    Positive association between population genetic differentiation and speciation rates in New World birds

    An implicit assumption of speciation biology is that population differentiation is an important stage of evolutionary diversification, but its significance as a rate-limiting control on phylogenetic speciation dynamics remains largely untested. If population differentiation within a species is related to its speciation rate over evolutionary time, the causes of differentiation could also be driving dynamics of organismal diversity across time and space. Alternatively, geographic variants might be short-lived entities with rates of formation that are unlinked to speciation rates, in which case the causes of differentiation would have only ephemeral impacts. By pairing population genetics datasets from173 NewWorld bird species (\u3e17,000 individuals) with phylogenetic estimates of speciation rate, we show that the population differentiation rates within species are positively correlated with their speciation rates over long timescales. Although population differentiation rate explains relatively little of the variation in speciation rate among lineages, the positive relationship between differentiation rate and speciation rate is robust to species-delimitation schemes and to alternative measures of both rates. Population differentiation occurs at least three times faster than speciation, which suggests that most populations are ephemeral. Speciation and population differentiation rates are more tightly linked in tropical species than in temperate species, consistent with a history of more stable diversification dynamics through time in the Tropics. Overall, our results suggest that the processes responsible for population differentiation are tied to those that underlie broad-scale patterns of diversity

    Este artigo discute o modo como a Cultura Digital afeta a atuação docente na área da Educação Musical, problematizando o uso de recursos tecnológicos e seus desafios na prática pedagógica. A partir da reflexão sobre os conceitos de Musicalidade e Cultura Digital, o trabalho analisa um recorte sobre as transformações dos processos de aprendizagem e autoaprendizagem das práticas musicais subsidiadas pelas Novas Tecnologias Digitais (NTD) no contexto da Educação Musical. A metodologia foi constituída de revisão de literatura, questionário on-line e entrevista semiestruturada com músicos profissionais experientes, cujos dados coletados foram analisados na perspectiva quali-quantitativa. Os resultados demonstram que a Cultura Digital permeia as práticas pedagógicas do planejamento e pesquisa de materiais à implementação de recursos de aprendizagem e autoaprendizagem. A pesquisa aponta desafios da docência no campo da Educação Musical, considerando o descompasso constatado entre a fluência na Cultura Digital dos estudantes e as limitações materiais de infraestrutura, de conectividade e de formação inicial e continuada de professores. Defende que a Educação Musical se beneficia com o emprego das novas tecnologias digitais, porém exige uma mudança paradigmática ao entender o estudante como um ser integral e protagonista do processo educativo-musical, cuja bagagem tecnológica pode ser crucial no processo educativo

    Extensive Monte Carlo simulations of helium-4 monolayer films adsorbed on weak substrates have been carried out, aimed at ascertaining the possible occurrence of a quasi-two-dimensional supersolid phase. Only crystalline films not registered with underlying substrates are considered. Numerical results yield strong evidence that helium-4 will not form a supersolid film on {any} substrate strong enough to stabilize a crystalline layer. On weaker substrates, continuous growth of a liquid film takes place

    On the possible "supersolid" character of parahydrogen clusters

    We present results of a theoretical study of structural and superfluid properties of parahydrogen clusters comprising 25, 26 and 27 molecules at low temperature. The microscopic model utilized here is based on the Silvera-Goldman pair potential. Numerical results are obtained by means of Quantum Monte Carlo simulations, making use of the continuous-space Worm Algorithm. The clusters are superfluid in the low temperature limit, but display markedly different physical behaviours. For N=25 and 27, superfluidity at low temperature arises as clusters melt, i.e., become progressively liquid-like as a result of quantum effects. On the other hand, for N = 26 the cluster remains rigid and solid-like. We argue that this cluster can be regarded as a mesoscopic "supersolid". This physical picture is supported by results of simulations in which a single parahydrogen molecule in the cluster is isotopically substituted.Comment: 18 pages, 7 figure

    Nitric Oxide Resistance in Leishmania (Viannia) braziliensis Involves Regulation of Glucose Consumption, Glutathione Metabolism and Abundance of Pentose Phosphate Pathway Enzymes

    In American Tegumentary Leishmaniasis production of cytokines, reactive oxygen species and nitric oxide (NO) by host macrophages normally lead to parasite death. However, some Leishmania braziliensis strains exhibit natural NO resistance. NO-resistant strains cause more lesions and are frequently more resistant to antimonial treatment than NO-susceptible ones, suggesting that NO-resistant parasites are endowed with specific mechanisms of survival and persistence. To tests this, we analyzed the effect of pro- and antioxidant molecules on the infectivity in vitro of L. braziliensis strains exhibiting polar phenotypes of resistance or susceptibility to NO. In addition, we conducted a comprehensive quantitative mass spectrometry-based proteomics analysis of those parasites. NO-resistant parasites were more infective to peritoneal macrophages, even in the presence of high levels of reactive species. Principal component analysis of protein concentration values clearly differentiated NO-resistant from NO-susceptible parasites, suggesting that there are natural intrinsic differences at molecular level among those strains. Upon NO exposure, NO-resistant parasites rapidly modulated their proteome, increasing their total protein content and glutathione (GSH) metabolism. Furthermore, NO-resistant parasites showed increased glucose analogue uptake, and increased abundance of phosphotransferase and G6PDH after nitrosative challenge, which can contribute to NADPH pool maintenance and fuel the reducing conditions for the recovery of GSH upon NO exposure. Thus, increased glucose consumption and GSH-mediated redox capability may explain the natural resistance of L. braziliensis against NO

    The Mare as a Model for Luteinized Unruptured Follicle Syndrome: Intrafollicular Endocrine Milieu.

