    An investigation of UK passenger attitudes towards the carbon offsetting of both flight and airport emissions

    Airlines and airports have to meet strict carbon emission reduction targets by 2050. Technological improvements and operational efficiencies can only go so far, in an industry that relies on hydrocarbons. To help the industry meet its targets, carbon offset schemes are a viable tool. These schemes are voluntary to passengers and so to become successful passengers in large numbers need to engage with them. At present less than 9% of passengers have ever donated to a carbon offset scheme. The industry has failed to promote such schemes with 60% of passengers unaware that they exist. Despite this, 82% would offset in the future, women and under 40’s in particular. Factors such as gender and a passenger’s belief in the existence of climate change play a key role in affecting a passenger’s likelihood of donating to a scheme. The airport offset scheme is a viable one, however there is mixed reaction from passengers

    Gendered experiences of academic staff in relation to research activity and the REF2014

    This report is based on research commissioned by the institutional Research and Knowledge Transfer Office between June 2015 and June 2016. This research has focused on generating qualitative and quantitative data as to the potential reasons why there appears to be a gender disparity in research productivity within the commissioning institution. In particular, the number of women self-selecting for representation in the REF2014 was comparatively low. This research was led by Dr Chantal Davies (as part of her broader remit in relation to the Forum for Research into Equality and Diversity) with Dr Ruth Healey as co-researcher and Anthony Cliffe as research assistant.     Worm Burden-Dependent Disruption of the Porcine Colon Microbiota by Trichuris suis Infection

    Helminth infection in pigs serves as an excellent model for the study of the interaction between human malnutrition and parasitic infection and could have important implications in human health. We had observed that pigs infected with Trichuris suis for 21 days showed significant changes in the proximal colon microbiota. In this study, interactions between worm burden and severity of disruptions to the microbial composition and metabolic potentials in the porcine proximal colon microbiota were investigated using metagenomic tools. Pigs were infected by a single dose of T. suis eggs for 53 days. Among infected pigs, two cohorts were differentiated that either had adult worms or were worm-free. Infection resulted in a significant change in the abundance of approximately 13% of genera detected in the proximal colon microbiota regardless of worm status, suggesting a relatively persistent change over time in the microbiota due to the initial infection. A significant reduction in the abundance of Fibrobacter and Ruminococcus indicated a change in the fibrolytic capacity of the colon microbiota in T. suis infected pigs. In addition, ∼10% of identified KEGG pathways were affected by infection, including ABC transporters, peptidoglycan biosynthesis, and lipopolysaccharide biosynthesis as well as α-linolenic acid metabolism. Trichuris suis infection modulated host immunity to Campylobacter because there was a 3-fold increase in the relative abundance in the colon microbiota of infected pigs with worms compared to naïve controls, but a 3-fold reduction in worm-free infected pigs compared to controls. The level of pathology observed in infected pigs with worms compared to worm-free infected pigs may relate to the local host response because expression of several Th2-related genes were enhanced in infected pigs with worms versus those worm-free. Our findings provided insight into the dynamics of the proximal colon microbiota in pigs in response to T. suis infection

    Nippostrongylus-induced intestinal hypercontractility requires IL-4 receptor alpha-responsiveness by T cells in mice

    Gut-dwelling helminthes induce potent IL-4 and IL-13 dominated type 2 T helper cell (T H 2) immune responses, with IL-13 production being essential for Nippostrongylus brasiliensis expulsion. This T H 2 response results in intestinal inflammation associated with local infiltration by T cells and macrophages. The resulting increased IL-4/IL-13 intestinal milieu drives goblet cell hyperplasia, alternative macrophage activation and smooth muscle cell hypercontraction. In this study we investigated how IL-4-promoted T cells contributed to the parasite induced effects in the intestine. This was achieved using pan T cell-specific IL-4 receptor alpha-deficient mice (iLck cre IL-4Rα −/lox ) and IL-4Rα-responsive control mice. Global IL-4Rα −/− mice showed, as expected, impaired type 2 immunity to N. brasiliensis . Infected T cell-specific IL-4Rα-deficient mice showed comparable worm expulsion, goblet cell hyperplasia and IgE responses to control mice. However, impaired IL-4-promoted T H 2 cells in T cell-specific IL-4Rα deficient mice led to strikingly reduced IL-4 production by mesenteric lymph node CD4 + T cells and reduced intestinal IL-4 and IL-13 levels, compared to control mice. This reduced IL-4/IL-13 response was associated with an impaired IL-4/IL-13-mediated smooth muscle cell hypercontractility, similar to that seen in global IL-4Rα −/− mice. These results demonstrate that IL-4-promoted T cell responses are not required for the resolution of a primary N. brasiliensis infection. However, they do contribute significantly to an important physiological manifestation of helminth infection; namely intestinal smooth muscle cell-driven hypercontractility

