    Receiver Mount Design, Transmitter Depth, and Wind Speed Affect Detection Probability of Acoustic Telemetry Transmitters in a Missouri River Tributary

    Background One of the most important considerations for acoustic telemetry study designs is det:ection probability between the transmitter and the receiver. Variation in environmental (i.e., wind and flow) and abiotic (i.e., bathymetry) conditions among aquatic systems can lead to differences in detection probability temporally or between systems. In this study we evaluate the effect of distance, receiver mount design, transmitter depth, and wind speed on detection probabilities of two models of acoustic transmitters in a mid-sized river. InnovaSea V16-6H (hereafter V16) and V13-1L (hereafter V13) tags were deployed in the James River, SD at 0.36 m (deep) and 2.29 m (V16 tag) or 1.98 m (V13 tag; shallow) above the benthic surface downstream of InnovaSea VR2W stationary receivers at distances of 100, 200, or 300 m. We used two receiver mount designs that included a fixed position within a PVC pipe on the downstream side of a bridge piling or a metal frame deployed in the middle of the river channel. Tags were deployed for 72 h at each location, and hourly detections were summarized. We evaluated downstream distance, receiver mount design, tag depth, and wind effects on tag detection using Bayesian logistic regression. Results: Detection probability decreased as distance increased for all combinations of tag types and mount designs and varied from nearly 100% at 100 m to less than 10% at 300 m. The V16 transmitter had greater detection probability by the receiver mounted in the pipe than in the midriver frame. For both mounts, the deep V16 transmitter had greater detection probability than the V16 shallow transmitter. Detection probability of the V13 transmitter was similar between receiver mounts or transmitter depths. Wind speed had a negative impact on detection probabilities of both transmitter types and depths, except the deep V16 transmitter. Conclusions: Deploying acoustic receivers in PVC pipes rather than midriver frames provided greater downstream detection probabilities for V16 transmitters under conditions evaluated in this study. In addition, V16 transmitters had greater detection probabilities when positioned deep within the water column rather than near the surface. We also demonstrated that wind speed can have a negative impact on detection probabilities

    The rise and fall of the king : the correlation between FO Aquarii's low states and the White Dwarf's Spindown

    The intermediate polar FO Aquarii experienced its first-reported low-accretion states in 2016, 2017, and 2018. We establish that these low states occurred shortly after the system's white dwarf (WD) began spinning down, after having spent a quarter-century spinning up. FO Aquarii is the only intermediate polar whose period derivative has undergone a sign change, and it has now done so twice. By combining our spin-pulse timings with previous data, we determine that the WD's spin period has varied quasi-sinusoidally since the system's discovery, and an extrapolation predicts that the white dwarf was spinning down during newly discovered low states in photographic plates from 1964, 1965, and 1974. Thus, FO Aquarii's low states appear to occur exclusively during epochs of spindown. Additionally, our time-series photometry of the 2016-18 low states reveals that the mode of accretion is extremely sensitive to the accretion rate; when the system is fainter than V~14.0, the accretion onto the WD is largely stream-fed, but when it is brighter, it is almost exclusively disk-fed. The system's grazing eclipse remained detectable throughout all observations, confirming the uninterrupted presence of a disk-like structure, regardless of the accretion state. Our observations are consistent with theoretical predictions that during the low states, the accretion disk dissipates into a ring of diamagnetic blobs. Finally, a new XMM-Newton observation from 2017 indicates that the system's anomalously soft X-ray spectrum and diminished X-ray luminosity in the wake of the 2016 low state appear to be long-lasting changes compared to pre-2016 observations.peer-reviewe

    The Gravity Collective: A Search for the Electromagnetic Counterpart to the Neutron Star-Black Hole Merger GW190814

