Molecular motion in cell membranes: analytic study of fence-hindered random walks
A theoretical calculation is presented to describe the confined motion of
transmembrane molecules in cell membranes. The study is analytic, based on
Master equations for the probability of the molecules moving as random walkers,
and leads to explicit usable solutions including expressions for the molecular
mean square displacement and effective diffusion constants. One outcome is a
detailed understanding of the dependence of the time variation of the mean
square displacement on the initial placement of the molecule within the
confined region. How to use the calculations is illustrated by extracting
(confinement) compartment sizes from experimentally reported published
observations from single particle tracking experiments on the diffusion of
gold-tagged G-protein coupled mu-opioid receptors in the normal rat kidney cell
membrane, and by further comparing the analytical results to observations on
the diffusion of phospholipids, also in normal rat kidney cells.Comment: 10 pages, 5 figure
The Phase Behavior of Mixed Lipid Membranes in Presence of the Rippled Phase
We propose a model describing liquid-solid phase coexistence in mixed lipid
membranes by including explicitly the occurrence of a rippled phase. For a
single component membrane, we employ a previous model in which the membrane
thickness is used as an order parameter. As function of temperature, this model
properly accounts for the phase behavior of the three possible membrane phases:
solid, liquid and the rippled phase. Our primary aim is to explore extensions
of this model to binary lipid mixtures by considering the composition
dependence of important model parameters. The obtained phase diagrams show
various liquid, solid and rippled phase coexistence regions, and are in
quantitative agreement with the experimental ones for some specific lipid

Meshfree finite differences for vector Poisson and pressure Poisson equations with electric boundary conditions
We demonstrate how meshfree finite difference methods can be applied to solve
vector Poisson problems with electric boundary conditions. In these, the
tangential velocity and the incompressibility of the vector field are
prescribed at the boundary. Even on irregular domains with only convex corners,
canonical nodal-based finite elements may converge to the wrong solution due to
a version of the Babuska paradox. In turn, straightforward meshfree finite
differences converge to the true solution, and even high-order accuracy can be
achieved in a simple fashion. The methodology is then extended to a specific
pressure Poisson equation reformulation of the Navier-Stokes equations that
possesses the same type of boundary conditions. The resulting numerical
approach is second order accurate and allows for a simple switching between an
explicit and implicit treatment of the viscosity terms.Comment: 19 pages, 7 figure
Mammography screening: views from women and primary care physicians in Crete
Background: Breast cancer is the most commonly diagnosed cancer among women and a leading cause of death from cancer in women in Europe. Although breast cancer incidence is on the rise worldwide, breast cancer mortality over the past 25 years has been stable or decreasing in some countries and a fall in breast cancer mortality rates in most European countries in the 1990s was reported by several studies, in contrast, in Greece have not reported these favourable trends. In Greece, the age-standardised incidence and mortality rate for breast cancer per 100.000 in 2006 was 81,8 and 21,7 and although it is lower than most other countries in Europe, the fall in breast cancer mortality that observed has not been as great as in other European countries. There is no national strategy for screening in this country. This study reports on the use of mammography among middleaged women in rural Crete and investigates barriers to mammography screening encountered by women and their primary care physicians.
Methods: Design: Semi-structured individual interviews. Setting and participants: Thirty women between 45–65
years of age, with a mean age of 54,6 years, and standard deviation 6,8 from rural areas of Crete and 28 qualified
primary care physicians, with a mean age of 44,7 years and standard deviation 7,0 serving this rural population.
Main outcome measure: Qualitative thematic analysis.
Results: Most women identified several reasons for not using mammography. These included poor knowledge
of the benefits and indications for mammography screening, fear of pain during the procedure, fear of a serious
diagnosis, embarrassment, stress while anticipating the results, cost and lack of physician recommendation.
Physicians identified difficulties in scheduling an appointment as one reason women did not use mammography
and both women and physicians identified distance from the screening site, transportation problems and the
absence of symptoms as reasons for non-use.
Conclusion: Women are inhibited from participating in mammography screening in rural Crete. The provision
of more accessible screening services may improve this. However physician recommendation is important in
overcoming women's inhibitions. Primary care physicians serving rural areas need to be aware of barriers
preventing women from attending mammography screening and provide women with information and advice in a sensitive way so women can make informed decisions regarding breast caner screening
Localization of Events in Space-Time
The present paper deals with the quantum coordinates of an event in
space-time, individuated by a quantum object. It is known that these
observables cannot be described by self-adjoint operators or by the
corresponding spectral projection-valued measure. We describe them by means of
a positive-operator-valued (POV) measure in the Minkowski space-time,
satisfying a suitable covariance condition with respect to the Poincare' group.
