    Inclusive Instructional Leadership: A Quasi-Experimental Study of a Professional Development Program for Principals

    This quasi-experiment tested whether or not a statewide professional development program for principals yielded measurable changes in self-reported attitudes and practices. The two-year program combined training events at central and regional locations with on-site coaching. The program attempted (1) to change attitudes toward inclusion, broadly understood and grounded in social justice, and (2) to cultivate practices constituting inclusive instructional leadership. The program positions the practices as efforts to serve all students well (i.e., including students of color, English Language Learners, and students with disabilities). This quasi-experiment used a post-only nonequivalent group design with propensity score matching to equate treatment and control groups. Dependent measures were principalsā€™ attitudes towards inclusion and self-reported use of inclusive instructional leadership practices. Treatment group members included 56 participating principals; control group members were 56 non-participating principals matched (one-to-one matching) via the R optimal match routine. Comparison of attitude scores exhibited a statistically significant effect favoring the treatment group (ES=0.47). Also observed was a statistically significant effect (ES=.38) for one of the nine practice itemsā€”principalsā€™ work with teachers on collaborative problem solving and professional learning. None of the observed values for the 21 items (i.e., across both scales, attitudes and practices) favored the control group

    Evaluating nursing students' engagement in an online course using flipped virtual classrooms

    Flipped classroom models allocate more time for active learning approaches compared with more traditional pedagogies, however what is less clear with the utilisation of flipped learning is evidence to support whether students in flipped classes are given more opportunities to develop higher order thinking skills (HOTs) to effect deep learning compared with the traditional ways of teaching. Focussing on this gap, this study compares on campus and off campus student engagement in two courses using different deliveries: online face-to-face (f2f) mixed mode (on campus students attend traditional f2f on campus classes and off campus students study exclusively online) versus fully online mode, utilising flipped classes (all student study off campus engaging in flipped virtual classes). Final course grades were similar for both deliveries; however, the study suggests flipped classes offered students more opportunities to develop HOTs and engage more deeply in the learning process. Studentsā€™ evaluations of the online flipped delivery were mixed, with those students previously enrolled exclusively as on campus, particularly dissatisfied with fully online delivery and virtual class tutor experience. Recommendations are made concerning both the timing of the introduction of fully online delivery in a program and the need for continual up-skilling of staff who teach in online environments

    To Go or Not to Go: Exploring brain activation during response inhibition reading tasks

    Objective: Response inhibition is an understudied component of reading that aids in the selection of appropriate responses amidst complicated tasks. Our objective was to explore the contribution of brain regions associated with response inhibition processing in reading tasks that vary in difficulty of response inhibition. Method: Participants (N = 15) completed two go/no-go reading tasks while in a functional magnetic resonance imaging (fMRI) scanner, with the instructions to ā€œname aloud the letter strings that spell a real word.ā€ For the minimal response inhibition condition, the foils, which are stimuli that should not be repsonded to, were nonwords with unfamiliar spelling and sound (e.g., ā€œbinkā€). For the maximal response inhibition condition, the foils were pseudohomophones with unfamiliar spelling but familiar sound (e.g., ā€œpyntā€). The following brain regions associated with decision-making processes were analyzed: the anterior cingulate cortex (ACC), the dorsomedial prefrontal cortex (DMPFC), the inferior frontal gyrus (IFG), the middle temporal gyrus (MTG), the middle occipital gyrus (MOG), and the posterior insula (PI). Results: Significant differences in activation within the nonword task were found for the DMPFC and the PI (the ACC approached significance). Significant differences in activation within the pseudohomophone task were found for the ACC, the MTG, and the PI. The IFG was found to be greatly activated for all words that had familiar phonemes (sounds). The MOG was found to be activated across all tasks. Conclusion: We provide evidence for differential response inhibition processing in the decision-making network during reading tasks. This work is a necessary step in better understanding response inhibition ability for individuals with and without reading impairments

    Developmental expression patterns of six6: a gene linked with spawning ecotypes in Atlantic salmon

    The Atlantic salmon has been studied extensively, particularly as a model for understanding the genetic and environmental contributions to the evolution and development of life history traits. Expression pattern analysis in situ, however, is mostly lacking in salmon. We examine the embryonic developmental expression of six6, a candidate gene previously identified to be associated with spawning ecotypes and age at sexual maturity, in Atlantic salmon. Six6 is a member of the sine oculis homeobox family of transcription factors and is known to regulate eye and brain development in other vertebrates. We assay the expression of this gene in embryonic Atlantic salmon Salmo salar by whole-mount in situ hybridization. In line with earlier studies in other vertebrate species, we find conserved expression in the developing brain and sensory organs, including optic and olfactory primordia. However, we also find previously unreported domains of expression that suggest additional roles in axial and appendicular development, cardiovascular, intestinal, and sensory organogenesis. Each of these systems are important in the sensory ecology of Atlantic salmon, suggesting it is plausible that six6 may have pleiotropic roles in this complex phenotype.Peer reviewe

    Chlamydia Hijacks ARF GTPases To Coordinate Microtubule Posttranslational Modifications and Golgi Complex Positioning.

