    Relativistic precession of quantum elliptical states in the Coulomb potential

    A special relativistic perturbation to non-relativistic quantum mechanics is shown to lead to the special relativistic prediction for the rate of precession for quantum states in the Coulomb potential. This behavior is shown using SO(4) coherent states as examples.     Variation in transcriptional regulation of cyclin dependent kinase inhibitor p21(waf1/cip1 )among human bronchogenic carcinomas

    BACKGROUND: Cell proliferation control depends in part on the carefully ordered regulation of transcription factors. The p53 homolog p73, contributes to this control by directly upregulating the cyclin dependent kinase inhibitor, p21(waf1/cip1). E2F1, an inducer of cell proliferation, directly upregulates p73 and in some systems upregulates p21 directly. Because of its central role in controlling cell proliferation, upregulation of p21 has been explored as a modality for treating bronchogenic carcinoma (BC). Improved understanding of p21 transcriptional regulation will facilitate identification of BC tissues that are responsive to p21-directed therapies. Toward this goal, we investigated the role that E2F1 and p73 each play in the transcriptional regulation of p21. RESULTS: Among BC samples (N = 21) p21 transcript abundance (TA) levels varied over two orders of magnitude with values ranging from 400 to 120,000 (in units of molecules/10(6 )molecules β-actin). The p21 values in many BC were high compared to those observed in normal bronchial epithelial cells (BEC) (N = 18). Among all BC samples, there was no correlation between E2F1 and p21 TA but there was positive correlation between E2F1 and p73α (p < 0.001) TA. Among BC cell lines with inactivated p53 and wild type p73 (N = 7) there was positive correlation between p73α and p21 TA (p < 0.05). Additionally, in a BC cell line in which both p53 and p73 were inactivated (H1155), E2F1 TA level was high (50,000), but p21 TA level was low (470). Transiently expressed exogenous p73α in the BC cell line Calu-1, was associated with a significant (p < 0.05) 90% increase in p21 TA and a 20% reduction in E2F1 TA. siRNA mediated reduction of p73 TA in the N417 BC cell line was associated with a significant reduction in p21 TA level (p < 0.01). CONCLUSION: p21 TA levels vary considerably among BC patients which may be attributable to 1) genetic alterations in Rb and p53 and 2) variation in TA levels of upstream transcription factors E2F1 and p73. Here we provide evidence that p73 upregulates p21 TA in BC tissues and upregulated p21 TA may result from E2F1 upregulation of p73 but not from E2F1 directly

    Beryllium-specific CD4\u3csup\u3e+\u3c/sup\u3e T cells induced by chemokine neoantigens perpetuate inflammation

    Discovering dominant epitopes for T cells, particularly CD4+ T cells, in human immune-mediated diseases remains a significant challenge. Here, we used bronchoalveolar lavage (BAL) cells from HLA-DP2–expressing patients with chronic beryllium disease (CBD), a debilitating granulomatous lung disorder characterized by accumulations of beryllium-specific (Be-specific) CD4+ T cells in the lung. We discovered lung-resident CD4+ T cells that expressed a disease-specific public CDR3β T cell receptor motif and were specific to Be-modified self-peptides derived from C-C motif ligand 4 (CCL4) and CCL3. HLADP2–CCL/Be tetramer staining confirmed that these chemokine-derived peptides represented major antigenic targets in CBD. Furthermore, Be induced CCL3 and CCL4 secretion in the lungs of mice and humans. In a murine model of CBD, the addition of LPS to Be oxide exposure enhanced CCL4 and CCL3 secretion in the lung and significantly increased the number and percentage of CD4+ T cells specific for the HLA-DP2–CCL/Be epitope. Thus, we demonstrate a direct link between Be-induced innate production of chemokines and the development of a robust adaptive immune response to those same chemokines presented as Be-modified self-peptides, creating a cycle of innate and adaptive immune activation

    A crossover randomised controlled trial of oral mandibular advancement devices for obstructive sleep apnoea-hypopnoea (TOMADO)

