    Facile synthesis of chitosan-capped ZnS quantum dots as an eco-friendly fluorescence sensor for rapid determination of bisphenol A in water and plastic samples

    This paper describes a novel eco-friendly fluorescence sensor for determination of bisphenol A (BPA) based on chitosan-capped ZnS quantum dots (QDs). By using safe and inexpensive materials, nontoxic ZnS QDs were synthesized via an environment-friendly method using chitosan as a capping agent. The as-prepared ZnS QDs exhibited characteristic absorption (absorbance edge at 310 nm) and emission (maxima at 430 nm) spectra with a relatively high fluorescence quantum yield of 11.8%. Quantitative detection of BPA was developed based on fluorescence quenching of chitosan-capped ZnS QDs with high sensitivity and selectivity. Under optimal conditions, the fluorescence response of ZnS QDs was linearly proportional to BPA concentration over a wide range from 0.50 to 300 mu g L-1 with a detection limit of 0.08 mu g L-1. Most of the potentially coexisting substances did not interfere with the BPA-induced quenching effect. The proposed analytical method for BPA was successfully applied to water and plastic real samples. The possible quenching mechanism is also discussed

    Molecular analysis and expression of phenylalanine ammonia-lyase from poinsettia (Euphorbia pulcherrima willd.)

    Phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) is a key regulatory enzyme that link primary and secondary metabolism in plants by catalyzing the conversion of l-phenylalanine to cinnamic acid. In this study, the cDNA and genomic DNA of PAL (named EpPAL) in poinsettia (Euphorbia pulcherrima willd.) were isolated and submitted in GenBank with accession number FJ594466 and FJ943503, respectively. The full-length of cDNA was 2429 bp with a poly (A) tail and contains a 2166-bp open reading frame (ORF) encoding 721 amino acids. The sequence of genomic DNA was 3315 bp, and the transcript was divided into two exons by an 886-bp long intron which located at 416 bp downstream initiation codon. Expression analysis of EpPAL in poinsettia revealed that expression levels were higher in roots and bracts, but lower in stems and green leaves. Meanwhile, expression levels increased in the order: green leaves - turning color leaves - bracts, which were consistent with their anthocyanin content during growth and development of bracts. The curve of diurnal variation of EpPAL expression level in bracts exhibited two highest peaks at 9:00 and 18:00, respectively, and reached the lowest level at 12:00 in a clear day. With the maturation and senescence of bracts, expression levels reduced gradually in both green leaves and bracts, but decreased more rapidly in bracts than green leaves.Keywords: Cloning, expression, phenylalanine ammonia-lyase, poinsetti

    How insurance affects altruistic provision in threshold public good games

    The occurrence and maintenance of cooperative behaviors in public goods systems have attracted great research attention across multiple disciplines. A threshold public goods game requires a minimum amount of contributions to be collected from a group of individuals for provision to occur. Here we extend the common binary-strategy combination of cooperation and defection by adding a third strategy, called insured cooperation, which corresponds to buying an insurance covering the potential loss resulted from the unsuccessful public goods game. Particularly, only the contributing agents can opt to be insured, which is an effort decreasing the amount of the potential loss occurring. Theoretical computations suggest that when agents face the potential aggregate risk in threshold public goods games, more contributions occur with increasing compensation from insurance. Moreover, permitting the adoption of insurance significantly enhances individual contributions and facilitates provision, especially when the required threshold is high. This work also relates the strategy competition outcomes to different allocation rules once the resulted contributions exceed the threshold point in populations nested within a dilemma

    An Operational Statistical Scheme for Tropical Cyclone-Induced Rainfall Forecast

    Nonparametric methods are used in this study to analyze and predict short-term rainfall due to tropical cyclones (TCs) in a coastal meteorological station. All 427 TCs during 1953–2011, which made landfall along the Southeast China coast with a distance less than 700 km to a certain meteorological station, Shenzhen, are analyzed and grouped according to their landfalling direction, distance, and intensity. The corresponding daily rainfall records at Shenzhen Meteorological Station (SMS) during TCs landfalling period (a couple of days before and after TC landfall) are collected. The maximum daily rainfall (R24) and maximum 3-day accumulative rainfall (R72) records at SMS for each TC category are analyzed by a nonparametric statistical method, percentile estimation. The results are plotted by statistical boxplot, expressing in the probability of precipitation. The performance of the statistical boxplots was evaluated to forecast the short-term rainfall at SMS during the TC seasons in 2012 and 2013. The results show that the boxplot scheme can be used as a valuable reference to predict the short-term rainfall at SMS due to TCs landfalling along the Southeast China coast

    ABI4 Mediates Antagonistic Effects of Abscisic Acid and Gibberellins at Transcript and Protein Levels