    The intracellular bacterium Chlamydia trachomatis develops in a parasitic compartment called the inclusion. Posttranslationally modified microtubules encase the inclusion, controlling the positioning of Golgi complex fragments around the inclusion. The molecular mechanisms by which Chlamydia coopts the host cytoskeleton and the Golgi complex to sustain its infectious compartment are unknown. Here, using a genetically modified Chlamydia strain, we discovered that both posttranslationally modified microtubules and Golgi complex positioning around the inclusion are controlled by the chlamydial inclusion protein CT813/CTL0184/InaC and host ARF GTPases. CT813 recruits ARF1 and ARF4 to the inclusion membrane, where they induce posttranslationally modified microtubules. Similarly, both ARF isoforms are required for the repositioning of Golgi complex fragments around the inclusion. We demonstrate that CT813 directly recruits ARF GTPases on the inclusion membrane and plays a pivotal role in their activation. Together, these results reveal that Chlamydia uses CT813 to hijack ARF GTPases to couple posttranslationally modified microtubules and Golgi complex repositioning at the inclusion.IMPORTANCEChlamydia trachomatis is an important cause of morbidity and a significant economic burden in the world. However, how Chlamydia develops its intracellular compartment, the so-called inclusion, is poorly understood. Using genetically engineered Chlamydia mutants, we discovered that the effector protein CT813 recruits and activates host ADP-ribosylation factor 1 (ARF1) and ARF4 to regulate microtubules. In this context, CT813 acts as a molecular platform that induces the posttranslational modification of microtubules around the inclusion. These cages are then used to reposition the Golgi complex during infection and promote the development of the inclusion. This study provides the first evidence that ARF1 and ARF4 play critical roles in controlling posttranslationally modified microtubules around the inclusion and that Chlamydia trachomatis hijacks this novel function of ARF to reposition the Golgi complex

    Dynamics of Polynesian Subsistence: Insights from Archaeofauna and Stable Isotope Studies, Aitutaki, Southern Cook Islands.

    v. ill. 23 cm.QuarterlyHuman colonists of Remote Oceania readily took advantage of the naive virgin fauna encountered on previously uninhabited islands, a bounty that was quickly depleted. Subsequent developments in Polynesian subsistence economies were more subtle, varied, and complex. These features are illustrated in a comparison of two quite different subsistence archives from the postcolonization period: archaeofaunal assemblages and stable isotope (d13C and d15N) records of humans, pigs, and dogs from the same archaeological contexts. The samples come from four stratified sites, with a total of 22 distinct occupational strata that represent a 600-year period on the small (18.4 km2) almost-atoll of Aitutaki in the southern Cook Islands. Benefits and challenges of integrating these quite different records are considered in the context of specific findings, with implications for subsistence studies elsewhere. In particular, differences in formation processes, taxonomic resolution, and contrasting spatial and temporal scales represented by each record are highlighted. A complex, multiscalar picture of subsistence change emerges, showing variability within and across the three species and the two subsistence archives. Findings support prior interpretations that established (not colonial) settlements are represented by the currently known Aitutaki archaeological record. Within the relatively stable and largely anthropogenic food web, humans occupy a central position throughout the sequence. Through time, a reduction in fishing and decreased consumption of marine carnivores is indicated; these changes are likely to be an outcome of both repeated storm events and considerable shoreline disruption in the fourteenth century A.D., and cultural decisions about the relative costs and benefits of various fishing activities vis-a`-vis other subsistence needs. An apparent reduction in variability of pig diets in late prehistory could reflect interspecific competition between pigs and their human managers, although small sample sizes constrain interpretations. Overall, use of two quite different subsistence archives provides a more robust, but also more complex, view of subsistence change across individuals and communities on Aitutaki

    Strong regulatory effects of vgll3 genotype on reproductive axis gene expression in juvenile male Atlantic salmon

    Age at maturity is a major contributor to the diversity of life history strategies in organisms. The process of maturation is influenced by both genetics and the environment, and includes changes in levels of sex hormones and behavior, but the specific factors leading to variation in maturation timing are poorly understood. gnrh1 regulates the transcription of gonadotropin genes at pubertal onset in many species, but this gene is lacking in certain teleost species including Atlantic salmon (Salmo salar), which raises the possibility of the involvement of other important regulatory factors during this process. Earlier research has reported a strong association of alternative alleles of the vgll3 gene with maturation timing in Atlantic salmon, suggesting it as a potential candidate regulating reproductive axis genes. Here, we investigated the expression of reproductive axis genes in one-year-old Atlantic salmon males with immature gonads and different vgll3 genotypes during the spawning period. We detected strong vgll3 genotype-dependent differential expression of reproductive axis genes (such as fshb, lhb, amh and igf3) tested in the pituitary, and testis. In addition, we observed differential expression of jun (ap1) and nr5a1b (sf1), potential upstream regulators of gonadotropins in the pituitary, as well as axin2, id3, insl3, itch, ptgs2a and ptger4b, the downstream targets of amh and igf3 in the testis. Hereby, we provide evidence of strong vgll3 genotype-dependent transcriptional regulation of reproductive axis genes prior to sexual maturation and suggest alternative models for distinct actions of vgll3 genotypes on the related molecular processes.Peer reviewe
