Cell Wall Composition and Structure Define the Developmental Fate of Embryogenic Microspores in Brassica napus

[EN] Microspore cultures generate a heterogeneous population of embryogenic structures that can be grouped into highly embryogenic structures [exine-enclosed (EE) and loose bicellular structures (LBS)] and barely embryogenic structures [compact callus (CC) and loose callus (LC) structures]. Little is known about the factors behind these different responses. In this study we performed a comparative analysis of the composition and architecture of the cell walls of each structure by confocal and quantitative electron microscopy. Each structure presented specific cell wall characteristics that defined their developmental fate. EE and LBS structures, which are responsible for most of the viable embryos, showed a specific profile with thin walls rich in arabinogalactan proteins (AGPs), highly and low methyl-esterified pectin and callose, and a callose-rich subintinal layer not necessarily thick, but with a remarkably high callose concentration. The different profiles of EE and LBS walls support the development as suspensorless and suspensor-bearing embryos, respectively. Conversely, less viable embryogenic structures (LC) presented the thickest walls and the lowest values for almost all of the studied cell wall components. These cell wall properties would be the less favorable for cell proliferation and embryo progression. High levels of highly methyl-esterified pectin are necessary for wall flexibility and growth of highly embryogenic structures. AGPs seem to play a role in cell wall stiffness, possibly due to their putative role as calcium capacitors, explaining the positive relationship between embryogenic potential and calcium levels. This work was supported by grant PID2020-115763RBI00 to JS-S from Spanish MICINN and by a Juan de la Cierva -Incorporacion Fellowship and a Marie Sklodowska-Curie Individual Fellowship (656579) to PC-M. The C programming language is widely used in computer and industrial engineering. Because of that, such programming language is also widely used as a language to teach programming to industrial engineering students. In Spain, many universities use this language compulsory in the first year, or even in higher courses. Our experience shows that learning computer programming in four months is an arduous task, but curricula require it. Such learning process is also tough by the fact that many students cannot attend classes regularly and, even if they attend, sometimes the class is no longer at the level they require. In this work we develop a series of files in "presentation" format (.ppsx) and videos that allow students to see several explanations about the most complicated programming C topics: functions, arrays, structures, strings, arrays with structures, etc. This multimedia material includes explanations (voice-over), and animations with examples. Students can watch and listen to the explanations whenever and wherever they want (tablet, PC, phone…). Surveys made to students reveal that it is also interesting for students who regularly attend classes, and they prefer to use this course material only at home, outside of regular classes.

Assess the effect that BAT (Bilateral Arm Training) produces on the upper limb motor control in patients with chronic brain injury assessed by the Wolf Motor Function Test (WMFT). Assess if there is a statistically significant correlation between motor control improvement and disability perceived by the Quick Dash questionnaire application. A quasi-experimental study with pre-post treatment measures during three months was performed in a sample of twelve patients with chronic brain injury. Patients received twelve sessions of forty-five minutes from bilateral training with a frequency of three times per week. WMFT and QD were used for the procedure assessment. Outcomes were statistically analyzed by the SPSS v 17.0 software. The study was executed at Polibea. Study´s inclusion criteria were as follows: no serious cognitive deficits, one or both upper limb´s control motor affected, attend to Polibea two times per week and no sensitive aphasia. After the BAT treatment we observed a statistically significant difference on motor control improvement in the WMFT. However the disability perceived through QD was not statistically significant. In conclusion, BAT improves motor control in our patients with chronic acquired brain injury

Manta birostris is a highly migratory species, as well as the largest member of the genus Manta. In 2009, the genus Manta was reevaluated and divided into two species: Manta alfredi and Manta birostris. Manta birostris is broadly distributed, ranging from tropical to semi-temperate oceans. Currently, unregulated fishing for gill rakers and low reproduction rates are reducing subpopulations worldwide. This species is particularly vulnerable to overfishing due to their late maturity, slow gestation periods and long life spans. A large group of M. birostris individuals visits the Isla de la Plata during the months of July to September. The present study characterize the genetic diversity of Isla de la Plata M. birostris individuals using microsatellites. A total of 57 samples collected during three years (2010, 2011 and 2012) were analyzed using eight pairs of heterologous primers. A total of 46 alleles were identified in the eight loci analyzed, with an average of 4,375 alleles per locus. Exclusive alleles were found in three loci (MA14, MA43 and MA30), and three loci (MA21, MA15, MA30) were not in Hardy-Weinberg equilibrium. The genetic diversity indicators analyzed (expected heterozygosity, Shannon index, fixation index) show a high level of genetic variability. The estimation of the population structure of M. birostris using Bayesian inference suggests that the samples studied possibly belong to one population. A greater insight about genetic diversity indexes and the population structure of M. birostris can be achieved through the analysis of more samples, over a broader period of time. . The information of genetic diversity and population structure obtained in this preliminary investigation can be used to establish specific conservation and management programs of this species.

Improved regeneration of eggplant doubled haploids from microspore-derived calli through organogenesis

[EN] Doubled haploid (DH) technology allows for the production of pure lines, useful for plant breeding, through a one-generation procedure that reduces considerably the time and resources needed to produce them. Despite the advantages of microspore culture to obtain DHs, this technique is still insufficiently developed in eggplant, where DHs are produced from microsporederived calli through organogenesis. At present, very little is known on the best in vitro conditions to promote this process. This is why in this work we addressed the optimization of the process of regeneration of eggplant DH plants from microspore-derived calli. We evaluated the effect of different media compositions in the induction of organogenesis, in the promotion of shoot growth and elongation, and in root growth. According to our results, we propose the repeated subculture of the calli in MS medium with 0.2 mg/l IAA and 4 mg/l zeatin to produce shoots, and then the repeated subculture of the excised shoots in basal MS medium to promote their conversion into entire plantlets. This procedure yielded 7.6 plants per 100 cultured calli, which represents a *49 increase with respect to previous reports. We also evaluated by flow cytometry and SSR molecular markers the effect of these in vitro culture conditions in the rate of DH plant production, finding that*70 % of the regenerated plants were true DHs. These results substantially improve the efficiencies of DH recovery published to date in eggplant, and may be useful to those working in the field of eggplant doubled haploidy and breeding. We acknowledge Dr. Rosa Peiro for her statistical advice, and the staff of the COMAV greenhouses for their valuable help.