This POV measure determines the probability that a measurement of the
coordinates of the event gives results belonging to a given set in space-time.
We show that this measure must vanish on the vacuum and the one-particle
states, which cannot define any event. We give a general expression for the
Poincare' covariant POV measures. We define the baricentric events, which lie
on the world-line of the centre-of-mass, and we find a simple expression for
the average values of their coordinates. Finally, we discuss the conditions
which permit the determination of the coordinates with an arbitrary accuracy.Comment: 31 pages, latex, no figure
Genome-wide association studies using single-nucleotide polymorphisms versus haplotypes: an empirical comparison with data from the North American Rheumatoid Arthritis Consortium
The high genomic density of the single-nucleotide polymorphism (SNP) sets that are typically surveyed in genome-wide association studies (GWAS) now allows the application of haplotype-based methods. Although the choice of haplotype-based vs. individual-SNP approaches is expected to affect the results of association studies, few empirical comparisons of method performance have been reported on the genome-wide scale in the same set of individuals. To measure the relative ability of the two strategies to detect associations, we used a large dataset from the North American Rheumatoid Arthritis Consortium to: 1) partition the genome into haplotype blocks, 2) associate haplotypes with disease, and 3) compare the results with individual-SNP association mapping. Although some associations were shared across methods, each approach uniquely identified several strong candidate regions. Our results suggest that the application of both haplotype-based and individual-SNP testing to GWAS should be adopted as a routine procedure
Real-life use of vitamin D<sub>3-</sub>fortified bread and milk during a winter season: the effects of CYP2R1 and GC genes on 25-hydroxyvitamin D concentrations in Danish families, the VitmaD study.
Common genetic variants rs10741657 and rs10766197 in CYP2R1 and rs4588 and rs842999 in GC and a combined genetic risk score (GRS) of these four variants influence late summer 25-hydroxyvitamin D (25(OH)D) concentrations. The objectives were to identify those who are most at risk of developing low vitamin D status during winter and to assess whether vitamin D(3)-fortified bread and milk will increase 25(OH)D concentrations in those with genetically determined low 25(OH)D concentrations at late summer. We used data from the VitmaD study. Participants were allocated to either vitamin D(3)-fortified bread and milk or non-fortified bread and milk during winter. In the fortification group, CYP2R1 (rs10741657) and GC (rs4588 and rs842999) were statistically significantly associated with winter 25(OH)D concentrations and CYP2R1 (rs10766197) was borderline significant. There was a negative linear trend between 25(OH)D concentrations and carriage of 0–8 risk alleles (p < 0.0001). No association was found for the control group (p = 0.1428). There was a significant positive linear relationship between different quintiles of total vitamin D intake and the increase in 25(OH)D concentrations among carriers of 0–2 (p = 0.0012), 3 (p = 0.0001), 4 (p = 0.0118) or 5 (p = 0.0029) risk alleles, but not among carriers of 6–8 risk alleles (p = 0.1051). Carriers of a high GRS were more prone to be vitamin D deficient compared to carriers of a low GRS. Furthermore, rs4588-AA carriers have a low but very stable 25(OH)D concentration, and interestingly, also low PTH level. HIV-1 Tat Promotes Kaposi's Sarcoma-Associated Herpesvirus (KSHV) vIL-6-Induced Angiogenesis and Tumorigenesis by Regulating PI3K/PTEN/AKT/GSK-3β Signaling Pathway
Kaposi's sarcoma (KS)-associated herpesvirus (KSHV) is etiologically associated with KS, the most common AIDS-related malignancy. KS is characterized by vast angiogenesis and hyperproliferative spindle cells. We have previously reported that HIV-1 Tat can trigger KSHV reactivation and accelerate Kaposin A-induced tumorigenesis. Here, we explored Tat promotion of KSHV vIL-6-induced angiogenesis and tumorigenesis. Tat promotes vIL-6-induced cell proliferation, cellular transformation, vascular tube formation and VEGF production in culture. Tat enhances vIL-6-induced angiogenesis and tumorigenesis of fibroblasts and human endothelial cells in a chicken chorioallantoic membrane (CAM) model. In an allograft model, Tat promotes vIL-6-induced tumorigenesis and expression of CD31, CD34, SMA, VEGF, b-FGF, and cyclin D1. Mechanistic studies indicated Tat activates PI3K and AKT, and inactivates PTEN and GSK-3β in vIL-6 expressing cells. LY294002, a specific inhibitor of PI3K, effectively impaired Tat's promotion of vIL-6-induced tumorigenesis. Together, these results provide the first evidence that Tat might contribute to KS pathogenesis by synergizing with vIL-6, and identify PI3K/AKT pathway as a potential therapeutic target in AIDS-related KS patients. © 2013 Zhou et al
