Cellulase production by the Thermophilic Fungus, Thermoascus aurantiacus
Thermoascus aurantiacus was the most active cellulase producer of several thermphilic fungi tested.
The optimum growth temperature for T. aurantiacus in liquid medium was 45° C and maximum cellulase
production from filter paper occurred at 40°C. The optimum temperatue for {3-glucosidase and carboxymethylcellulase
activity was 70° C; for filter paper degrading activity it was 65°C. Maximum activity was
found at pH 5.0 for the filter paper degrading enzyme and glucosidase and pH 4.3 for carboxymethylcellulase
activity
The Isolation of a New S-Methyl Benzothioate Compound from a Marine-Derived Streptomyces sp.
The application of an HPLC bioactivity profiling/microtiter plate technique in conjunction with microprobe NMR instrumentation and access to the AntiMarin database has led to the isolation of a new 1. In this example, 1 was isolated from a cytotoxic fraction of an extract obtained from marine-derived Streptomyces sp. cultured on Starch Casein Agar (SCA) medium. The 1D and 2D 1H NMR and ESIMS data obtained from 20 μg of compound 1 fully defined the structure. The known 2 was also isolated and readily dereplicated using this approach
The Velocity Function of Galaxies
We present a galaxy circular velocity function, Psi(log v), derived from
existing luminosity functions and luminosity-velocity relations. Such a
velocity function is desirable for several reasons. First, it enables an
objective comparison of luminosity functions obtained in different bands and
for different galaxy morphologies, with a statistical correction for dust
extinction. In addition, the velocity function simplifies comparison of
observations with predictions from high-resolution cosmological N-body
simulations.
We derive velocity functions from five different data sets and find rough
agreement among them, but about a factor of 2 variation in amplitude. These
velocity functions are then compared with N-body simulations of a LCDM model
(corrected for baryonic infall) in order to demonstrate both the utility and
current limitations of this approach. The number density of dark matter halos
and the slope of the velocity function near v_*, the circular velocity
corresponding to an ~L_* spiral galaxy, are found to be comparable to that of
observed galaxies. The primary sources of uncertainty in construction of
Psi(log v) from observations and N-body simulations are discussed and

Experimental and Theoretical Results for Weak Charge Current Backward Proton Production
In this paper, we do three things in the study of deuteron break-up by high
energy neutrino beams. (1) We present previously unpublished data on neutrino
induced backward protons from deuteron targets; (2) we calculate the
contributions from both the two-nucleon (2N) and six-quark (6q) deuteron
components, which depend upon the overall normalization of the part that is 6q;
and (3) we suggest other signatures for distinguishing the 2N and 6q clusters.
We conclude that the 6q cluster easily explains the shape of the high momentum
backward proton spectrum, and its size is nicely explained if the amount of 6q
is one or a few percent by normalization of the deuteron. There is a crossover,
above which the 6q contribution is important or dominant, at 300--400 MeV/c
backward proton momentum.Comment: 8 pages, 5 figure
Genetic variants and physical activity interact to affect bone density in Hispanic children
Background: Our aim was to investigate if moderate to vigorous physical activity (MVPA), calcium intake interacts with bone mineral density (BMD)-related single nucleotide polymorphisms (SNPs) to influence BMD in 750 Hispanic children (4-19y) of the cross-sectional Viva La Familia Study.
Methods: Physical activity and dietary intake were measured by accelerometers and multiple-pass 24 h dietary recalls, respectively. Total body and lumbar spine BMD were measured by dual energy X-ray absorptiometry. A polygenic risk score (PRS) was computed based on SNPs identified in published literature. Regression analysis was conducted with PRSs, MVPA and calcium intake with total body and lumbar spine BMD.
Results: We found evidence of statistically significant interaction effects between the PRS and MVPA on total body BMD and lumbar spine BMD (p \u3c 0.05). Higher PRS was associated with a lower total body BMD (β = − 0.040 ± 0.009, p = 1.1 × 10− 5 ) and lumbar spine BMD (β = − 0.042 ± 0.013, p = 0.0016) in low MVPA group, as compared to high MVPA group (β = − 0.015 ± 0.006, p = 0.02; β = 0.008 ± 0.01, p = 0.4, respectively).
