    Brazilian gamma detection device for sentinel lymph node biopsy

    OBJECTIVE: To test the effectiveness of an intra-operative gamma detection Brazilian device (IPEN) on sentinel lymph node biopsy (SLNB) procedures. METHODS: Forty melanoma or breast cancer patients with indication for undergoing SLNB were studied. Lymphoscintigraphy was done 2 to 24 hours prior to surgery. Lymphatic mapping with vital dye and gamma detection were performed intraoperatively. For gamma detection Neoprobe ® 1500 was used followed by IPEN (equipment under test) in the first 20 patients and for the remaining half IPEN was used first to verify its ability to locate the sentinel node (SN). Measurements were taken from the radiopharmaceutical product injection site, from SN (in vivo and ex vivo) and from background. It was recorded if the SN was stained or not and if it was found easily by surgeon. RESULTS: There were 33 (82.5%) breast cancer and 7 (17.5%) melanoma patients. Ages varied from 21 to 68 year-old (median age of 46). Sex distribution was 35 (87.5%) women and 5 (12.5%) men. Sentinel node was found in all but one patient. There was no statistical difference between the reasons ex vivo/ background obtained with the measures of both equipments (p=0, 2583-ns). The SN was easily found by the surgeon with both devices. CONCLUSION: The SLNB was successfully performed using either equipment. It was possible to do SLNB with the Brazilian device developed by IPEN without prejudice for the patient.OBJETIVO: Testar a eficácia de equipamento de detecção gama intra-operatória (DGI) desenvolvido pelo IPEN (Brasil), em procedimentos de biópsia de linfonodo sentinela (BLS) no melanoma e no câncer de mama. MÉTODOS: Foram estudados 40 pacientes portadores de melanoma ou câncer de mama com indicação para realização de BLS.Todos pacientes foram submetidos à linfocintilografia e a BLS ocorreu entre 2 a 24 horas após a mesma. Concomitantemente à DGI, realizou-se o mapeamento linfático com corante vital. Foram feitas leituras com o equipamento convencional Neoprobe® 1500 e com o equipamento em teste (IPEN) dos valores de captação do sítio de injeção do radiofármaco, do LS in vivo e ex vivo e da captação de fundo. Foi registrado se o LS estava corado e se o cirurgião teve facilidade para encontrá-lo. Nos primeiros 20 pacientes utilizou-se o equipamento convencional e depois o de teste; nos outros 20, utilizou-se primeiro o equipamento em teste, com objetivo de verificar se o mesmo identificava primariamente o LS. RESULTADOS: Dos quarenta pacientes, 33 eram portadores de tumor de mama e sete de melanoma cutâneo; variação da idade: 21 a 68 anos (mediana= 46 anos); 35 mulheres e 5 homens. Em apenas um paciente o LS não foi encontrado, nem pela DGI nem pelo corante vital. Não houve diferença estatística entre as razões ex vivo/fundo obtidas com os dois equipamentos (p=0, 2583-ns).     Characterisation of the fumarate hydratase repertoire in Trypanosoma cruzi.

    Nifurtimox and benznidazole represent the only treatments options available targeting Chagas disease, the most important parasitic infection in the Americas. However, use of these is problematic as they are toxic and ineffective against the more severe stages of the disease. In this work, we used a multidisciplinary approach to characterise the fumarases from Trypanosoma cruzi, the causative agent of Chagas Disease. We showed this trypanosome expresses cytosolic and mitochondrial fumarases that via an iron-sulfur cluster mediate the reversible conversion of fumarate to S-malate. Based on sequence, biochemical properties and co-factor binding, both T. cruzi proteins share characteristics with class I fumarases, enzymes found in bacteria and some other protozoa but absent from humans, that possess class II isoforms instead. Gene disruption suggested that although the cytosolic or mitochondrial fumarase activities are individually dispensable their combined activity is essential for parasite viability. Finally, based on the mechanistic differences with the human (host) fumarase, we designed and validated a selective inhibitor targeting the parasite enzyme.     Antiaterogenicidad in vitro de extractos del alga marina Halimeda incrassata: actividad antioxidante y estudios de migración en células de la musculatura lisa

