Optimization of a viability PCR method for the detection of Listeria monocytogenes in food samples

Inventario de las emisiones de contaminantes atmosféricos por tráfico rodado en España en el año 2009
Este PFC tiene como finalidad la elaboración de un inventario de emisiones de
contaminantes atmosféricos debidos al tráfico rodado en España, para el año 2009. En éste,
se estiman las emisiones producidas por el parque vehicular español (turismos, camiones,
furgonetas, motocicletas y ciclomotores) con una resolución espacial de 1 km2 y una
resolución horaria de 1 h.
Para la modelización de las emisiones del tráfico rodado se ha conseguido información de la
intensidad media diaria de circulación (IMD) de más de 24000 puntos, cubriendo asà más de
111000 km de carreteras. En especial, se destaca el gran número de aforos de las ciudades
de Madrid (3198) y Barcelona (2575), las cuales soportan una parte muy importante del
tráfico presente en todo el estado. Además se han conseguido más de 27600 perfiles
(horarios, semanales y mensuales) de distribución temporal de la IMD, que permiten mostrar
cual es la evolución del tráfico al largo de las horas, dÃas y meses; en especial las
diferencias entre dÃas laborables y festivos.
La metodologÃa usada para la estimación de emisiones es la planteada por EMEP/EEA
(2009), en la que se distinguen cuatro tipos de emisiones: en caliente, en frÃo, evaporativas
y de desgaste (neumáticos, frenos y abrasión de pavimento). Además se destaca la
inclusión de las emisiones por resuspensión. Esta metodologÃa describe factores de emisión
para 246 categorÃas de vehÃculos. Los contaminantes considerados en el presente
inventario son los principales primarios: NOx (óxidos de nitrógeno), CO (monóxido de
carbono), NMVOC (compuestos orgánicos volátiles distintos del metano), SOx (dióxido de
sodio), PM (material particulado) y amonÃaco (NH3). Nótese que en el caso de NMVOC y PM
se nombran siguiendo su expresión en inglés para más comodidad con la literatura
consultada.
Mediante el soporte informático del programa ArcGIS Desktop 10 y la cartografÃa digital
proporcionada por Tele Atlas Multinet, se ha podido obtener la visualización de los
resultados espacialmente y siguiendo una distribución horaria. De esta forma se ha podido
identificar las principales focos de emisión asà como su evolución a lo largo de un perÃodo.
Los resultados obtenidos muestran como existe una elevada emisión de PM y NOx
producidos, esencialmente por los vehÃculos diésel, y elevadas concentraciones de CO y
NMVOC producidas por vehÃculos gasolina. Tanto el SOx como el NH3 presentan cantidades
poco significativas.
Para la validación de los resultados, se hace una comparación con los resultados que
reporta el Inventario Nacional de Emisiones (INESP), elaborado por el Ministerio de Medio
Ambiente Rural y Marino (MARM)
Tumorous area detection in breast cancer biopsy images using multi-resolution networks
En aquest projecte, volem fer detecció de zona tumoral de mostres de teixits mamaris tenyides amb Hematoxilina-Eosina. En particular, volem detectar tumors in situ i invasius per separat. Com els patòlegs utilitzen múltiples resolucions en la realització d'aquesta tasca, hem entrenat una xarxa de multiresolució per segmentació semà ntica anomenada HookNet. L'arquitectura d'aquesta xarxa permet al model de segmentació prendre un parell d'imatges concèntrices amb diferents resolucions, d'aquesta manera proporcionem a la xarxa informació contextual, aixà com detalls d'alta resolució de les cèl·lules. Les prediccions de HookNet van obtenir un F-score mitjà de 0,71, superant una U-Net entrenada amb imatges a un nivell de zoom de 10x, que va obtenir un F-score de 0,49. No obstant això, una U-Net entrenada amb imatges amb una magnificació de 2,5x millora els resultats de HookNet i obté un F-score de 0,81.In this project we want to perform tumorous area segmentation of breast tissue samples stained with Hematoxylin-Eosin. In particular, we want to detect in situ and invasive tumors separately. Since pathologists use multiple resolutions when performing this task, we have chosen and trained a multiresolution network for semantic segmentation called HookNet. The architecture of this network allows the segmentation model to take a pair of concentric images with different resolutions, this way we provide the network with contextual information as well as high resolution details of the cells. The predictions of HookNet obtained an average F-score of 0.71, outperforming a U-Net trained on image patches at 10x magnification level which obtained an F-score of 0.49. However, a single-resolution U-Net trained with imaged at 2.5x magnification level still beat HookNet with an F-score of 0.81

On the construction and algebraic semantics of relevance logic

Visible light enhances the antimicrobial effect of some essential oils
The photodisinfection is a topical, broad spectrum antimicrobial technology, targeting bacteria, virus, fungi, and protozoa effective for single cells as for biofilms. Natural molecules have been studied less than synthetic agents in the process but they are currently receiving great interest. Therefore, the aim of this study is to evaluate for the first time if non-coherent blue and red light enhances the antimicrobial activity of some essential oils when standard strains for antibiotic or fungicide tests are enlightened in vitro. Staphylococcus epidermidis, Pseudomonas aeruginosa and Candida albicans collection strains were irradiated with monochromatic visible light from light emitting diodes in the presence of 5% and 0.5% eucalyptus (Eucalyptus globulus), clove (Eugenia caryophyllata), and thyme (Thymus vulgaris) essential oils. Microbial levels were measured by plate count on culture media. In this preliminary report, the results differ according to the kind and concentration of antimicrobial oils, the wavelength of light, and the prokaryotic or eukaryotic microorganism. The results support the idea that mainly blue light enhances the innate antimicrobial activity of the essential oils, especially phenols, and could offer a very efficient and natural way to combat microorganisms in several industries and medical applications.Postprint (author's final draft

GeNIUL, experts on viability PCR
The standard PCR technique allows the rapid detection of microorganisms, difficult to detect by traditional culture methods. However, the control of samples through PCR has the drawback of false positives due to amplification of DNA/RNA of non-viable cells. This can be overcome by adding intercalating agents (EMA or PMA) to the sample, which selectively penetrates nonviable cells and blocks their DNA/RNA after a photo-activation process, preventing its subsequent amplification by PCR. This process is known as viability PCR (v-PCR). v-PCR has been used as an accurate, reliable and simple method to detect and quantify viable microorganism involved in environmental samples, food production process and clinical samples.
