    Stabilisation of BGK modes by relativistic effects

    Context. We examine plasma thermalisation processes in the foreshock region of astrophysical shocks within a fully kinetic and self-consistent treatment. We concentrate on proton beam driven electrostatic processes, which are thought to play a key role in the beam relaxation and the particle acceleration. Our results have implications for the effectiveness of electron surfing acceleration and the creation of the required energetic seed population for first order Fermi acceleration at the shock front. Aims. We investigate the acceleration of electrons via their interaction with electrostatic waves, driven by the relativistic Buneman instability, in a system dominated by counter-propagating proton beams. Methods. We adopt a kinetic Vlasov-Poisson description of the plasma on a fixed Eulerian grid and observe the growth and saturation of electrostatic waves for a range of proton beam velocities, from 0.15c to 0.9c. Results. We can report a reduced stability of the electrostatic wave (ESW) with increasing non-relativistic beam velocities and an improved wave stability for increasing relativistic beam velocities, both in accordance with previous findings. At the highest beam speeds, we find the system to be stable again for a period of ≈160 plasma periods. Furthermore, the high phase space resolution of the Eulerian Vlasov approach reveals processes that could not be seen previously with PIC simulations. We observe a, to our knowledge, previously unreported secondary electron acceleration mechanism at low beam speeds. We believe that it is the result of parametric couplings to produce high phase velocity ESW’s which then trap electrons, accelerating them to higher energies. This allows electrons in our simulation study to achieve the injection energy required for Fermi acceleration, for beam speeds as low as 0.15c in unmagnetised plasma

    Improving evaluation of the distribution and density of immunostained cells in breast cancer using computerized video image analysis

    Quantitation of cell density in tissues has proven problematic over the years. The manual microscopic methodology, where an investigator visually samples multiple areas within slides of tissue sections, has long remained the basic ‘standard’ for many studies and for routine histopathologic reporting. Nevertheless, novel techniques that may provide a more standardized approach to quantitation of cells in tissue sections have been made possible by computerized video image analysis methods over recent years. The present study describes a novel, computer-assisted video image analysis method of quantitating immunostained cells within tissue sections, providing continuous graphical data. This technique enables the measurement of both distribution and density of cells within tissue sections. Specifically, the study considered immunoperoxidase-stained tumor infiltrating lymphocytes within breast tumor specimens, using the number of immunostained pixels within tissue sections to determine cellular density and number. Comparison was made between standard manual graded quantitation methods and video image analysis, using the same tissue sections. The study demonstrates that video image techniques and computer analysis can provide continuous data on cell density and number in immunostained tissue sections, which compares favorably with standard visual quantitation methods, and may offer an alternative

    Achieving equal standards in medical student education: is a national exit examination the answer?

There is substantial variability in the content of medical school curricula, and the process of curriculum change is becoming more challenging because of intense competition for time and space in the course. A national exit examination could provide a uniform standard of assessment for all medical school graduates in Australia, as well as foreign graduates applying to work in Australia.     Immune profiling in human breast cancer using high-sensitivity detection and analysis techniques

DESIGN: Cohort comparative analysis studies of breast tissue. SETTING: Human hospital and laboratory healthcare facilities. PARTICIPANTS: Women over 18 years. MAIN OUTCOME MEASURES: Evaluation of the comparative immunophenotype of human breast carcinoma and normal breast tissues. RESULTS: Leukocyte density and specific subgroups of lymphocytes and macrophages were generally higher in breast cancers compared to normal breast tissues. CD3, CD4, CD45RO, CD45RA(2H4), CD45 and HLA Class II (on TIL) were significantly expressed on breast tumour tissues compared with normal tissues (p < .01). Some 30% of T-cells were γδ-TCR positive, but the majority were αβ-TCR in type. CD19 (B-cell), CD14 (FMC32 and 33) and HLA Class I levels (epithelial and TIL) showed no significant differences. IL-2α receptor expression was low or absent on most TIL. CONCLUSIONS: High-sensitivity and image analysis techniques permitted accurate characterisation of the TIL infiltrate for immune profiling. Breast carcinoma showed predominance of CD4 T-cells of mainly memory phenotype. Normal breast tissues showed low leukocyte infiltration.     Quantitative trait loci for yield and grain plumpness relative to maturity in three populations of barley (Hordeum vulgare L.) grown in a low rain-fall environment

