    Individualized and Clinically Derived Stimuli Activate Limbic Structures in Depression: An fMRI Study

    In the search for neurobiological correlates of depression, a major finding is hyperactivity in limbic-paralimbic regions. However, results so far have been inconsistent, and the stimuli used are often unspecific to depression. This study explored hemodynamic responses of the brain in patients with depression while processing individualized and clinically derived stimuli.Eighteen unmedicated patients with recurrent major depressive disorder and 17 never-depressed control subjects took part in standardized clinical interviews from which individualized formulations of core interpersonal dysfunction were derived. In the patient group such formulations reflected core themes relating to the onset and maintenance of depression. In controls, formulations reflected a major source of distress. This material was thereafter presented to subjects during functional magnetic resonance imaging (fMRI) assessment.Increased hemodynamic responses in the anterior cingulate cortex, medial frontal gyrus, fusiform gyrus and occipital lobe were observed in both patients and controls when viewing individualized stimuli. Relative to control subjects, patients with depression showed increased hemodynamic responses in limbic-paralimbic and subcortical regions (e.g. amygdala and basal ganglia) but no signal decrease in prefrontal regions.This study provides the first evidence that individualized stimuli derived from standardized clinical interviewing can lead to hemodynamic responses in regions associated with self-referential and emotional processing in both groups and limbic-paralimbic and subcortical structures in individuals with depression. Although the regions with increased responses in patients have been previously reported, this study enhances the ecological value of fMRI findings by applying stimuli that are of personal relevance to each individual's depression

    EM703 improves bleomycin-induced pulmonary fibrosis in mice by the inhibition of TGF-β signaling in lung fibroblasts

    BACKGROUND: Fourteen-membered ring macrolides have been effective in reducing chronic airway inflammation and also preventing lung injury and fibrosis in bleomycin-challenged mice via anti-inflammatory effects. EM703 is a new derivative of erythromycin (EM) without the bactericidal effects. We investigated the anti-inflammatory and antifibrotic effects of EM703 in an experimental model of bleomycin-induced lung injury and subsequent fibrosis in mice. METHODS: Seven-week-old male ICR mice were used. All experiments used eight mice/group, unless otherwise noted in the figure legends. Bleomycin was administered intravenously to the mice on day 0. EM703 was orally administered daily to mice. All groups were examined for cell populations in the bronchoalveolar lavage (BAL) fluid and for induction of messenger RNA (mRNA) of Smad3 and Smad4 in the lung tissues by reverse transcriptase (RT)-polymerase chainreaction (PCR) on day 7. Fibroblastic foci were assessed histologically, and the hydroxyproline content was chemically determined in the lung tissues on day 28. We performed assay of proliferation and soluble collagen production, and examined the induction of mRNA of Smad3 and Smad4 by RT-PCR in murine lung fibroblast cell line MLg2908. We also examined Smad3, Smad4 and phosphorylated Smad2/3 (p-Smad2/3) protein assay by western blotting in MLg2908. RESULTS: Bleomycin-induced lung fibrosis, and the infiltration of macrophages and neutrophils into the airspace were inhibited by EM703. The expression of Smad3 and Smad4 mRNA was clearly attenuated by bleomycin, but was recovered by EM703. EM703 also inhibited fibroblast proliferation and the collagen production in lung fibroblasts induced by Transforming growth factor-beta (TGF-β). The expression of Smad3 and Smad4 mRNA in murine lung fibroblasts disappeared due to TGF-β, but was recovered by EM703. EM703 inhibited the expression of p-Smad2/3 and Smad4 protein in murine lung fibroblasts induced by TGF-β. CONCLUSION: These findings suggest that EM703 improves bleomycin-induced pulmonary fibrosis in mice by actions of anti-inflammation and regulation of TGF-β signaling in lung fibroblasts

    Money Matters: a Nuanced Approach to Understanding the Relationship between Household Income and Child Subjective Well-Being

