases are difficult to treat and are mainly unresponsive to chemotherapy. To investigate why UM are so chemo-resistant we explored the effect of interstrand cross-linking agents mitomycin C (MMC) and cisplatin in comparison with hydroxyurea (HU). methods: Sensitivity to MMC, cisplatin and HU was tested in established UM cell lines using clonogenic assays. The response of UM to MMC was confirmed in MTT assays using short-term cultures of primary UM. The expression of cytochrome P450 reductase (CYP450R) was analysed by western blotting, and DNA cross-linking was assessed using COMET analysis supported by γ-H2AX foci formation. results: Both established cell lines and primary cultures of UM were resistant to the cross-linking agent MMC (in each case P<0.001 in Student's t-test compared with controls). In two established UM cell lines, DNA cross-link damage was not induced by MMC (in both cases P<0.05 in Students's t-test compared with damage induced in controls). In all, 6 out of 6 UMs tested displayed reduced expression of the metabolising enzyme CYP450R and transient expression of CYP450R increased MMC sensitivity of UM. conclusion: We suggest that reduced expression of CYP450R is responsible for MMC resistance of UM, through a lack of bioactivation, which can be reversed by complementing UM cell lines with CYP450R

It is unclear how the development of B cells and plasma cells in the gastrointestinal mucosa differs between healthy humans and those with food allergy, and how B cells contribute to, or are affected by, the breakdown of oral tolerance. In particular, the antibody gene repertoires associated with symptomatic allergy have only begun to be characterized in full molecular detail.     A Collaboratory, Multi-Disciplinary Approach to Risk Mitigation during HIV Analytical Treatment Interruption

    Analytic treatment interruptions (ATIs) are currently the standard for assessing the impact of experimental interventions aimed at inducing sustained antiretroviral therapy (ART)-free remission in trials related to HIV cure. ATIs are associated with substantial risk to both study participants and their sexual partner(s). Two documented HIV transmissions occurring in the context of ATIs have been recently reported, but recommendations for mitigating the risk of such events during ATIs are limited. We outline a practical approach to risk mitigation during ATI studies and describe strategies we are utilising in an upcoming clinical trial that may be applicable to other centres