    Rationale Mandibular advancement devices (MADs) are used to treat obstructive sleep apnoea-hypopnoea syndrome (OSAHS) but evidence is lacking regarding their clinical and cost-effectiveness in less severe disease. Objectives To compare clinical- and cost-effectiveness of a range of MADs against no treatment in mild to moderate OSAHS. Measurements and methods This open-label, randomised, controlled, crossover trial was undertaken at a UK sleep centre. Adults with Apnoea-Hypopnoea Index (AHI) 5–<30/h and Epworth Sleepiness Scale (ESS) score ≥9 underwent 6 weeks of treatment with three nonadjustable MADs: self-moulded (SleepPro 1; SP1); semi-bespoke (SleepPro 2; SP2); fully-bespoke MAD (bMAD); and 4 weeks no treatment. Primary outcome was AHI scored by a polysomnographer blinded to treatment. Secondary outcomes included ESS, quality of life, resource use and cost. Main results 90 patients were randomised and 83 were analysed. All devices reduced AHI compared with no treatment by 26% (95% CI 11% to 38%, p=0.001) for SP1, 33% (95% CI 24% to 41%) for SP2 and 36% (95% CI 24% to 45%, p<0.001) for bMAD. ESS was 1.51 (95% CI 0.73 to 2.29, p<0.001, SP1) to 2.37 (95% CI 1.53 to 3.22, p<0.001, bMAD) lower than no treatment (p<0.001 for all). Compliance was lower for SP1, which was the least preferred treatment at trial exit. All devices were cost-effective compared with no treatment at a £20 000/quality-adjusted life year (QALY) threshold. SP2 was the most cost-effective up to £39 800/QALY. Conclusions Non-adjustable MADs achieve clinically important improvements in mild to moderate OSAHS and are cost-effective

    The impact of networks on clinical trials in the United Kingdom

    The conduct of clinical trials in the UK has been affected by the recent introduction of managed clinical networks, clinical research networks and rigorous governance regulations. This commentary considers the challenges that these changes have posed for clinical triallists in the UK, based on experiences derived in the conduct of a multicentre neonatal clinical trial under the conditions that now prevail. We conclude that the considerable skills and knowledge that are now required to be an effective Principal Investigator should be recognised and that application processes, including issuing honorary contracts, should be simplified and centralised

    FUSE Observations of Molecular Hydrogen in Translucent Interstellar Clouds: II. The Line of Sight Toward HD 110432

    We report the second study from the FUSE survey of molecular hydrogen in translucent clouds, for the line of sight toward HD 110432. This star lies beyond the Coalsack dark nebula, and with E(B-V) = 0.40, and A_V = 1.32 this line of sight bridges the gap between less extinguished diffuse cloud lines of sight with A_V \sim 1, such as Zeta Oph, and the translucent clouds with A_V \gtrsim 2 such as HD 73882. Through profile fitting and a curve-of-growth analysis, we have derived rotational populations for H2 for J = 0--7. The line of sight has a total molecular hydrogen column density, log N(H2) = 20.68 \pm 0.05 cm^{-2}, nearly identical to that toward Zeta Oph, but a factor of three less than HD 73882. The ratio of N(J=1) to N(J=0) yields a kinetic temperature T_{kin} = 63 \pm 7 K, similar to other lines of sight with A_V \gtrsim 1. The high-J lines show considerable excitation above this temperature. The high-J excitation is similar to that toward Zeta Oph, but much smaller than that toward HD 73882. Chemical modeling indicates that the physical conditions in the cloud(s) are very similar to those in the cloud(s) toward Zeta Oph. An analysis of IUE spectra of the Lyman-alpha line gives log N(H I) = 20.85 \pm 0.15 cm^{-2}. Combined with N(H2), we derive a hydrogen molecular fraction, f_{H2} = 0.58 pm 0.12, statistically identical to that found for the lines of sight toward Zeta Oph and HD 73882. We also report column densities for the HD and CO molecules. From the combined measurements of hydrogen and carbon- containing molecules, the line of sight toward HD 110432 appears quite similar to the diffuse cloud line of sight toward Zeta Oph, and quite dissimilar to the translucent cloud line of sight toward HD 73882.Comment: 28 pages, 7 figures, accepted for publication in The Astrophysical Journa
