    Development And Testing Of The Inertial Electrostatic Confinement Diffusion Thruster

    The Inertial Electrostatic Confinement (IEC) diffusion thruster is an experiment in active development that takes advantage of physical phenomenon that occurs during operation of an IEC device. The IEC device has been proposed as a fusion reactor design that relies on traditional electrostatic ion acceleration and is typically arranged in a spherical geometry. The design incorporates two radially-symmetric spherical electrodes. Often the inner electrode utilizes a grid of wire shaped in a sphere with a radius 15 to 50 percent of the radius of the outer electrode. The inner electrode traditionally has 90 percent or more transparency to allow particles (ions) to pass to the center of the spheres and collide/recombine in the dense plasma core at r=0. When operating the IEC, an unsteady plasma leak is typically observed passing out one of the gaps in the lattice grid of the inner electrode. The IED diffusion thruster is based upon the idea that this plasma leak can be used for propulsive purposes. The IEC diffusion thruster utilizes the radial symmetry found in the IEC device. A cylindrical configuration is employed here as it will produce a dense core of plasma the length of the cylindrical grid while promoting the plasma leak to exhaust through an electromagnetic nozzle at one end of the apparatus. A proof-of-concept IEC diffusion thruster is operational and under testing using argon as propellant (Figure 1)

    Medical, Recreational, and Mixed Marijuana Users: An Examination of Physical and Mental Health Correlates

    Marijuana use in the U.S. doubled between 2001 and 2013, largely due to increases in legalization laws. Little attention, however, is given to the type of marijuana user (e.g., recreational or medical), particularly with health outcomes. Our study used data from the 2017 Behavioral Risk Factor Surveillance System (N=5,349) to examine physical health, mental health, and demographic variables by marijuana user type (including non-marijuana users). In physical health, the non-marijuana group was generally healthier, getting the most sleep, lowest BMI, and lowest alcoholic consumption. Medical users self-reported the poorest physical health, BMI, and sleep. Similar results were found in the mental health category between non-marijuana and medical users. Future longitudinal research is needed to investigate whether medical users, over time, increase their marijuana use to include recreational use (i.e., become mixed users) as a method of coping with the combination of health, emotional, and quality of life problems. Although this is among the first nationally representative studies to examine unique marijuana user groups, future studies should track user groups over time to understand the implications of transitioning into medical or recreational user groups

    Love-bombing: A Narcissistic Approach to Relationship Formation

    The current study examined the relationship between attachment style, self-esteem, and narcissism as they pertain to behavioral tendencies, termed love-bombing behaviors, among a sample of young adult millennials. Love-bombing was identified as the presence of excessive communication at the beginning of a romantic relationship in order to obtain power and control over another’s life as a means of narcissistic self-enhancement. Millennials have shown a drastic increase in narcissism compared to generations prior, and the need for psychological services on college campuses has also increased. This study sought to establish empirical evidence for the presence of love-bombing behaviors amongst millennials as a gateway for further research to address the problem facing young adult relationships today. The sample consisted of 484 college students from a large southern university who ranged in age from 18 to 30. Results indicated that love-bombing was positively correlated with narcissistic tendencies and insecure attachment styles (lack of trust or value in self and others), and negatively associated with self-esteem. Secure attachment was a positive indicator of love-bombing behaviors. Lastly, love-bombing was also associated with more text and media usage within romantic relationships. In conclusion, love-bombing was found to be a logical and potentially necessary strategy for romantic relationships among individuals with high displays of narcissism and low levels of self-esteem. This is the first study to empirically examine love-bombing behaviors; thus, future research should address the impact that these behaviors may have on young adult relationships. The potential for negative psychological impact on both love-bombers and the subject of their attacks are discussed

    Immunomodulation with IL-4Rα antisense oligonucleotide prevents respiratory syncytial virus-mediated pulmonary disease

    Respiratory syncytial virus (RSV) causes significant morbidity and mortality in infants worldwide. Severe RSV infections in infants cause bronchiolitis, wheeze, and/or cough and significantly increase the risk for developing asthma. RSV pathogenesis is thought to be due to a Th2-type immune response initiated in response to RSV infection, specifically in the infant. Using a neonatal mouse system as an appropriate model for human infants, we sought to determine whether local inhibition of IL-4Rα expression during primary RSV infection in the neonate would prevent Th2-skewed responses to secondary RSV infection and improve longterm pulmonary function. To reduce IL-4Rα expression, antisense oligonucleotides (ASOs) specific for IL-4Rα were administered intranasally to neonatal mice at the time of primary infection. Mice were initially infected with RSV at 1 wk of age and were reinfected at 6 wk of age. Administration of IL-4Rα ASOs during primary RSV infection in neonatal mice abolished the pulmonary dysfunction normally observed following reinfection in the adult. This ablation of pulmonary dysfunction correlated with a persistent rebalancing of the Th cell compartment with decreased Th2 responses (i.e., reduced goblet cell hyperplasia, Th2 cells, and cytokine secretion) and increased Th1 responses (i.e., elevated Th1 cell numbers and type I Abs and cytokines). Our data support our hypothesis that a reduction in the Th2 immune response during primary infection in neonates prevents Th2-mediated pulmonary pathology initially and upon reinfection and further suggest that vaccine strategies incorporating IL-4Rα ASOs may be of significant benefit to infants.     Reduction in \u3ci\u3eMusca domestica\u3c/i\u3e fecundity by dsRNA-mediated gene knockdown

