    Sviluppo di un software per l’analisi di immagini di Diffusion Kurtosis Imaging

    L’analisi mediante RM del tensore di diffusione (Diffusion Tensor Imaging, DTI) consente di valutare anche in vivo e con modalità non invasive il processo di diffusione delle molecole d’acqua nei tessuti biologici. La peculiare organizzazione di alcuni tessuti biologici (es: muscoli, sostanza bianca del sistema nervoso centrale e tessuti ad alta cellularità) influenza tale fenomeno rendendolo anisotropo e quindi ben valutabile con tali tecniche di studio. Nonostante i grandi vantaggi di tale tecnica, il DTI è basato su un modello molto semplificato che assume che lo spostamento per diffusione segua un profilo gaussiano il che è molto raro in un ambiente variegato come i tessuti biologici. Per caratterizzare la natura non gaussiana della diffusione dell’acqua nei tessuti è stata sviluppata negli ultimi anni la Diffusion Kurtosis Imaging (DKI) che permette di ottenere ulteriori e più accurate informazioni sulle caratteristiche ultrastrutturali tissutali. Nel presente lavoro si è posto come obiettivo lo sviluppo di un software in grado di ricostruire le mappe tipiche della DKI. In particolare, il software è stato sviluppato in linguaggio di programmazione “Python” e permette di estrarre i parametri DTI e DKI da una serie di dati acquisiti per vari valori di b e per un vario numero di direzioni di gradienti

    After more than ten years of operation of the EPIC camera on board the X-ray observatory XMM-Newton we have reviewed the status of its thin and medium filters by performing both analysis of data collected in-flight and laboratory measurements on on-ground back-up filters. We have investigated the status of the EPIC thin and medium filters by performing an analysis of the optical loading in the PN offset maps to gauge variations in the optical and UV transmission of the filters. We both investigated repeated observations of single optically bright targets and performed a statistical analysis of the extent of loading versus visual magnitude at different epochs. We report the results of these measurements

    Blood circulating miR-28-5p and let-7d-5p associate with premature ageing in Down syndrome

    Persons with Down syndrome (DS) undergo a premature ageing with early onset of age-related diseases. The main endpoint of this study was the identification of blood circulating microRNAs (c-miRs) signatures characterizing DS ageing process. A discovery phase based on array was performed in plasma samples obtained from 3 young (31 Â± 2 years-old) and 3 elderly DS persons (66 Â± 2 years-old). Then, a validation phase was carried out for relevant miRs by RT-qPCR in an enlarged cohort of 43 DS individuals (from 19 up to 68 years-old). A group of 30 non-trisomic subjects, as representative of physiological ageing, was compared. In particular miR-628-5p, miR-152-3p, miR-28-5p, and let-7d-5p showed a lower level in younger DS persons (age ≤ 50 years) respect to the age-matched controls. Among those, miR-28-5p and let-7d-5p were found significantly decreased in physiological ageing ( oldest group ), thus they emerged as possible biomarkers of premature ageing in DS. Moreover, measuring blood levels of beta amyloid peptides, Aβ-42 was assessed at the lowest levels in physiological ageing and correlated with miR-28-5p and let-7d-5p in DS, while Aβ-40 correlated with miR-628-5p in the same cohort. New perspectives in terms of biomarkers are discussed

    Cytochalasin B Influences Cytoskeletal Organization and Osteogenic Potential of Human Wharton's Jelly Mesenchymal Stem Cells

    Among perinatal stem cells of the umbilical cord, human Wharton's jelly mesenchymal stem cells (hWJ-MSCs) are of great interest for cell-based therapy approaches in regenerative medicine, showing some advantages over other MSCs. In fact, hWJ-MSCs, placed between embryonic and adult MSCs, are not tumorigenic and are harvested with few ethical concerns. Furthermore, these cells can be easily cultured in vitro, maintaining both stem properties and a high proliferative rate for several passages, as well as trilineage capacity of differentiation. Recently, it has been demonstrated that cytoskeletal organization influences stem cell biology. Among molecules able to modulate its dynamics, Cytochalasin B (CB), a cyto-permeable mycotoxin, influences actin microfilament polymerization, thus affecting several cell properties, such as the ability of MSCs to differentiate towards a specific commitment. Here, we investigated for the first time the effects of a 24 h-treatment with CB at different concentrations (0.1-3 mu M) on hWJ-MSCs. CB influenced the cytoskeletal organization in a dose-dependent manner, inducing changes in cell number, proliferation, shape, and nanomechanical properties, thus promoting the osteogenic commitment of hWJ-MSCs, as confirmed by the expression analysis of osteogenic/autophagy markers

    The thin and medium filters of the EPIC camera on-board XMM-Newton: measured performance after more than 15 years of operation

    After more than 15 years of operation of the EPIC camera on board the XMM-Newton X-ray observatory, we have reviewed the status of its Thin and Medium filters. We have selected a set of Thin and Medium back-up filters among those still available in the EPIC consortium and have started a program to investigate their status by different laboratory measurements including: UV/VIS transmission, Raman scattering, X-Ray Photoelectron Spectroscopy, and Atomic Force Microscopy. Furthermore, we have investigated the status of the EPIC flight filters by performing an analysis of the optical loading in the PN offset maps to gauge variations in the optical and UV transmission. We both investigated repeated observations of single optically bright targets and performed a statistical analysis of the extent of loading versus visual magnitude at different epochs. We report the results of the measurements conducted up to now. Most notably, we find no evidence for change in the UV/VIS transmission of the back-up filters in ground tests spanning a 2 year period and we find no evidence for change in the optical transmission of the thin filter of the EPIC-pn camera from 2002 to 2012. We point out some lessons learned for the development and calibration programs of filters for X-ray detectors in future Astronomy missions
