    Line Optical Tweezers Instrument for Measuring Nanoscale Interactions and Kinetics

    We describe an optical tweezers instrument for measuring short-ranged colloidal interactions, based on a combination of a continuous wave line optical tweezers, high speed video microscopy, and laser illumination. Our implementation can measure the separation of two nearly contacting microspheres to better than 4 nm at rates in excess of 10 kHz. A simple image analysis algorithm allows us to sensibly remove effects from diffraction blurring and microsphere image overlap for separations ranging from contact to at least 100 nm. The result is a versatile instrument for measuring steric, chemical and single-molecular interactions and dynamics, with a force resolution significantly better than achievable with current atomic force microscopy. We demonstrate the effectiveness of the instrument with measurements of the pair interactions and dynamics of microspheres in the presence of transient molecular bridges of DNA or surfactant micelles

    Fluid Mechanical and Electrical Fluctuation Forces in Colloids

    Fluctuations in fluid velocity and fluctuations in electric fields may both give rise to forces acting on small particles in colloidal suspensions. Such forces in part determine the thermodynamic stability of the colloid. At the classical statistical thermodynamic level, the fluid velocity and electric field contributions to the forces are comparable in magnitude. When quantum fluctuation effects are taken into account, the electric fluctuation induced van der Waals forces dominate those induced by purely fluid mechanical motions. The physical principles are applied in detail for the case of colloidal particle attraction to the walls of the suspension container and more briefly for the case of forces between colloidal particles.Comment: ReVTeX format, one *.eps figur

    The specific binding of complementary DNA strands has been suggested as an ideal method for directing the controlled self-assembly of microscopic objects. We report the first direct measurements of such DNA-induced interactions between colloidal microspheres, as well as the first colloidal crystals assembled using them. The interactions measured with our optical tweezer method can be modeled in detail by well-known statistical physics and chemistry, boding well for their application to directed selfassembly. The microspheres’ binding dynamics, however, have a surprising power-law scaling that can significantly slow annealing and crystallization

    We describe direct imaging measurements of the collective and relative diffusion of two colloidal spheres near a flat plate. The bounding surface modifies the spheres' dynamics, even at separations of tens of radii. This behavior is captured by a stokeslet analysis of fluid flow driven by the spheres' and wall's no-slip boundary conditions. In particular, this analysis reveals surprising asymmetry in the normal modes for pair diffusion near a flat surface.Comment: 4 pages, 4 figure

    The Dynamics of Credit Spreads in Hotel Mortgages and Signaling Implications

    This paper investigates the loan pricing of risk in a market with short term leases (hotels) relative to longer term leases (office properties) with respect to how news on the economy, capital and real estate markets is incorporated in loan pricing using a vector autoregression (VAR) framework. The hotel loan pricing data provides a unique laboratory to study loan pricing adjustments given the short-term nature of the hotel leases. We examine the information content of hotel credit spreads in two stages. After establishing the impact of economic variables on loan pricing and the informational content of the incremental risk spread, we next examine how loan pricing adjusts in response to expected delinquencies. We find that improvement in general economic conditions, an increase in forward looking corporate profitability, an increase in capital availability and/or an increase in the demand for hotel services forecast a decline in the hotel risk premium differential. Thus, the relative loan prices—the spread—reflect systematic risk. We also find that hotel spreads themselves contain important economic information. Unexpected increases in hotel spreads predict hotel delinquencies. In other words, lenders appear to set interest rates on hotel mortgages in anticipation of hotel delinquencies and foreclosures in future periods. Lenders do not appear to consider past delinquencies in their setting their rate

    Blubber Transciptome Response to Acute Stress Axis Activation Involves Transient Charges in Adipogenesis and Lipolysis in Fast-Adapted Marine Mammal

    Stress can compromise an animal\u27s ability to conserve metabolic stores and participate in energy-demanding activities that are critical for fitness. Understanding how wild animals, especially those already experiencing physiological extremes (e.g. fasting), regulate stress responses is critical for evaluating the impacts of anthropogenic disturbance on physiology and fitness, key challenges for conservation. However, studies of stress in wildlife are often limited to baseline endocrine measurements and few have investigated stress effects in fasting-adapted species. We examined downstream molecular consequences of hypothalamic-pituitary-adrenal (HPA) axis activation by exogenous adrenocorticotropic hormone (ACTH) in blubber of northern elephant seals due to the ease of blubber sampling and its key role in metabolic regulation in marine mammals. We report the first phocid blubber transcriptome produced by RNAseq, containing over 140,000 annotated transcripts, including metabolic and adipocytokine genes of interest. The acute response of blubber to stress axis activation, measured 2 hours after ACTH administration, involved highly specific, transient (lasting \u3c24 \u3ehours) induction of gene networks that promote lipolysis and adipogenesis in mammalian adipocytes. Differentially expressed genes included key adipogenesis factors which can be used as blubber-specific markers of acute stress in marine mammals of concern for which sampling of other tissues is not possible

    Strong Attraction between Charged Spheres due to Metastable Ionized States

    We report a mechanism which can lead to long range attractions between like-charged spherical macroions, stemming from the existence of metastable ionized states. We show that the ground state of a single highly charged colloid plus a few excess counterions is overcharged. For the case of two highly charged macroions in their neutralizing divalent counterion solution we demonstrate that, in the regime of strong Coulomb coupling, the counterion clouds are very likely to be unevenly distributed, leading to one overcharged and one undercharged macroion. This long-living metastable configuration in turn leads to a long range Coulomb attraction.Comment: REVTEX-published versio

    Microrheology, stress fluctuations and active behavior of living cells

    We report the first measurements of the intrinsic strain fluctuations of living cells using a recently-developed tracer correlation technique along with a theoretical framework for interpreting such data in heterogeneous media with non-thermal driving. The fluctuations' spatial and temporal correlations indicate that the cytoskeleton can be treated as a course-grained continuum with power-law rheology, driven by a spatially random stress tensor field. Combined with recent cell rheology results, our data imply that intracellular stress fluctuations have a nearly 1/ω21/\omega^2 power spectrum, as expected for a continuum with a slowly evolving internal prestress.Comment: 4 pages, 2 figures, to appear in Phys.