    Aim: The aim of this work was to evaluate the in vitro atheroprotective potential of the seaweed Halimeda incrassata in smooth muscle cell migration and lipoprotein oxidation in relation to its antioxidant activity.Material and methods: Antioxidant activity was determinate by DPPH• radical scavenging assay and ORAC method. The inhibitory effect of the aqueous extract on LDL oxidation mediated by Cu2+ ions was determinate by TBARS and conjugated diene quantification. The effect of the seaweed aqueous extract on smooth muscle cell migration was evaluated in MOVAS-1 mouse aortic smooth muscle cell.Results: The inhibitory effect of the aqueous extract on lipoprotein oxidation mediated by Cu2+ was demonstrated. Seaweed extract caused dose-dependent inhibition of TBARS (IC50 = 0.8 mg/mL) and conjugated dienes formation. The seaweed had a high antioxidant activity in the assays performed. The activity could be related to the phenolic content of Halimeda incrassata.Conclusions: In summary, the results of this study represent a further step in the characterization of the atheroprotective action of Halimeda incrassata and indicate the seaweed could be used for a nutraceutical and/or phytoterapeutic application.Objetivos: El objetivo de este trabajo fue evaluar el potencial ateroprotector in vitro del alga Halimeda incrassata en la migración de células de músculo liso de ratón y la oxidación de lipoproteínas en relación con su actividad antioxidante.Material y métodos: La actividad antioxidante fue determinada mediante los métodos de inhibición de radicales DPPH y la Capacidad antioxidante total (ORAC). La actividad inhibitoria de la oxidación de LDL mediada por iones Cu2+ se determinó por la cuantificación de TBARS y dienos conjugados. El efecto del extracto acuoso sobre la migración de las células de músculo liso se evaluó en la línea de células de músculo liso aórtica de ratón MOVAS-1.Resultados: Se demostró el efecto inhibidor del extracto sobre la oxidación de LDL mediada por Cu2+. El extracto del alga causa inhibición dosis-dependiente de la formación de TBARS (IC50 = 0,8 mg/mL) y dienos conjugados. Las algas tuvieron una alta actividad antioxidante en los ensayos realizados y podría estar relacionada con el contenido de compuestos fenólicos.Conclusiones: Los resultados de este trabajo representan un paso más en la caracterización de la acción ateroprotectora de Halimeda incrassata y evidencian sus posibles aplicaciones como nutracéutico y/o fitofármaco

    SOD2 immunoexpression predicts lymph node metastasis in penile cancer

    BACKGROUND: Superoxide dismutase-2 (SOD2) is considered one of the most important antioxidant enzymes that regulate cellular redox state in normal and tumorigenic cells. Overexpression of this enzyme in lung, gastric, colorectal, breast cancer and cervical cancer malignant tumors has been observed. Its relationship with inguinal lymph node metastasis in penile cancer is unknown. METHODS: SOD2 protein expression levels were determined by immunohistochemistry in 125 usual type squamous cell carcinomas of the penis from a Brazilian cancer center. The casuistic has been characterized by means of descriptive statistics. An exploratory logistic regression has been proposed to evaluate the independent predictive factors of lymph node metastasis. RESULTS: SOD2 expression in more than 50% of cells was observed in 44.8% of primary penile carcinomas of the usual type. This expression pattern was associated with lymph node metastasis both in the uni and multivariate analysis. CONCLUSIONS: Our results indicate that SOD2 expression predicts regional lymph node metastasis.     Bird-spiders (Arachnida, Mygalomorphae) as perceived by the inhabitants of the village of Pedra Branca, Bahia State, Brazil

    This paper deals with the conceptions, knowledge and attitudes of the inhabitants of the county of Pedra Branca, Bahia State, on mygalomorph spiders locally known as 'caranguejeiras' (bird-spiders). It is launched here a new filed within ethnozoology: ethnoarachnology, which is defined as the transdisciplinary study of the relationships between human beings and bird-spiders. Data were collected from February to June 2005 by means of open-ended interviews carried out with 30 individuals, which ages ranged from 13 to 86 years old. It was recorded some traditional knowledge regarding the following items: taxonomy, biology, habitat, ecology, seasonality, and behavior. Results show that bird-spiders are classified as "insects". The most commented aspect of the interaction between bird-spiders and inhabitants of Pedra Branca is related to their dangerousness, since they said these spiders are very venomous and can cause health problems. In general, the traditional zoological knowledge of Pedra Branca's inhabitants concerning these spiders is coherent with the academic knowledge

Atualmente esses dispositivos dispõem de terapia antitaquicardia (ATP), cardioversão com baixa energia, desfibrilação com alta energia e função antibradicardia de backup.
