    Combining blue native polyacrylamide gel electrophoresis with liquid chromatography tandem mass spectrometry as an effective strategy for analyzing potential membrane protein complexes of Mycobacterium bovis bacillus Calmette-Guérin

    <p>Abstract</p> <p>Background</p> <p>Tuberculosis is an infectious bacterial disease in humans caused primarily by <it>Mycobacterium tuberculosis</it>, and infects one-third of the world's total population. <it>Mycobacterium bovis </it>bacillus Calmette-Guérin (BCG) vaccine has been widely used to prevent tuberculosis worldwide since 1921. Membrane proteins play important roles in various cellular processes, and the protein-protein interactions involved in these processes may provide further information about molecular organization and cellular pathways. However, membrane proteins are notoriously under-represented by traditional two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) and little is known about mycobacterial membrane and membrane-associated protein complexes. Here we investigated <it>M. bovis </it>BCG by an alternative proteomic strategy coupling blue native PAGE to liquid chromatography tandem mass spectrometry (LC-MS/MS) to characterize potential protein-protein interactions in membrane fractions.</p> <p>Results</p> <p>Using this approach, we analyzed native molecular composition of protein complexes in BCG membrane fractions. As a result, 40 proteins (including 12 integral membrane proteins), which were organized in 9 different gel bands, were unambiguous identified. The proteins identified have been experimentally confirmed using 2-D SDS PAGE. We identified MmpL8 and four neighboring proteins that were involved in lipid transport complexes, and all subunits of ATP synthase complex in their monomeric states. Two phenolpthiocerol synthases and three arabinosyltransferases belonging to individual operons were obtained in different gel bands. Furthermore, two giant multifunctional enzymes, Pks7 and Pks8, and four mycobacterial Hsp family members were determined. Additionally, seven ribosomal proteins involved in polyribosome complex and two subunits of the succinate dehydrogenase complex were also found. Notablely, some proteins with high hydrophobicity or multiple transmembrane helixes were identified well in our work.</p> <p>Conclusions</p> <p>In this study, we utilized LC-MS/MS in combination with blue native PAGE to characterize modular components of multiprotein complexes in BCG membrane fractions. The results demonstrated that the proteomic strategy was a reliable and reproducible tool for analysis of BCG multiprotein complexes. The identification in our study may provide some evidence for further study of BCG protein interaction.</p

    Peroxiredoxin-2 as a candidate biomarker to test oxidative stress levels of stored red blood cells under blood bank conditions

    BACKGROUND: Several researches on aging red blood cells (RBCs) - performed both in vivo and under blood bank conditions - revealed that RBC membrane proteins undergo a number of irreversible alterations, mainly due to oxidative stress. The individuation of proteins to be used as indicators of irreversible RBC injury and to be proposed as candidate biomarkers of oxidative damage or aging status during blood storage is therefore of great interest. STUDY DESIGN AND METHODS: Based on this purpose we performed proteomic analysis of the membranes of RBCs during various storage periods under blood bank conditions. Changes in protein composition of RBC membranes were monitored as a function of the storage period by means of polyacrylamide gel electrophoresis coupled with immunoblotting and mass spectrometry analyses. RESULTS: During storage, a progressive linkage of typical cytosolic proteins to the membrane was detected, including both antioxidant and metabolic enzymes (such as catalase, peroxiredoxin-2 [Prx2], and 2,3-bisphosphoglycerate-mutase), as well as nonreducible cross-linkings of probably oxidized or denatured hemoglobin. This phenomenon was unequivocally related to oxidative stress, since storage of RBCs under anaerobic conditions showed a suppression of these protein recruitments to the membrane. CONCLUSION: The detailed analysis of these protein associations to the membrane of aged RBCs allowed Prx2 to be suggested as a potential RBC oxidative stress marker for the sake of developing new approaches in quality assurance of blood components. © 2011 American Association of Blood Banks

    Prevalence and antimicrobial susceptibility of salmonella in European wild boar (Sus scrofa); Latium Region - Italy.

    The prevalence of Salmonella spp. infection was determined in 499 wild boars harvested during the 2010–2011 and 2011–2012 hunting seasons in the Latium Region of Italy. We conducted a microbiological assessment on faeces collected at slaughter and we examined serum samples for the presence of antibodies to Salmonella spp. by ELISA assay. Out of 383 serum samples examined, 255 (66.5%) were positive for Salmonella spp. antibodies. Overall, 10.8% (54/499) of the animals were positive by microbiological assessment. The Salmonellae most frequently isolated were S. enterica subsp. salamae II (24%), S. enterica subsp. Diarizonae III b (12.9%), S. enterica subsp. houtenae IV (11.1%) and S. Fischerhuette (7.4%); less common Salmonella isolates included S. Veneziana (5.5%), S. Napoli (5.5%), S. Kottbus (5.5%), S. Thompson (5.5%), S. enterica subsp. arizonae III a (3.7%), S. Toulon (3.7%), S. Burgas (1.8%), S. Tennelhone (1.8%), S. Ferruch (1.8%), S. choleraesuis (1.8%), S. Paratyphi (1.8%), S. Stanleyville (1.8%), S. Typhimurium (1.8%) and S. enterica subsp. enterica 4,5,12:1:- (1.8%). These isolates were tested against 16 antimicrobial agents and exhibited resistance to sulphonamides (92.5%), sulphonamides and thrimetroprim (14.8%), colistin (14.8%), streptomycin (18.5%), gentamycin (5.5%), tetracycline (5.5%), ceftiofur (3.7%), cefazoline (1.8%), cefotaxime (1.8%), nalidixic acid (1.8%), amoxicillin and clavulanic acid (1.8%) and ampicillin (3.7%). Our data, the first collected on this species in Italy, suggest that European wild boars are frequent carriers of antimicrobial-resistant Salmonellae and are likely involved in the transmission of antimicrobial resistance throughout the environment