    We present optical follow-up imaging obtained with the Katzman Automatic Imaging Telescope, Las Cumbres Observatory Global Telescope Network, Nickel Telescope, Swope Telescope, and Thacher Telescope of the LIGO/Virgo gravitational wave (GW) signal from the neutron star-black hole (NSBH) merger GW190814. We searched the GW190814 localization region (19 deg2^{2} for the 90th percentile best localization), covering a total of 51 deg2^{2} and 94.6% of the two-dimensional localization region. Analyzing the properties of 189 transients that we consider as candidate counterparts to the NSBH merger, including their localizations, discovery times from merger, optical spectra, likely host-galaxy redshifts, and photometric evolution, we conclude that none of these objects are likely to be associated with GW190814. Based on this finding, we consider the likely optical properties of an electromagnetic counterpart to GW190814, including possible kilonovae and short gamma-ray burst afterglows. Using the joint limits from our follow-up imaging, we conclude that a counterpart with an rr-band decline rate of 0.68 mag day1^{-1}, similar to the kilonova AT 2017gfo, could peak at an absolute magnitude of at most 17.8-17.8 mag (50% confidence). Our data are not constraining for ''red'' kilonovae and rule out ''blue'' kilonovae with M>0.5MM>0.5 M_{\odot} (30% confidence). We strongly rule out all known types of short gamma-ray burst afterglows with viewing angles <<17^{\circ} assuming an initial jet opening angle of \sim5.25.2^{\circ} and explosion energies and circumburst densities similar to afterglows explored in the literature. Finally, we explore the possibility that GW190814 merged in the disk of an active galactic nucleus, of which we find four in the localization region, but we do not find any candidate counterparts among these sources.Comment: 86 pages, 9 figure

    The 2021 WHO catalogue of Mycobacterium tuberculosis complex mutations associated with drug resistance: a genotypic analysis.

    Background: Molecular diagnostics are considered the most promising route to achievement of rapid, universal drug susceptibility testing for Mycobacterium tuberculosis complex (MTBC). We aimed to generate a WHO-endorsed catalogue of mutations to serve as a global standard for interpreting molecular information for drug resistance prediction. Methods: In this systematic analysis, we used a candidate gene approach to identify mutations associated with resistance or consistent with susceptibility for 13 WHO-endorsed antituberculosis drugs. We collected existing worldwide MTBC whole-genome sequencing data and phenotypic data from academic groups and consortia, reference laboratories, public health organisations, and published literature. We categorised phenotypes as follows: methods and critical concentrations currently endorsed by WHO (category 1); critical concentrations previously endorsed by WHO for those methods (category 2); methods or critical concentrations not currently endorsed by WHO (category 3). For each mutation, we used a contingency table of binary phenotypes and presence or absence of the mutation to compute positive predictive value, and we used Fisher's exact tests to generate odds ratios and Benjamini-Hochberg corrected p values. Mutations were graded as associated with resistance if present in at least five isolates, if the odds ratio was more than 1 with a statistically significant corrected p value, and if the lower bound of the 95% CI on the positive predictive value for phenotypic resistance was greater than 25%. A series of expert rules were applied for final confidence grading of each mutation. Findings: We analysed 41 137 MTBC isolates with phenotypic and whole-genome sequencing data from 45 countries. 38 215 MTBC isolates passed quality control steps and were included in the final analysis. 15 667 associations were computed for 13 211 unique mutations linked to one or more drugs. 1149 (7·3%) of 15 667 mutations were classified as associated with phenotypic resistance and 107 (0·7%) were deemed consistent with susceptibility. For rifampicin, isoniazid, ethambutol, fluoroquinolones, and streptomycin, the mutations' pooled sensitivity was more than 80%. Specificity was over 95% for all drugs except ethionamide (91·4%), moxifloxacin (91·6%) and ethambutol (93·3%). Only two resistance mutations were identified for bedaquiline, delamanid, clofazimine, and linezolid as prevalence of phenotypic resistance was low for these drugs. Interpretation: We present the first WHO-endorsed catalogue of molecular targets for MTBC drug susceptibility testing, which is intended to provide a global standard for resistance interpretation. The existence of this catalogue should encourage the implementation of molecular diagnostics by national tuberculosis programmes.
