    Geologic Provinces Beneath the Greenland Ice Sheet Constrained by Geophysical Data Synthesis

    Present understanding of Greenland's subglacial geology is derived mostly from interpolation of geologic mapping of its ice‐free margins and unconstrained by geophysical data. Here we refine the extent of its geologic provinces by synthesizing geophysical constraints on subglacial geology from seismic, gravity, magnetic and topographic data. North of 72°N, no province clearly extends across the whole island, leaving three distinct subglacial regions yet to be reconciled with margin geology. Geophysically coherent anomalies and apparent province boundaries are adjacent to the onset of faster ice flow at both Petermann Glacier and the Northeast Greenland Ice Stream. Separately, based on their subaerial expression, dozens of unusually long, straight and sub‐parallel subglacial valleys cross Greenland's interior and are not yet resolved by current syntheses of its subglacial topography

    A prospective evaluation of thiamine and magnesium status in relation to clinicopathological characteristics and 1-year mortality in patients with Alcohol Withdrawal Syndrome

    Background: Alcohol withdrawal syndrome (AWS) is routinely treated with B-vitamins. However, the relationship between thiamine status and outcome is rarely examined. The aim of the present study was to examine the relationship between thiamine and magnesium status in patients with AWS. Methods: Patients (n = 127) presenting to the Emergency Department with AWS were recruited to a prospective observational study. Blood samples were drawn to measure whole blood thiamine diphosphate (TDP) and serum magnesium concentrations. Routine biochemistry and haematology assays were also conducted. The Glasgow Modified Alcohol Withdrawal Score (GMAWS) measured severity of AWS. Seizure history and current medications were also recorded. Results: The majority of patients (99%) had whole blood TDP concentration within/above the reference interval (275–675 ng/gHb) and had been prescribed thiamine (70%). In contrast, the majority of patients (60%) had low serum magnesium concentrations (< 0.75 mmol/L) and had not been prescribed magnesium (93%). The majority of patients (66%) had plasma lactate concentrations above 2.0 mmol/L. At 1 year, 13 patients with AWS had died giving a mortality rate of 11%. Male gender (p < 0.05), BMI < 20 kg/m2 (p < 0.01), GMAWS max ≄ 4 (p < 0.05), elevated plasma lactate (p < 0.01), low albumin (p < 0.05) and elevated serum CRP (p < 0.05) were associated with greater 1-year mortality. Also, low serum magnesium at time of recruitment to study and low serum magnesium at next admission were associated with higher 1-year mortality rates, (84% and 100% respectively; both p < 0.05). Conclusion: The prevalence of low circulating thiamine concentrations were rare and it was regularly prescribed in patients with AWS. In contrast, low serum magnesium concentrations were common and not prescribed. Low serum magnesium was associated more severe AWS and increased 1-year mortality

    Inhibition of Hippocampal Synaptic Activity by ATP, Hypoxia or Oxygen-Glucose Deprivation Does Not Require CD73

    Adenosine, through activation of its A1 receptors, has neuroprotective effects during hypoxia and ischemia. Recently, using transgenic mice with neuronal expression of human equilibrative nucleoside transporter 1 (hENT1), we reported that nucleoside transporter-mediated release of adenosine from neurons was not a key mechanism facilitating the actions of adenosine at A1 receptors during hypoxia/ischemia. The present study was performed to test the importance of CD73 (ecto-5â€Č-nucleotidase) for basal and hypoxic/ischemic adenosine production. Hippocampal slice electrophysiology was performed with CD73+/+ and CD73−/− mice. Adenosine and ATP had similar inhibitory effects in both genotypes, with IC50 values of approximately 25 ”M. In contrast, ATP was a less potent inhibitor (IC50 = 100 ”M) in slices from mice expressing hENT1 in neurons. The inhibitory effects of ATP in CD73+/+ and CD73−/− slices were blocked by the adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine (DPCPX) and were enhanced by the nucleoside transport inhibitor S-(4-nitrobenzyl)-6-thioinosine (NBTI), consistent with effects that are mediated by adenosine after metabolism of ATP. AMP showed a similar inhibitory effect to ATP and adenosine, indicating that the response to ATP was not mediated by P2 receptors. In comparing CD73−/− and CD73+/+ slices, hypoxia and oxygen-glucose deprivation produced similar depression of synaptic transmission in both genotypes. An inhibitor of tissue non-specific alkaline phosphatase (TNAP) was found to attenuate the inhibitory effects of AMP and ATP, increase basal synaptic activity and reduce responses to oxygen-glucose deprivation selectively in slices from CD73−/− mice. These results do not support an important role for CD73 in the formation of adenosine in the CA1 area of the hippocampus during basal, hypoxic or ischemic conditions, but instead point to TNAP as a potential source of extracellular adenosine when CD73 is absent

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Voice and visibility: tackling the ‘invisibility’ of the sexual orientation strand in UK organisation equality and diversity research

    This covering statement introduces the nine published outputs in this submission, and explains their genesis. It considers the implications of the growing visibility of the sexual orientation strand for individuals and organisations operating in the UK context before and after the introduction of the Employment Equality (Sexual Orientation) Regulations 2003 (hereafter the Regulations). The PhD advanced knowledge and understanding of the following: the rationale for and drivers of sexual orientation equality work in UK trade union, public, private and voluntary sector organisations over the last two decades; the equality/diversity structures, policies and practices introduced by organisations in order to become more inclusive of lesbian gay and bisexual (LGB) people; LGB people’s perceptions and experiences of these sexual orientation equality and diversity policies and practices; LGB people’s voice, activism and agency in influencing the changes which have been taking place. The covering document provides details of the research projects and the interpretivist case study methodology on which the published outputs draw. It summarises and links the aims and principal findings of each output demonstrating that they form a coherent body of work. It concludes that although the introduction of the Regulations has been a positive trigger to sexual orientation equality work in the UK, progress remains uneven within and across organisations. Thus, it identifies voice mechanisms such as LGBT trade union and company network groups as key tools for inclusion. The conclusion locates the PhD as an original contribution to the advancement of sexual orientation organisation equality/diversity research. It does so by discussing its empirical and theoretical contributions to a sexual orientation research agenda which has been developing in waves subject to social, political and legal change and mobilisation in LGBT communities in different parts of the world

    Negotiating the self between past and present: narratives of older women moving towards self-employment

    Older people are encouraged into self-employment as a means to extend their working lives; however both age and gender are thought to constrain the capacity of individuals to take on an enterprising identity. This paper explores the narrative identity work of women over 50 contemplating a move into self-employment. It reveals how they negotiated a provisional self-employed identity in relation to an aged identity, an enterprising identity and an identity as organizational outsider. It discusses the implications of contrasting forms of engagement with these identities for the subsequent enactment of participants’ business plans. The paper briefly considers the implications of its findings for the following areas: enacting and realising provisional identities; the relationship of self-employment to enterprise; and a process-based conceptualisation of age