    This paper examines the links between household income and child subjective well-being. Previous studies produce contradictory findings: qualitative investigations indicate a strong relationship which is elusive in quantitative studies. I hypothesise that the reason for this discrepancy is that household-level measures of child poverty do not adequately capture children’s active roles in forming views on their material needs, assessing their comparative socio-economic status, and contributing to processes and outcomes of intra-household resource sharing. Thus a relationship between household income and subjective well-being is hypothesised to exist, but to be mediated by factors including (among others) material deprivation, perceptions of fairness in the processes and outcomes of intra-household allocation, and subjective material well-being. Drawing on survey data from a sample of 1010 parent-child (aged 10–16) pairs in England, structural equation modelling is used to examine these potential mediating effects. Findings indicate that income has a complex role to play in child subjective well-being, with significant direct and indirect associations. Income, deprivation, perceptions of the fairness of intra-household allocation processes and outcomes, and subjective material well-being are all significantly interrelated and are all predictors of subjective well-being. The complex nature of these relationships illustrates the multi-dimensional nature of child poverty and its impacts. Household income is an important factor; but alone it cannot capture children’s active roles in assessing their needs and material living conditions. This confirms the importance of considering children’s agency in understandings of child poverty and material well-being, and including their reports in studies of child poverty and intra-household allocation

    Fear of missing out, need for touch, anxiety and depression are related to problematic smartphone use

    Problematic smartphone use is an important public health challenge and is linked with poor mental health outcomes. However, little is known about the mechanisms that maintain this behavior. We recruited a sample of 308 participants from Amazon\u27s Mechanical Turk labor market. Participants responded to standardized measures of problematic smartphone use, and frequency of smartphone use, depression and anxiety and possible mechanisms including behavioral activation, need for touch, fear of missing out (FoMO), and emotion regulation. Problematic smartphone use was most correlated with anxiety, need for touch and FoMO. The frequency of use was most correlated (inversely) with depression. In regression models, problematic smartphone use was associated with FoMO, depression (inversely), anxiety, and need for touch. Frequency of use was associated with need for touch, and (inversely) with depressive symptoms. Behavioral activation mediated associations between smartphone use (both problematic and usage frequency) and depression and anxiety symptoms. Emotional suppression also mediated the association between problematic smartphone use and anxiety. Results demonstrate the importance of social and tactile need fulfillment variables such as FoMO and need for touch as critical mechanisms that can explain problematic smartphone use and its association with depression and anxiety

    Genital condylomata acuminata are nonmalignant human papillomavirus (HPV)-induced tumors in which HPV types 6 and 11 are most commonly found. Usual treatments for condylomata acuminata are nonspecific and are based on the destruction or removal of infected tissue. These procedures are often painful and are characterized by a high relapse rate. We report here what is to our knowledge the first double-blind, placebo-controlled study of the use of cidofovir, a nucleotide analogue, for the treatment of genital papillomavirus infections. Thirty patients were enrolled in the study; 19 received cidofovir, and 11 received placebo. The median number of warts and the median baseline wart area were comparable for both groups. Nine (47%) of 19 patients in the cidofovir group had a complete response (total healing), compared with 0 of the patients in the placebo group (P=.006). None of the patients in the cidofovir group experienced progression of the disease, compared with 5 (45%) of 11 patients in the placebo group.     Evolutionary Robotics: Exploring New Horizons

    This paper considers the field of Evolutionary Robotics (ER) from the perspective of its potential users: roboticists. The core hypothesis motivating this field of research is discussed, as well as the potential use of ER in a robot design process. Four main aspects of ER are presented: (a) ER as an automatic parameter tuning procedure, which is the most mature application and is used to solve real robotics problem, (b) evolutionary-aided design, which may benefit the designer as an efficient tool to build robotic systems (c) ER for online adaptation, i.e. continuous adaptation to changing environment or robot features and (d) automatic synthesis, which corresponds to the automatic design of a mechatronic device and its control system. Critical issues are also presented as well as current trends and pespectives in ER. A section is devoted to a roboticist's point of view and the last section discusses the current status of the field and makes some suggestions to increase its maturity