    House flies (Musca domestica) are worldwide agricultural pests with estimated control costs at $375 million annually in the U.S. Non-target effects and widespread resistance challenge the efficacy of traditional chemical control. Double stranded RNA (dsRNA) has been suggested as a biopesticide for M. domestica but a phenotypic response due to the induction of the RNAi pathway has not been demonstrated in adults. In this study female house flies were injected with dsRNA targeting actin-5C or ribosomal protein (RP) transcripts RPL26 and RPS6. Ovaries showed highly reduced provisioning and clutch reductions of 94±99% in RP dsRNA treated flies but not in actin-5C or GFP treated flies. Gene expression levels were significantly and specifically reduced in dsRNA injected groups but remained unchanged in the control dsGFP treated group. Furthermore, injections with an Aedes aegypti conspecific dsRNA designed against RPS6 did not impact fecundity, demonstrating species specificity of the RNAi response. Analysis of M. domestica tissues following RPS6 dsRNA injection showed significant reduction of transcript levels in the head, thorax, and abdomen but increased expression in ovarian tissues. This study demonstrates that exogenous dsRNA is specifically effective and has potential efficacy as a highly specific biocontrol intervention in adult house flies. Further work is required to develop effective methods for delivery of dsRNA to adult flies

    DREADDs in Drosophila: A Pharmacogenetic Approach for Controlling Behavior, Neuronal Signaling, and Physiology in the Fly

    SummaryWe have translated a powerful genetic tool, designer receptors exclusively activated by designer drugs (DREADDs), from mammalian systems to Drosophila melanogaster to selectively, rapidly, reversibly, and dose-dependently control behaviors and physiological processes in the fly. DREADDs are muscarinic acetylcholine G protein-coupled receptors evolved for loss of affinity to acetylcholine and for the ability to be fully activated by an otherwise biologically inert chemical, clozapine-N-oxide. We demonstrate its ability to control a variety of behaviors and processes in larvae and adults, including heart rate, sensory processing, diurnal behavior, learning and memory, and courtship. The advantages of this particular technology include the dose-responsive control of behaviors, the lack of a need for specialized equipment, and the capacity to remotely control signaling in essentially all neuronal and nonneuronal fly tissues

    A Comparison of Match Outcomes Between Traditional Medical Degree and Dual-Degree Applicants

    Background: Dual degrees combining and MD with another professional degree (MPH, MBA, or PhD) are becoming more common in an attempt to increase an applicant’s competitivity for a residency. Objective: This study was designed to assess differences in MD-only and dual degree MD applicants with respect to applicant characteristics and match outcomes. Methods: Utilizing the voluntarily-reported publicly available 2017–2019 Texas STAR database, we assessed applicants from 115 medical schools. Texas STAR indicates that over this time period, there were 18,224 responses for a response rate of 43.8%. Comparisons were made between groups using student’s t-test and chi-squared analysis. Results: Compared to MD only students, MD/MPH applicants had a higher propensity towards primary care specialties. MD/PhD applicants did not differ versus MD only applicants in their selection of primary care specialties, or of competitive specialties. MD/MBA applicants chose more competitive specialties and less primary care specialties. Despite all these differences, match rates were not different comparing MD only and dual-degree students. Conclusions: Despite the growing popularity of combined MD programs, such programs do not appear to increase applicant match competitivity

    Modulation of lung inflammation by vessel dilator in a mouse model of allergic asthma

    <p>Abstract</p> <p>Background</p> <p>Atrial natriuretic peptide (ANP) and its receptor, NPRA, have been extensively studied in terms of cardiovascular effects. We have found that the ANP-NPRA signaling pathway is also involved in airway allergic inflammation and asthma. ANP, a C-terminal peptide (amino acid 99–126) of pro-atrial natriuretic factor (proANF) and a recombinant peptide, NP73-102 (amino acid 73–102 of proANF) have been reported to induce bronchoprotective effects in a mouse model of allergic asthma. In this report, we evaluated the effects of vessel dilator (VD), another N-terminal natriuretic peptide covering amino acids 31–67 of proANF, on acute lung inflammation in a mouse model of allergic asthma.</p> <p>Methods</p> <p>A549 cells were transfected with pVD or the pVAX1 control plasmid and cells were collected 24 hrs after transfection to analyze the effect of VD on inactivation of the extracellular-signal regulated receptor kinase (ERK1/2) through western blot. Luciferase assay, western blot and RT-PCR were also performed to analyze the effect of VD on NPRA expression. For determination of VD's attenuation of lung inflammation, BALB/c mice were sensitized and challenged with ovalbumin and then treated intranasally with chitosan nanoparticles containing pVD. Parameters of airway inflammation, such as airway hyperreactivity, proinflammatory cytokine levels, eosinophil recruitment and lung histopathology were compared with control mice receiving nanoparticles containing pVAX1 control plasmid.</p> <p>Results</p> <p>pVD nanoparticles inactivated ERK1/2 and downregulated NPRA expression in vitro, and intranasal treatment with pVD nanoparticles protected mice from airway inflammation.</p> <p>Conclusion</p> <p>VD's modulation of airway inflammation may result from its inactivation of ERK1/2 and downregulation of NPRA expression. Chitosan nanoparticles containing pVD may be therapeutically effective in preventing allergic airway inflammation.</p