Discussion: The study indicated that calcium intake does not modify the relationship between genetic variants and BMD, while it implied physical activity interacts with genetic variants to affect BMD in Hispanic children. Due to limited sample size of our study, future research on gene by environment interaction on bone health and functional studies to provide biological insights are needed
Robotic milking technologies and renegotiating situated ethical relationships on UK dairy farms
Robotic or automatic milking systems (AMS) are novel technologies that take over the labor of dairy farming and reduce the need for human-animal interactions. Because robotic milking involves the replacement of 'conventional' twice-a-day milking managed by people with a system that supposedly allows cows the freedom to be milked automatically whenever they choose, some claim robotic milking has health and welfare benefits for cows, increases productivity, and has lifestyle advantages for dairy farmers. This paper examines how established ethical relations on dairy farms are unsettled by the intervention of a radically different technology such as AMS. The renegotiation of ethical relationships is thus an important dimension of how the actors involved are re-assembled around a new technology. The paper draws on in-depth research on UK dairy farms comparing those using conventional milking technologies with those using AMS. We explore the situated ethical relations that are negotiated in practice, focusing on the contingent and complex nature of human-animal-technology interactions. We show that ethical relations are situated and emergent, and that as the identities, roles, and subjectivities of humans and animals are unsettled through the intervention of a new technology, the ethical relations also shift. © 2013 Springer Science+Business Media Dordrecht
In search of the authentic nation: landscape and national identity in Canada and Switzerland
While the study of nationalism and national identity has flourished in the last decade, little attention has been devoted to the conditions under which natural environments acquire significance in definitions of nationhood. This article examines the identity-forming role of landscape depictions in two polyethnic nation-states: Canada and Switzerland. Two types of geographical national identity are identified. The first – what we call the ‘nationalisation of nature’– portrays zarticular landscapes as expressions of national authenticity. The second pattern – what we refer to as the ‘naturalisation of the nation’– rests upon a notion of geographical determinism that depicts specific landscapes as forces capable of determining national identity. The authors offer two reasons why the second pattern came to prevail in the cases under consideration: (1) the affinity between wild landscape and the Romantic ideal of pure, rugged nature, and (2) a divergence between the nationalist ideal of ethnic homogeneity and the polyethnic composition of the two societies under consideration
Using the RDP Classifier to Predict Taxonomic Novelty and Reduce the Search Space for Finding Novel Organisms
BACKGROUND: Currently, the naïve Bayesian classifier provided by the Ribosomal Database Project (RDP) is one of the most widely used tools to classify 16S rRNA sequences, mainly collected from environmental samples. We show that RDP has 97+% assignment accuracy and is fast for 250 bp and longer reads when the read originates from a taxon known to the database. Because most environmental samples will contain organisms from taxa whose 16S rRNA genes have not been previously sequenced, we aim to benchmark how well the RDP classifier and other competing methods can discriminate these novel taxa from known taxa. PRINCIPAL FINDINGS: Because each fragment is assigned a score (containing likelihood or confidence information such as the boostrap score in the RDP classifier), we "train" a threshold to discriminate between novel and known organisms and observe its performance on a test set. The threshold that we determine tends to be conservative (low sensitivity but high specificity) for naïve Bayesian methods. Nonetheless, our method performs better with the RDP classifier than the other methods tested, measured by the f-measure and the area-under-the-curve on the receiver operating characteristic of the test set. By constraining the database to well-represented genera, sensitivity improves 3-15%. Finally, we show that the detector is a good predictor to determine novel abundant taxa (especially for finer levels of taxonomy where novelty is more likely to be present). CONCLUSIONS: We conclude that selecting a read-length appropriate RDP bootstrap score can significantly reduce the search space for identifying novel genera and higher levels in taxonomy. In addition, having a well-represented database significantly improves performance while having genera that are "highly" similar does not make a significant improvement. On a real dataset from an Amazon Terra Preta soil sample, we show that the detector can predict (or correlates to) whether novel sequences will be assigned to new taxa when the RDP database "doubles" in the future
Narrative constructions of anorexia and abuse: An athlete's search for meaning in trauma
Interpretive approaches to the study of eating disorders are scarce. Narrative analysis provides an attractive means to address this shortfall and is applied to the life story of Beth, a former elite athlete with experience of anorexia nervosa and, as she revealed, sexual abuse. Six unstructured life history interviews took place yielding more than 9 hours of interview data. Throughout our conversations, Beth constructed multiple, fragile, and sometimes contrasting narrative coherences indicative of a fragmented and uncertain understanding of her life. It is argued that how Beth makes sense of her trauma is consequential for her future experiences