    The absence of MyD88 has no effect on the induction of alternatively activated macrophage during Fasciola hepatica infection

    <p>Abstract</p> <p>Background</p> <p>Alternatively activated macrophages (AAMϕ) play important roles in allergies and responses to parasitic infections. However, whether signaling through toll-like receptors (TLRs) plays any role in AAMϕ induction when young <it>Fasciola hepatica </it>penetrates the liver capsule and migrates through the liver tissue is still unclear.</p> <p>Results</p> <p>The data show that the lack of myeloid differentiation factor 88 (MyD88) has no effect on the AAMϕ derived from the bone marrow (BMMϕ) <it>in vitro </it>and does not impair the mRNA expression of arginase-1, resistin-like molecule (RELMα), and Ym1 in BMMϕs. The Th2 cytokine production bias in splenocytes was not significantly altered in <it>F. hepatica</it>-infected mice in the absence of MyD88 <it>in vitro </it>and in the pleural cavity lavage <it>in vivo</it>. In addition, MyD88-deficiency has no effect on the arginase production of the <it>F. hepatica </it>elicited macrophages (Fe Mϕs), production of RELMα and Ym1 proteins and mRNA expression of Ym1 and RELMα of macrophages in the peritoneal cavity 6 weeks post <it>F. hepatica </it>infection.</p> <p>Conclusions</p> <p>The absence of MyD88 has no effect on presence of AAMϕ 6 weeks post <it>F. hepatica </it>infection.</p

    Scaling the mountain: an exploration of gendered experience of academic staff in relation to the Research Excellence Framework 2014

    This paper will explore the experiences of female academics within the research institution in relation to the recent Research Excellence Framework in 2014 (REF 2014). Pursuant to the Equality Act 2010, there is now a legal duty (the Public Sector Equality Duty or PSED) for public bodies and therefore Higher Education Institutions (HEI) to have ‘due regard’ to the need to eliminate unlawful discrimination; advance equality of opportunity; and foster good relations between people who share a protected characteristic and people who do not share it (Equality and Human Rights Commission, 2015). It is considered that the PSED thus requires individual institutions to consider and explore the experience of female academics with a view to generating qualitative data about the experience of this group in relation to the REF 2014. This paper will explore some of the early findings of an institutionally funded research project in relation to the gendered experience of academic staff in the research institution in relation to the REF 2014 process and provide the opportunity for local perspectives in this regard to feed into lessons learnt at a national and global level

    IL-4Ralpha-associated antigen processing by B cells promotes immunity in Nippostrongylus brasiliensis infection.

    In this study, B cell function in protective T(H)2 immunity against N. brasiliensis infection was investigated. Protection against secondary infection depended on IL-4Ralpha and IL-13; but not IL-4. Protection did not associate with parasite specific antibody responses. Re-infection of B cell-specific IL-4Ralpha(-)/(-) mice resulted in increased worm burdens compared to control mice, despite their equivalent capacity to control primary infection. Impaired protection correlated with reduced lymphocyte IL-13 production and B cell MHC class II and CD86 surface expression. Adoptive transfer of in vivo N. brasiliensis primed IL-4Ralpha expressing B cells into naive BALB/c mice, but not IL-4Ralpha or IL-13 deficient B cells, conferred protection against primary N. brasiliensis infection. This protection required MHC class II compatibility on B cells suggesting cognate interactions by B cells with CD4(+) T cells were important to co-ordinate immunity. Furthermore, the rapid nature of these protective effects by B cells suggested non-BCR mediated mechanisms, such as via Toll Like Receptors, was involved, and this was supported by transfer experiments using antigen pulsed Myd88(-)/(-) B cells. These data suggest TLR dependent antigen processing by IL-4Ralpha-responsive B cells producing IL-13 contribute significantly to CD4(+) T cell-mediated protective immunity against N. brasiliensis infection
