    Recall and recognition of dreams and waking events: A diary paradigm

    Summary: Dream recall is often considered to be poor, although it is rarely systematically compared to the retrieval of waking event memories. A diary paradigm was implemented to explore the memory profile of recalled dreams over time, in comparison to recalled waking experiences. Twenty-five participants completed a dream and waking-event diary over two weeks. Titles of reports were re-presented in a surprise recall task. Subsequently, extracts of the reports were represented for recognition. No significant differences were found between dream and waking event memories in terms of either recall or recognition although some differences were found in terms of recollective experience, with waking-events being "remembered"as episodic memories more than dreams. Diary dream memories that are recalled, reported and to some extent rehearsed are therefore accessible and detailed in a similar manner to waking experiences, providing that they are adequately encoded close to their time of occurrence.     Circulatory contributors to the phenotype in hereditary hemorrhagic telangiectasia

    Hereditary hemorrhagic telangiectasia (HHT) is mechanistically and therapeutically challenging, not only because of the molecular and cellular perturbations that generate vascular abnormalities, but also the modifications to circulatory physiology that result, and are likely to exacerbate vascular injury. First, most HHT patients have visceral arteriovenous malformations (AVMs). Significant visceral AVMs reduce the systemic vascular resistance: supra-normal cardiac outputs are required to maintain arterial blood pressure, and may result in significant pulmonary venous hypertension. Secondly, bleeding from nasal and gastrointestinal telangiectasia leads to iron losses of such magnitude that in most cases, diet is insufficient to meet the ‘hemorrhage adjusted iron requirement.’ Resultant iron deficiency restricts erythropoiesis, leading to anemia and further increases in cardiac output. Low iron levels are also associated with venous and arterial thromboses, elevated Factor VIII, and increased platelet aggregation to circulating 5HT (serotonin). Third, recent data highlight that reduced oxygenation of blood due to pulmonary AVMs results in a graded erythrocytotic response to maintain arterial oxygen content, and higher stroke volumes and/or heart rates to maintain oxygen delivery. Finally, HHT-independent factors such as diet, pregnancy, sepsis and other intercurrent illnesses also influence vascular structures, hemorrhage, and iron handling in HHT patients. These considerations emphasize the complexity of mechanisms that impact on vascular structures in HHT, and also offer opportunities for targeted therapeutic approaches

    Any medium can be represented as an isometric subgraph of the hypercube, with each token of the medium represented by a particular equivalence class of arcs of the subgraph. Such a representation, although useful, is not especially revealing of the structure of a particular medium. We propose an axiomatic definition of the concept of a `mediatic graph'. We prove that the graph of any medium is a mediatic graph. We also show that, for any non-necessarily finite set S, there exists a bijection from the collection M of all the media on a given set S (of states) onto the collection G of all the mediatic graphs on S.Comment: Four axioms replaced by two; two references added; Fig.6 correcte

    The dynamics of the one-tangle and the concurrence is analyzed in the Lipkin-Meshkov-Glick model which describes many physical systems such as the two-mode Bose-Einstein condensates. We consider two different initial states which are physically relevant and show that their entanglement dynamics are very different. A semiclassical analysis is used to compute the one-tangle which measures the entanglement of one spin with all the others, whereas the frozen-spin approximation allows us to compute the concurrence using its mapping onto the spin squeezing parameter.Comment: 11 pages, 11 EPS figures, published versio

    Primary cilia disassembly down-regulates mechanosensitive hedgehog signalling: a feedback mechanism controlling ADAMTS-5 expression in chondrocytes

    SummaryObjectiveHedgehog signalling is mediated by the primary cilium and promotes cartilage degeneration in osteoarthritis. Primary cilia are influenced by pathological stimuli and cilia length and prevalence are increased in osteoarthritic cartilage. This study aims to investigate the relationship between mechanical loading, hedgehog signalling and cilia disassembly in articular chondrocytes.MethodsPrimary bovine articular chondrocytes were subjected to cyclic tensile strain (CTS; 0.33 Hz, 10% or 20% strain). Hedgehog pathway activation (Ptch1, Gli1) and A Disintegrin And Metalloproteinase with Thrombospondin Motifs 5 (ADAMTS-5) expression were assessed by real-time PCR. A chondrocyte cell line generated from the Tg737ORPK mouse was used to investigate the role of the cilium in this response. Cilia length and prevalence were quantified by immunocytochemistry and confocal microscopy.ResultsMechanical strain upregulates Indian hedgehog expression and activates hedgehog signalling. Ptch1, Gli1 and ADAMTS-5 expression were increased following 10% CTS, but not 20% CTS. Pathway activation requires a functioning primary cilium and is not observed in Tg737ORPK cells lacking cilia. Mechanical loading significantly reduced cilium length such that cilia became progressively shorter with increasing strain magnitude. Inhibition of histone deacetylase 6 (HDAC6), a tubulin deacetylase, prevented cilia disassembly and restored mechanosensitive hedgehog signalling and ADAMTS-5 expression at 20% CTS.ConclusionsThis study demonstrates for the first time that mechanical loading activates primary cilia-mediated hedgehog signalling and ADAMTS-5 expression in adult articular chondrocytes, but that this response is lost at high strains due to HDAC6-mediated cilia disassembly. The study provides new mechanistic insight into the role of primary cilia and mechanical loading in articular cartilage
