    Winter Ecosystem Respiration and Sources of CO2 From the High Arctic Tundra of Svalbard: Response to a Deeper Snow Experiment

    Currently, there is a lack of understanding on how the magnitude and sources of carbon (C) emissions from High Arctic tundra are impacted by changing snow cover duration and depth during winter. Here we investigated this issue in a graminoid tundra snow fence experiment on shale-derived gelisols in Svalbard from the end of the growing season and throughout the winter. To characterize emissions, we measured ecosystem respiration (Reco) along with its radiocarbon (14C) content. We assessed the composition of soil organic matter (SOM) by measuring its bulk-C and nitrogen (N), 14C content, and n-alkane composition. Our findings reveal that greater snow depth increased soil temperatures and winter Reco (25 mg C m−2 d−1 under deeper snow compared to 13 mg C m−2 d−1 in ambient conditions). At the end of the growing season, Reco was dominated by plant respiration and microbial decomposition of C fixed within the past 60 years (Δ14C = 62 Â± 8‰). During winter, emissions were significantly older (Δ14C = âˆ’64 Â± 14‰), and likely sourced from microorganisms decomposing aged SOM formed during the Holocene mixed with biotic or abiotic mineralization of the carbonaceous, fossil parent material. Our findings imply that snow cover duration and depth is a key control on soil temperatures and thus the magnitude of Reco in winter. We also show that in shallow Arctic soils, mineralization of carbonaceous parent materials can contribute significant proportions of fossil C to Reco. Therefore, permafrost-C inventories informing C emission projections must carefully distinguish between more vulnerable SOM from recently fixed biomass and more recalcitrant ancient sedimentary C sources

    Acoustic suppression of the coffee-ring effect

    We study the influence of acoustic fields on the evaporative self-assembly of solute particles suspended inside sessile droplets of complex fluids. The self-assembly process often results in an undesirable ring-like heterogeneous residue, a phenomenon known as the coffee-ring effect. Here we show that this ring-like self-assembly can be controlled acoustically to form homogeneous disc-like or concentrated spot-like residues. The principle of our method lies in the formation of dynamic patterns of particles in acoustically excited droplets, which inhibits the evaporation-driven convective transport of particles towards the contact line. We elucidate the mechanisms of this pattern formation and also obtain conditions for the suppression of the coffee-ring effect. Our results provide a more general solution to suppress the coffee-ring effect without any physiochemical modification of the fluids, the particles or the surface, thus potentially useful in a broad range of industrial and analytical applications that require homogenous solute depositions

    CONSERVING SURFACE-NESTING SEABIRDS AT THE PRINCE EDWARD ISLANDS: THE ROLES OF RESEARCH, MONITORING AND LEGISLATION

    South Africa's subantarctic Prince Edward Islands support substantial proportions of the global populations of a number of surface-nesting seabirds. Populations of most of these have decreased at the islands since the 1980s and 12 of 16 species are regarded as Threatened or Near Threatened regionally or internationally. The main causes of population decreases are thought to be by-catch mortality of albatrosses and giant petrels in longline fisheries, and environmental change influencing availability of prey to penguins and the Crozet shag Phalacrocorax [atriceps] melanogenis. It is proposed that the Prince Edward Islands Special Nature Reserve be expanded to include surrounding territorial waters so as to afford additional protection for seabirds breeding there, especially those species feeding near to the islands. Consideration needs also to be given to listing species as threatened or protected in terms of planned new legislation in South Africa and then developing management plans for them, preferably linked closely with the Agreement on the Conservation of Albatrosses and Petrels and the National Plan of Action (NPOA) – Seabirds. The islands should also be nominated as a Ramsar Wetland of International Importance in recognition of their importance to seabirds, with 13 of the 16 species exceeding the 1&#37 of the global population criterion. A combination of research, monitoring and legislation will help conserve the surface-nesting seabirds of the Prince Edward Islands into the 21st century, but only providing the effects of climate change can somehow be addressed.Afr.     Pramipexole reduces phosphorylation of α-Synuclein at Serine-129

    α-Synuclein is a central component of the pathogenesis of Parkinson's disease (PD). Phosphorylation at serine-129 represents an important post-translational modification and constitutes the major form of the protein in Lewy bodies. Several kinases have been implicated in the phosphorylation of α-synuclein. The targeting of kinase pathways as a potential to influence the pathogenesis of PD is an important focus of attention, given that mutations of specific kinases (LRRK2 and PINK1) are causes of familial PD. Pramipexole (PPX) is a dopamine agonist developed for the symptomatic relief of PD. Several in vitro and in vivo laboratory studies have demonstrated that PPX exerts neuroprotective properties in model systems of relevance to PD. The present study demonstrates that PPX inhibits the phosphorylation of α-synuclein and that this is independent of dopamine receptor activation. PPX blocks the increase in phosphorylated α-synuclein induced by inhibition of the ubiquitin proteasomal system. The phosphorylation of α-synuclein occurs in part at least through casein kinase 2, and PPX in turn reduces the phosphorylation of this enzyme, thereby inhibiting its activity. Thus, PPX decreases the phosphorylation of α-synuclein, and this mechanism may contribute to its protective properties in PD models

    Enhancement of Gap Junction Function During Acute Myocardial Infarction Modifies Healing and Reduces Late Ventricular Arrhythmia Susceptibility