    Luteinized unruptured follicle (LUF) syndrome is a recurrent anovulatory dysfunction that affects up to 23% of women with normal menstrual cycles and up to 73% with endometriosis. Mechanisms underlying the development of LUF syndrome in mares were studied to provide a potential model for human anovulation. The effect of extended increase in circulating LH achieved by administration of recombinant equine LH (reLH) or a short surge of LH and decrease in progesterone induced by prostaglandin F2α (PGF2α) on LUF formation (Experiment 1), identification of an optimal dose of COX-2 inhibitor (flunixin meglumine, FM; to block the effect of prostaglandins) for inducing LUFs (Experiment 2), and evaluation of intrafollicular endocrine milieu in LUFs (Experiment 3) were investigated. In Experiment 1, mares were treated with reLH from Day 7 to Day 15 (Day 0=ovulation), PGF2α on Day 7, or in combination. In Experiment 2, FM at doses of 2.0 or 3.0 mg/kg every 12 h and human chorionic gonadotropin (hCG) (1500 IU) were administered after a follicle ≥32 mm was detected. In Experiment 3, FM at a dose of 2.0 mg/kg every 12 h plus hCG was used to induce LUFs and investigate the intrafollicular endocrine milieu. No LUFs were induced by reLH or PGF2α treatment; however, LUFs were induced in 100% of mares using FM. Intrafollicular PGF2α metabolite, PGF2α, and PGE2 were lower and the ratio of PGE2:PGF2α was higher in the induced LUF group. Higher levels of intrafollicular E2 and total primary sex steroids were observed in the induced LUF group along with a tendency for higher levels of GH, cortisol, and T; however, LH, PRL, VEGF-A, and NO did not differ between groups. In conclusion, this study reveals part of the intrafollicular endocrine milieu and the association of prostaglandins in LUF formation, and indicates that the mare might be an appropriate model for studying the poorly understood LUF syndrome

    In-depth quantitative proteomics uncovers specie-specific metabolic programs in Leishmania (Viannia) species

    Author summary Leishmania braziliensis,L.panamensis, andL.guyanensisare responsible for most of the cases of tegumentary leishmaniasis (TL) in the Americas. These species are associated with a variety of clinical manifestations of TL ranging from self-healing localized cutaneous lesions to disseminated and mucocutaneous presentations that may result in severe oropharyngeal mutilation. Here, we report a comprehensive quantitative comparison of the proteome of those species. Assessment of absolute titers of similar to 7000 proteins revealed a very clear differentiation among them. Significant differences in energy metabolism, membrane proteins, transporters, and lipids are contributing for species-specific traits and provide rich substrate for exploring new molecules for diagnosing purposes. Leishmaniaspecies are responsible for a broad spectrum of diseases, denominated Leishmaniasis, affecting over 12 million people worldwide. During the last decade, there have been impressive efforts for sequencing the genome of most of the pathogenicLeishmaniaspp. as well as hundreds of strains, but large-scale proteomics analyses did not follow these achievements and theLeishmaniaproteome remained mostly uncharacterized. Here, we report a comprehensive comparative study of the proteomes of strains representingL.braziliensis,L.panamensisandL.guyanensisspecies. Proteins extracted by SDS-mediated lysis were processed following the multi-enzyme digestion-filter aided sample preparation (FASP) procedure and analysed by high accuracy mass spectrometry. "Total Protein Approach" and "Proteomic Ruler" were applied for absolute quantification of proteins. Principal component analysis demonstrated very high reproducibility among biological replicates and a very clear differentiation of the three species. Our dataset comprises near 7000 proteins, representing the most completeLeishmaniaproteome yet known, and provides a comprehensive quantitative picture of the proteomes of the three species in terms of protein concentration and copy numbers. Analysis of the abundance of proteins from the major energy metabolic processes allow us to highlight remarkably differences among the species and suggest that these parasites depend on distinct energy substrates to obtain ATP. WhereasL.braziliensisrelies the more on glycolysis,L.panamensisandL.guyanensisseem to depend mainly on mitochondrial respiration. These results were confirmed by biochemical assays showing opposite profiles for glucose uptake and O(2)consumption in these species. In addition, we provide quantitative data about different membrane proteins, transporters, and lipids, all of which contribute for significant species-specific differences and provide rich substrate for explore new molecules for diagnosing purposes.     Solidification of small para-H2 clusters at zero temperature

    We have determined the ground-state energies of para-H2_2 clusters at zero temperature using the diffusion Monte Carlo method. The liquid or solid character of each cluster is investigated by restricting the phase through the use of proper importance sampling. Our results show inhomogeneous crystallization of clusters, with alternating behavior between liquid and solid phases up to N=55. From there on, all clusters are solid. The ground-state energies in the range N=13--75 are established and the stable phase of each cluster is determined. In spite of the small differences observed between the energy of liquid and solid clusters, the corresponding density profiles are significantly different, feature that can help to solve ambiguities in the determination of the specific phase of H2_2 clusters.Comment: 17 pages, accepted for publication in J.     Thin helium film on a glass substrate

    We investigate by Monte Carlo simulations the structure, energetics and superfluid properties of thin helium-four films (up to four layers) on a glass substrate, at low temperature. The first adsorbed layer is found to be solid and "inert", i.e., atoms are localized and do not participate to quantum exchanges. Additional layers are liquid, with no clear layer separation above the second one. It is found that a single helium-three impurity resides on the outmost layer, not significantly further away from the substrate than helium-four atoms on the same layer.Comment: Six figures, submitted for publication to the Journal of Low Temperature Physic
