    Clinical significance of epithelial-to-mesenchymal transition in laryngeal carcinoma: Its role in the different subsites

    Background: During epithelial-to-mesenchymal transition, cancer cells lose adhesion capacity gaining migratory properties. The role of the process on prognosis has been evaluated in 50 cases of laryngeal carcinoma. Methods: E-cadherin, N-cadherin, β-catenin, α-catenin, γ-catenin, caveolin-1, and vimentin immunohistochemical expression were evaluated using a double score based on staining intensity and cellular localization. Results: Cytoplasmic E-cadherin and α/γ catenin staining were associated with a decrease in survival, cytoplasmic β-catenin was associated with advanced stage, and N-cadherin and vimentin expression were associated with poor differentiation and tumor relapse. On the basis of cancer cells, epithelial or mesenchymal morphological and immunophenotypic similarity we identified 4 main subgroups correlated with a transition to a more undifferentiated phenotype, which have a different pattern of relapse and survival. Conclusion: The negative prognostic role of epithelial-to-mesenchymal transition has been confirmed and a predictive role in glottic tumors has been suggested, leading us to propose epithelial-to-mesenchymal transition as an additional adverse feature in laryngeal carcinoma

    Selection of an Endophytic Streptomyces sp. Strain DEF09 From Wheat Roots as a Biocontrol Agent Against Fusarium graminearum

    Selection of biological control agents (BCA) profits from an integrated study of the tripartite interactions occurring among the BCA, the plant and the pathogen. The environment plays a crucial role in the efficacy of BCA, therefore, the selection process shall utmost mimic naturally occurring conditions. To identify effective biocontrol strains against Fusarium graminearum, the major cause of Fusarium head blight (FHB) in wheat and deoxynivalenol (DON) accumulation in grains, a workflow consisting of in vitro and in vivo assays was set up. Twenty-one Streptomyces strains, 16 of which were endophytes of different plants, were analyzed. In vitro and in vivo tests characterized their plant growth promoting (PGP) traits. Biocontrol activity against F. graminearum was firstly assessed with a dual culture assay. An in vivo germination blotter assay measured Fusarium foot rot and root rot symptoms (FFR-FRR) reduction as well as growth parameters of the plant treated with the Streptomyces strains. A selected subset of Streptomyces spp. strains was then assessed in a growth chamber measuring FFR symptoms and growth parameters of the wheat plant. The approach led to the identification of an effective Streptomyces sp. strain, DEF09, able to inhibit FHB on wheat in controlled conditions by blocking the spread of the pathogen at the infection site. The results were further confirmed in field conditions on both bread and durum wheat, where DEF09 decreased disease severity up to 60%.     HQR-Scheme: A High Quality and resilient virtual primary key generation approach for watermarking relational data

    Most of the watermarking techniques designed to protect relational data often use the Primary Key (PK) of relations to perform the watermark synchronization. Despite offering high confidence to the watermark detection, these approaches become useless if the PK can be erased or updated. A typical example is when an attacker wishes to use a stolen relation, unlinked to the rest of the database. In that case, the original values of the PK lose relevance, since they are not employed to check the referential integrity. Then, it is possible to erase or replace the PK, compromising the watermark detection with no need to perform the slightest modification on the rest of the data. To avoid the problems caused by the PK-dependency some schemes have been proposed to generate Virtual Primary Keys (VPK) used instead. Nevertheless, the quality of the watermark synchronized using VPKs is compromised due to the presence of duplicate values in the set of VPKs and the fragility of the VPK schemes against the elimination of attributes. In this paper, we introduce the metrics to allow precise measuring of the quality of the VPKs generated by any scheme without requiring to perform the watermark embedding. This way, time waste can be avoided in case of low-quality detection. We also analyze the main aspects to design the ideal VPK scheme, seeking the generation of high-quality VPK sets adding robustness to the process. Finally, a new scheme is presented along with the experiments carried out to validate and compare the results with the rest of the schemes proposed in the literature

    A Double Fragmentation Approach for Improving Virtual Primary Key-Based Watermark Synchronization

    Relational data watermarking techniques using virtual primary key schemes try to avoid compromising watermark detection due to the deletion or replacement of the relation's primary key. Nevertheless, these techniques face the limitations that bring high redundancy of the generated set of virtual primary keys, which often compromises the quality of the embedded watermark. As a solution to this problem, this paper proposes double fragmentation of the watermark by using the existing redundancy in the set of virtual primary keys. This way, we guarantee the right identification of the watermark despite the deletion of any of the attributes of the relation. The experiments carried out to validate our proposal show an increment between 81.04% and 99.05% of detected marks with respect to previous solutions found in the literature. Furthermore, we found out that our approach takes advantage of the redundancy present in the set of virtual primary keys. Concerning the computational complexity of the solution, we performed a set of scalability tests that show the linear behavior of our approach with respect to the processes runtime and the number of tuples involved, making it feasible to use no matter the amount of data to be protected

    THGEM-based detectors for sampling elements in DHCAL: laboratory and beam evaluation

    We report on the results of an extensive R&D program aimed at the evaluation of Thick-Gas Electron Multipliers (THGEM) as potential active elements for Digital Hadron Calorimetry (DHCAL). Results are presented on efficiency, pad multiplicity and discharge probability of a 10x10 cm2 prototype detector with 1 cm2 readout pads. The detector is comprised of single- or double-THGEM multipliers coupled to the pad electrode either directly or via a resistive anode. Investigations employing standard discrete electronics and the KPiX readout system have been carried out both under laboratory conditions and with muons and pions at the CERN RD51 test beam. For detectors having a charge-induction gap, it has been shown that even a ~6 mm thick single-THGEM detector reached detection efficiencies above 95%, with pad-hit multiplicity of 1.1-1.2 per event; discharge probabilities were of the order of 1e-6 - 1e-5 sparks/trigger, depending on the detector structure and gain. Preliminary beam tests with a WELL hole-structure, closed by a resistive anode, yielded discharge probabilities of <2e-6 for an efficiency of ~95%. Methods are presented to reduce charge-spread and pad multiplicity with resistive anodes. The new method showed good prospects for further evaluation of very thin THGEM-based detectors as potential active elements for DHCAL, with competitive performances, simplicity and robustness.