    Identifying yield and grain plumpness QTL that are independent of developmental variation or phenology is of paramount importance for developing widely adapted and stable varieties through the application of marker assisted selection. The current study was designed to dissect the genetic basis of yield performance and grain plumpness in southern Australia using three doubled haploid (DH) populations developed from crosses between adapted parents that are similar in maturity and overall plant development. Three interconnected genetic populations, Commander x Fleet (CF), Commander x WI4304 (CW), and Fleet x WI4304 (FW) developed from crossing of Australian elite barley genotypes, were used to map QTL controlling yield and grain plumpness. QTL for grain plumpness and yield were analysed using genetic linkage maps made of genotyping-by-sequencing markers and major phenology genes, and field trials at three drought prone environments for two growing seasons. Seventeen QTL were detected for grain plumpness. Eighteen yield QTL explaining from 1.2% to 25.0% of the phenotypic variation were found across populations and environments. Significant QTL x environment interaction was observed for all grain plumpness and yield QTL, except QPlum.FW-4H.1 and QYld.FW-2H.1. Unlike previous yield QTL studies in barley, none of the major developmental genes, including Ppd-H1, Vrn-H1, Vrn-H2 and Vrn-H3, that drive barley adaption significantly affected grain plumpness and yield here. Twenty-two QTL controlled yield or grain plumpness independently of known maturity QTL or genes.     Towards testing of a second-generation bladed receiver

    A bladed receiver design concept is presented which offers a >2% increase in overall receiver efficiency after considering spillage, reflection, emission and convection losses, based on an integrated optical-thermal model, for a design where the working fluid is conventional molten salt operating in the standard 290–565°C temperature range. A novel testing methodology is described, using air and water to test the receiver when molten salt facilities are not available. Technoeconomic analysis shows that the receiver could achieve a 4 AUD/MWhe saving in levelised cost of energy, but only if the bladed receiver design can be implemented at no additional cost

    Filifactor alocis infection and inflammatory responses in the mouse subcutaneous chamber model

    Recent microbiome studies have implicated a role for Filifactor alocis in periodontal disease. In this study, we investigated the colonization and survival properties of F. alocis in a mouse subcutaneous chamber model of infection and characterized host innate immune responses. An infection of 10(9) F. alocis successfully colonized all chambers; however, the infection was cleared after 72 h. F. alocis elicited a local inflammatory response with neutrophils recruited into the chambers at 2 h postinfection along with an increase in levels of the proinflammatory cytokines interleukin 1β (IL-1β), IL-6, and tumor necrosis factor (TNF). F. alocis also induced apoptosis in chamber epithelial cells and neutrophils. Consistent with resolution of infection, neutrophil numbers and cytokine levels returned to baseline by 72 h. Fluorescent in situ hybridization (FISH) and quantitative PCR demonstrated that F. alocis exited the chambers and spread to the spleen, liver, lung, and kidney. Massive neutrophil infiltration was observed in the spleen and lungs, and the recruited neutrophils were in close proximity to the infecting bacteria. Significant epithelial injury was observed in the kidneys. Infection of all tissues was resolved after 7 days. This first in vivo study of the pathogenicity of F. alocis shows that in the chamber model the organism can establish a proinflammatory, proapoptotic local infection which is rapidly resolved by the host concordant with neutrophil influx. Moreover, F. alocis can spread to, and transiently infect, remote tissues where neutrophils can also be recruited

    CD1a-positive infiltrating-dendritic cell density and 5-year survival from human breast cancer

    © Churchill LivingstoneInfiltrating CD1a+ dendritic cells (DCs) have been associated with increased survival in a number of human cancers. This study investigated DC infiltration within breast cancers and the association with survival. Classical established prognostic factors, of tumour size, lymph node status, histological grade, lympho-vascular invasion, the KI-67 (MIB-1) fraction and the Nottingham Prognostic Index (NPI) were also compared. A total of 48 breast cancer patients were followed from the time of surgery and CD1a density analysis for 5 years or until death. Our data set validated previous studies, which show a relationship between survival and the NPI (P<0.001), tumour size (P<0.01) and lymph node status (P<0.05). Although more patients were alive at the 5-year time point in the group with higher CD1a DC density than the lower CD1a DC group, this failed to reach statistical significance at the P=0.05 level.