    Objectives: To investigate the effects of enhancing gap junction (GJ) coupling during acute myocardial infarction (MI) on the healed infarct scar morphology and late post-MI arrhythmia susceptibility. Background: Increased heterogeneity of myocardial scarring after MI is associated with greater arrhythmia susceptibility. We hypothesized that short-term enhancement of GJ coupling during acute MI can produce more homogeneous infarct scars, reducing late susceptibility to post-MI arrhythmias. Methods: Following arrhythmic characterisation of the rat 4-week post-MI model (n=24), a further 27 Sprague-Dawley rats were randomised to receive rotigaptide to enhance GJ coupling (n=13) or saline control (n=14) by osmotic minipump immediately prior to, and for the first 7 days following surgical MI. At 4 weeks post-MI, hearts were explanted for ex vivo programmed electrical stimulation (PES) and optical mapping. Heterogeneity of infarct border zone (IBZ) scarring was quantified by histomorphometry. Results: Despite no detectable difference in infarct size at 4 weeks post-MI, rotigaptide-treated hearts had reduced arrhythmia susceptibility during PES (Inducibility score: rotigaptide 2.40.8, control 5.00.6, p=0.02) and less heterogeneous IBZ scarring (standard deviation of IBZ Complexity Score: rotigaptide 1.10.1, control 1.40.1, p=0.04), associated with an improvement in IBZ conduction velocity (rotigaptide 43.13.4 cm/s, control 34.82.0 cm/s, p=0.04). Conclusions: Enhancement of GJ coupling for only 7 days at the time of acute MI produced more homogeneous IBZ scarring and reduced arrhythmia susceptibility at 4 weeks post-MI. Short-term GJ modulation at the time of MI may represent a novel treatment strategy to modify the healed infarct scar morphology and reduce late post-MI arrhythmic risk

    Identification of systems containing nonlinear stiffnesses using backbone curves

The final version is available from Elsevier via the DOI in this record.This paper presents a method for the dynamic identification of structures containing discrete nonlinear stiffnesses. The approach requires the structure to be excited at a single resonant frequency, enabling measurements to be made in regimes of large displacements where nonlinearities are more likely to be significant. Measured resonant decay data is used to estimate the system backbone curves. Linear natural frequencies and nonlinear parameters are identified using these backbone curves assuming a form for the nonlinear behaviour. Numerical and experimental examples, inspired by an aerospace industry test case study, are considered to illustrate how the method can be applied.     Necrotizing meningoencephalitis in atypical dog breeds: a case series and literature review.

    BackgroundCanine necrotizing meningoencephalitis (NME) is a fatal, noninfectious inflammatory disease of unknown etiology. NME has been reported only in a small number of dog breeds, which has led to the presumption that it is a breed-restricted disorder.Hypothesis/objectivesOur objective was to describe histopathologically confirmed NME in dog breeds in which the condition has not been reported previously and to provide preliminary evidence that NME affects a wider spectrum of dog breeds than previously reported.AnimalsFour dogs with NME.MethodsArchives from 3 institutions and from 1 author's (BS) collection were reviewed to identify histopathologically confirmed cases of NME in breeds in which the disease has not been reported previously. Age, sex, breed, survival from onset of clinical signs, and histopathologic findings were evaluated.ResultsNecrotizing meningoencephalitis was identified in 4 small dog breeds (Papillon, Shih Tzu, Coton de Tulear, and Brussels Griffon). Median age at clinical evaluation was 2.5 years. Histopathologic abnormalities included 2 or more of the following: lymphoplasmacytic or histiocytic meningoencephalitis or encephalitis, moderate-to-severe cerebrocortical necrosis, variable involvement of other anatomic locations within the brain (cerebellum, brainstem), and absence of detectable infectious agents.Conclusions and clinical importanceUntil now, NME has only been described in 5 small dog breeds. We document an additional 4 small breeds previously not shown to develop NME. Our cases further illustrate that NME is not a breed-restricted disorder and should be considered in the differential diagnosis for dogs with signalment and clinical signs consistent with inflammatory brain disease

    Highly Efficient Spatially Offset Raman Spectroscopy to Profile Molecular Composition in Bone

    Spatially offset Raman spectroscopy (SORS) offers the prospect of collecting spectral information detailing the molecular composition of biomaterials at greater depths below the surface layers than are normally probed by conventional Raman spectroscopy. By collecting off-axial scattered light, the technique overcomes the large background from in-line light within scattering media. In this paper we present a configuration which enables the highly efficient collection of spectral markers, indicative of bone health, including Raman signatures to assess phosphate, collagen and carbonate content, at millimeter depths. We demonstrate the effectiveness of the technique by performing spectral decompositions to analyze the molecular distribution of these markers non-invasively, using in vitro model systems, comprising bone and tissue, in situ

Using prices to improve the efficiency with which water resources are allocated is now widely accepted in principle if somewhat difficult to achieve in practice. Whilst there are some technical difficulties associated with full-cost recovery in irrigation, the lack of political will to tackle reform remains a significant impediment. This article reports the results of an empirical investigation into farmers’ preferences for changes to water prices and tariff structures. We conclude that some of the preferences of farmers are conducive to price reform. We also find evidence that public subsidy of infrastructure in irrigation is not always aligned with the preferences of farmers
