    The Co-management Survey report: an executive summary

    Co-management is typically known to be a resource management system that shares managerial responsibility between the state and other stakeholders of a resource. In the case of Lake Victoria, one would expect the state to be represented by the fisheries departments of Kenya, Uganda and Tanzania, while stakeholder groups may comprise fishing communities, fish processing factories and municipalities. Taking that into account, the survey's objectives were defined as: (a) To identify the difficulties and impracticalities inherent in implementing state-based regulations via a "top-down" management strategy. (b) To assess the prevalence of community-based institutions that either seek to regulate the fishery or have the potential to be used to regulate it. (c) To identify ways in which community-based regulatory and monitory systems may be established, and how these will fare over time. (d) To identify roles for national Fisheries Departments, industrial fish processors and other stakeholders. (e) To develop well-founded policy suggestions for the establishment of a co-management framework to manage the fisheries of Lake Victoria

    This paper examines the data recently collected from Lake Victoria (SEDAWOG, 2000a). In the first section, it provides the results from the survey that examines fishermen's perceptions of the status of the resource base. In the second section, it starts with an examination of boundaries, followed by an examination of these data insofar as they relate to fishermen's understanding of roles. We conclude the paper with a brief discussion of what these findings mean for fisheries management on Lake Victoria in particular, and our understanding of co-managerial applications more generall

    Reliable in silico ranking of engineered therapeutic TCR binding affinities with MMPB/GBSA

    Accurate and efficient in silico ranking of proteinprotein binding affinities is useful for protein design with applications in biological therapeutics. One popular approach to rank binding affinities is to apply the molecular mechanics Poisson-Boltzmann/generalized Born surface area (MMPB/ GBSA) method to molecular dynamics (MD) trajectories. Here, we identify protocols that enable the reliable evaluation of T-cell receptor (TCR) variants binding to their target, peptide-human leukocyte antigens (pHLAs). We suggest different protocols for variant sets with a few (<= 4) or many mutations, with entropy corrections important for the latter. We demonstrate how potential outliers could be identified in advance and that just 5-10 replicas of short (4 ns) MD simulations may be sufficient for the reproducible and accurate ranking of TCR variants. The protocols developed here can be applied toward in silico screening during the optimization of therapeutic TCRs, potentially reducing both the cost and time taken for biologic development

    Rabbit and Human Non-Keratinising Stratified Squamous Oesophageal Epithelium Displays Similar Microridge Structure by Scanning Electron Microscopy

    Since the oesophageal epithelium of common laboratory animals, rats and mice, is keratinized it is unsuitable for comparison with typical non-keratinized stratified squamous human epithelium. It is thus important to find a suitable animal model for the study of human oesophageal tissue changes. This study investigated the microridge structure of immature and adult rabbit specimens, and adult human biopsies by scanning electron microscopy and morphometry. The investigation revealed a similarity between typical squamous human and adult rabbit oesophageal mucosal epithelium. While human epithelium specimens subdivided into two other groups (non-typical squamous and non-squamous); all typical squamous human biopsies were from patients who had normal endoscopy reports and no reflux symptoms. The surface cells of typical squamous human epithelium displayed complex microridge patterns (64% of cell surface) but patterns in non-typical squamous specimens were more variable (38%) (P \u3c 0.001) and cell boundaries less obvious. Rabbit squames displayed clear microridge patterns with an elevation in the percentage of cell surface covered by microridges, with increasing age, from immature to adult specimens (P \u3c 0.001). There was no statistically significant differences between adult rabbit, and \u27typical squamous\u27 human biopsies (range 51-65%), results which suggest potential use of a rabbit model to study changes in human oesophageal tissue

    Improved timed-mating, non-invasive method using fewer unproven female rats with pregnancy validation via early body mass increases

    For studies requiring accurate conception-timing, reliable, efficient methods of detecting oestrus reduce time and costs, whilst improving welfare. Standard methods use vaginal cytology to stage cycle, and breeders are paired–up using approximately five proven females with proven males to achieve at least one conception on a specific day. We describe an alternative, fast, consistent, non-invasive method of timed-mating using detection of lordosis behaviour in Wistar and Lister-Hooded rats that used unproven females with high success rates. Rats under reverse-lighting had body masses recorded pre-mating, day (d) 3-4, d8, d10 and d18 of pregnancy. Using only the presence of the oestrus dance to time-mate females for 24-hrs, 89% Wistar and 88% Lister-Hooded rats successfully conceived. We did not observe behavioural oestrus in Sprague-Dawleys without males present. Significant body mass increases following mating distinguished pregnant from non-pregnant rats, as early as d4 of pregnancy (10% ± 1.0 increase cf 3% ± 1.2). The pattern of increases throughout gestation was similar for all pregnant rats until late pregnancy, when there were smaller increases for primi- and multiparous rats (32% ± 2.5; 25% ± 2.4), whereas nulliparous rats had highest gains (38% ± 1.5). This method demonstrated a distinct refinement of the previous timed-mating common practice used, as disturbance of females was minimised. Only the number required of nulli-, primi- or multiparous rats were mated, and body mass increases validated pregnancy status. This new breeding-management method is now established practice for two strains of rat and resulted in a reduction in animal use