    Abscisic acid (ABA) and gibberellins (GA) are plant hormones which antagonistically mediate numerous physiological processes, and their optimal balance is essential for normal plant development. However, the molecular mechanism underlying ABA and GA antagonism still needs to be determined. Here, we report that ABA- INSENSITIVE 4 (ABI4) is a central factor for GA/ABA homeostasis and antagonism in post-germination stages. ABI4 over-expression in Arabidopsis (OE-ABI4) leads to developmental defects including a decrease in plant height and poor seed production. The transcription of a key ABA biosynthetic gene, NCED6, and of a key GA catabolic gene, GA2ox7, is significantly enhanced by ABI4 over-expression. ABI4 activates NCED6 and GA2ox7 transcription by directly binding to the promoters, and genetic analysis revealed that mutation in these two genes partially rescues the dwarf phenotype of ABI4 overexpressing plants. Consistently, ABI4 overexpressing seedlings have a lower GA/ABA ratio compared to the wild type. We further show that ABA induces GA2ox7 transcription while GA represses NCED6 expression in an ABI4-dependent manner; and that ABA stabilizes the ABI4 protein, whereas GA promotes its degradation. Taken together, these results propose that ABA and GA antagonize each other by oppositely acting on ABI4 transcript and protein levels

Based on the six teaching links in the BOPPPS teaching mode, the case implementation process is analyzed and summarized. Practice has shown that in the project-based teaching process, the use of a new teaching mode that combines the BOPPPS teaching mode and information based teaching methods can effectively improve the remote teaching level of teachers and the remote learning effect of students

    Crucial role of strategy updating for coexistence of strategies in interaction networks

    Network models are useful tools for studying the dynamics of social interactions in a structured population. After a round of interactions with the players in their local neighborhood, players update their strategy based on the comparison of their own payoff with the payoff of one of their neighbors. Here we show that the assumptions made on strategy updating are of crucial importance for the strategy dynamics. In the first step, we demonstrate that seemingly small deviations from the standard assumptions on updating have major implications for the evolutionary outcome of two cooperation games: cooperation can more easily persist in a Prisoner's Dilemma game, while it can go more easily extinct in a Snowdrift game. To explain these outcomes, we develop a general model for the updating of states in a network that allows us to derive conditions for the steady-state coexistence of states (or strategies). The analysis reveals that coexistence crucially depends on the number of agents consulted for updating. We conclude that updating rules are as important for evolution on a network as network structure and the nature of the interaction

    The enzymatic activity of Arabidopsis protein arginine methyltransferase 10 is essential for flowering time regulation

    Arabidopsis AtPRMT10 is a plant-specific type I protein arginine methyltransferase that can asymmetrically dimethylate arginine 3 of histone H4 with auto-methylation activity. Mutations of AtPRMT10 derepress FLOWERING LOCUS C (FLC) expression resulting in a late-flowering phenotype. Here, to further investigate the biochemical characteristics of AtPRMT10, we analyzed a series of mutated forms of the AtPRMT10 protein. We demonstrate that the conserved "VLD" residues and "double-E loop" are essential for enzymatic activity of AtPRMT10. In addition, we show that Arg54 and Cys259 of AtPRMT10, two residues unreported in animals, are also important for its enzymatic activity. We find that Arg13 of AtPRMT10 is the auto-methylation site. However, substitution of Arg13 to Lys13 does not affect its enzymatic activity. In vivo complementation assays reveal that plants expressing AtPRMT10 with VLD-AAA, E143Q or E152Q mutations retain high levels of FLC expression and fail to rescue the late-flowering phenotype of atprmt10 plants. Taken together, we conclude that the methyltransferase activity of AtPRMT10 is essential for repressing FLC expression and promoting flowering in Arabidopsis

    Smad Ubiquitination Regulatory Factor 1 (Smurf1) Promotes Thyroid Cancer Cell Proliferation and Migration via Ubiquitin-Dependent Degradation of Kisspeptin-1

    Background/Aims: Thyroid cancer is the most common malignancy in human endocrine system. Smad ubiquitination regulatory factor 1 (Smurf1) is an E3 ubiquitin-protein ligase in ubiquitin-proteasome pathway (UPP) system. This study aimed to investigate the effects of Smurf1 on thyroid cancer proliferation and metastasis, as well as underlying potential mechanism. Methods: The expression levels of Smurf1 in thyroid tumor tissues and thyroid cancer cells were detected by western blotting and qRT-PCR. Then, the effects of up-regulation or down-regulation of Smurf1 on thyroid cancer cell viability, migration, invasion, proliferation and apoptosis were measured using trypan blue exclusion assay, two-chamber migration (invasion) assay, cell colony formation assay and Guava Nexin assay, respectively. The ubiquitination of kisspeptin-1 (KISS-1) was assessed by protein ubiquitination assay. Finally, the effects of KISS-1 overexpression on activity of nuclear factor-kappa B (NF-ÎşB) signaling pathway, as well as thyroid cancer cell viability, migration, invasion, proliferation and apoptosis were also detected, respectively. Results: Smurf1 was highly expressed in thyroid tumor tissues and thyroid cancer cells. Up-regulation of Smurf1 promoted the viability, migration, invasion and proliferation of thyroid cancer cells. Knockdown of Smurf1 had opposite effects. Moreover, smurf1 promoted the ubiquitination of KISS-1. Overexpression of KISS-1 inactivated NF-ÎşB pathway, suppressed thyroid cancer cell viability, migration, invasion and proliferation, and induced cell apoptosis. Conclusion: Up-regulation of Smurf1 exerted important roles in thyroid cancer formation and development by promoting thyroid cancer proliferation and metastasis. The ubiquitin-dependent degradation of KISS-1 induced by Smurf1 and the activation of NF-ÎşB signaling pathway might be involved in this process. Smurf1 could be an effective therapy target and biomarker for thyroid cancer treatment
