    A comparison of structural reform scenarios across the EU member states - Simulation-based analysis using the QUEST model with endogenous growth

    This paper calibrates the Roeger-Varga-Veld (2008) micro-founded DSGE model with endogenous growth for all EU member states using country specific structural characteristics and employs the individual country models to analyse the macroeconomic impact of various structural reforms. We analyse the costs and benefits of reforms in terms of fiscal policy instruments such as taxes, benefits, subsidies and administrative costs faced by firms. We find that less R&D intensive countries would benefit the most from R&D promoting and skill-upgrading policies.     Refinement strategies for polygonal meshes applied to adaptive VEM discretization

    In the discretization of differential problems on complex geometrical domains, discretization methods based on polygonal and polyhedral elements are powerful tools. Adaptive mesh refinement for such kind of problems is very useful as well and states new issues, here tackled, concerning good quality mesh elements and reliability of the simulations. In this paper we propose several new polygonal refinement strategies and numerically investigate the quality of the meshes generated by an adaptive mesh refinement process, as well as optimal rates of convergence with respect to the number of degrees of freedom. Among the several possible problems in which these strategies can be applied, here we have considered a geometrically complex geophysical problem as test problem that naturally yields to a polygonal mesh and tackled it by the Virtual Element Method. All the adaptive strategies here proposed, but the “Trace Direction strategy”, can be applied to any problem for which a polygonal element method can be useful and any numerical method based on polygonal elements and can generate good quality isotropic mesh elements

    Non-perturbative triality in heterotic and type II N=2 strings

    The non-perturbative equivalence of four-dimensional N=2 superstrings with three vector multiplets and four hypermultiplets is analysed. These models are obtained through freely acting orbifold compactifications from the heterotic, the symmetric and the asymmetric type II strings. The heterotic scalar manifolds are (SU(1,1) / U(1))^3 for the S,T,U moduli sitting in the vector multiplets and SO(4,4)/ (SO(4) X SO(4)) for those in the hypermultiplets. The type II symmetric duals correspond to a self-mirror Calabi-Yau threefold compactification with Hodge numbers h(1,1)=h(2,1)=3, while the type II asymmetric construction corresponds to a spontaneous breaking of the N=(4,4) supersymmetry to N=(2,0). Both have already been considered in the literature. The heterotic construction instead is new and we show that there is a weak/strong coupling S-duality relation between the heterotic and the asymmetric type IIA ground state with S(Het)=-1/S(As); we also show that there is a partial restoration of N=8 supersymmetry in the heterotic strong-coupling regime. The design and construction of the Gamma Beam System complex as well as the integration of the technical plants and the commissioning of the overall facility, was awarded to the Eurogammas Consortium in March 2014.     Polydatin Incorporated in Polycaprolactone Nanofibers Improves Osteogenic Differentiation

    Polycaprolactone nanofibers are used as scaffolds in the field of tissue engineering for tissue regeneration or drug delivery. Polycaprolactone (PCL) is a biodegradable hydrophobic polyester used to obtain implantable nanostructures, which are clinically applicable due to their biological safety. Polydatin (PD), a glycosidic precursor of resveratrol, is known for its antioxidant, antitumor, antiosteoporotic, and bone regeneration activities. We aimed to use the osteogenic capacity of polydatin to create a biomimetic innovative and patented scaffold consisting of PCL-PD for bone tissue engineering. Both osteosarcoma cells (Saos-2) and mesenchymal stem cells (MSCs) were used to test the in vitro cytocompatibility of the PD-PCL scaffold. Reverse-phase (RP) HPLC was used to evaluate the timing release of PD from the PCL-PD nanofibers and the MTT assay, scanning electron microscopy, and alkaline phosphatase (ALP) activity were used to evaluate the proliferation, adhesion, and cellular differentiation in both osteosarcoma and human mesenchymal stem cells (MSCs) seeded on PD-PCL nanofibers. The proliferation of osteosarcoma cells (Saos-2) on the PD-PCL scaffold decreased when compared to cells grown on PLC nanofibers, whereas the proliferation of MSCs was comparable in both PCL and PD-PCL nanofibers. Noteworthy, after 14 days, the ALP activity was higher in both Saos-2 cells and MSCs cultivated on PD-PCL than on empty scaffolds. Moreover, the same cells showed a spindle-shaped morphology after 14 days when grown on PD-PCL as shown by SEM. In conclusion, we provide evidence that nanofibers appropriately coated with PD support the adhesion and promote the osteogenic differentiation of both human osteosarcoma cells and MSCs

    The impact of the Hippo pathway and cell metabolism on pathological complete response in locally advanced Her2+ breast cancer: the TRISKELE multicenter prospective study

    The Hippo pathway and its two key effectors, Yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ), are consistently altered in breast cancer. Pivotal regulators of cell metabolism such as the AMP-activated protein kinase (AMPK), Stearoyl-CoA-desaturase 1 (SCD1), and HMG-CoA reductase (HMGCR) are relevant modulators of TAZ/YAP activity. In this prospective study, we measured the tumor expression of TAZ, YAP, AMPK, SCD1, and HMGCR by immunohistochemistry in 65 Her2+ breast cancer patients who underwent trastuzumab-based neoadjuvant treatment. The aim of the study was to assess the impact of the immunohistochemical expression of the Hippo pathway transducers and cell metabolism regulators on pathological complete response. Low expression of cytoplasmic TAZ, both alone and in the context of a composite signature identified by machine learning including also low nuclear levels of YAP and HMGCR and high cytoplasmic levels of SCD1, was a predictor of residual disease in the univariate logistic regression. This finding was not confirmed in the multivariate model including estrogen receptor > 70% and body mass index > 20. However, our findings were concordant with overall survival data from the TCGA cohort. Our results, possibly affected by the relatively small sample size of this study population, deserve further investigation in adequately sized, ad hoc prospective studies

MORFEO has officially passed the Preliminary Design Review and it is entering the final design phase.
