    Expressions of toll-like receptors 2 and 4, and relative cellular factors in HIV patients with tuberculosis infection

    Purpose: To investigate the expressions of toll-like receptor 2 (TLR2), toll-like receptor 4 (TLR4), tumor necrosis factor alpha (TNF-α), IFN-γ (IFN- gamma), interleukin 2 (IL-2), interleukin 6 (IL-6) and interleukin 10 (IL-10) in human immunodeficiency virus (HIV) patients with tuberculosis (TB) infection.Methods: Two groups of HIV patients (68 in each group) were used for this study. These were HIV with TB (HIV/TB) group and HIV without TB group. A third group (68 healthy people) served as control. Quantitative polymerase chain reaction (qPCR) was adopted to measure TLR-2 and TLR-4 expressions in peripheral blood mononuclear cells (PBMC), while the serum levels of TNF-α, IFN-γ, IL-2, IL-6 and IL-10 were determined by ELISA.Results: The △Ct values of TLR-2 and TLR-4 in HIV/TB and HIV groups were significantly lower than those in the control group (p < 0.05). Compared to control group, the serum levels of TNF-α, IL-6 and IL-10 significantly increased, while IFN-γ and IL-2 in HIV/TB and HIV groups significantly decreased (p < 0.05). However, IFN-γ and IL-2 decreased significantly in HIV/TB group (p < 0.05). Expression of TLR2 correlated positively with serum levels of TNF-α, IL-6 and IL-10, but negatively with IFN-γ and IL-2 (p < 0.05).Conclusion: TLR2 signal pathway plays a role in HIV patients with TB infection by promoting the expressions of TNF-α, IL-6 and IL-10, while inhibiting IFN-γ and IL-2 cellular factors, and thus may provide a new pathway for the treatment of patients with HIV/TB.Keywords: HIV, Tuberculosis, Toll-like receptor, Cellular factors, Tumor necrosis factor, Interleuki

    Study on the experiment and reaction kinetics of sulfur removal from coal by microorganisms

    To solve the safety problem of spontaneous combustion of high-sulfur coal, applied microbiology, physical chemistry, reaction kinetics theory, combined with the SEM, FTIR and TG-DTG-DSC experiments and analysis of testing methods, the microbial desulfurization experiments were carried out, and the change law of the desulfurization reaction of coal before and after the element composition, main physical and chemical properties, the coal spontaneous combustion point was studied. The results show that when the temperature is 30°C, the coal particle size is 120 mesh, the initial pH value is 2.0 and the bacteria liquid amount is 15 mL, the desulfurization effect of the coal sample is the best, and the maximum desulfurization rate can reach 75.12%. There is obvious erosion on the surface of the coal sample after microbial desulfurization, the pyrite in the coal is obviously reduced, and the molecular structure in the coal is basically unchanged. Under the action of microorganism, part of inorganic sulfur in coal is removed, the spontaneous combustion point of coal is increased by 50°C, the activation energy of coal has increased more than three times, and the possibility of spontaneous combustion of coal is reduced. By analyzing the reaction kinetics of the microbial desulfurization process, it can be seen that the microbial desulfurization reaction is controlled by external diffusion, internal diffusion and chemical reaction, among which internal diffusion is the main influencing factor

    Prognostic value of vitamin D in patients with pneumonia: A systematic review and meta-analysis

    Purpose: To investigate the prognostic role of vitamin D in pneumonia patients  through meta-analysis.Methods: PubMed and Embase were systematically searched for relevant studies that assessed the impact of vitamin D on the risk of adverse outcomes among patients with pneumonia. Risk ratios (RR) with 95 % confidence intervals (95 % CI) were pooled using meta-analysis. Q-test and I2 statistics were used to evaluate between-study heterogeneity.Results: Six studies were finally included in the meta-analysis. The results of meta-analysis of these studies indicated that low vitamin D status was associated with higher risk of mortality among pneumonia patients (RR = 2.59, 95 % CI = 1.32-5.08; p = 0.005). Results from meta-analysis of studies with adjusted estimates suggest that low vitamin D status was independently associated with higher risk of mortality among pneumonia patients (RR = 3.15, 95 % CI 1.54-6.44, p = 0.002). There was no significant risk of bias in the meta-analysis.Conclusion: This study demonstrates that low vitamin D level is associated with a higher risk of adverse outcomes in patients with pneumonia.Keywords: Pneumonia, Vitamin D, Prognosis, Meta-analysis, Systematic revie

    Effect modification of environmental factors on influenza-associated mortality: a time-series study in two Chinese cities

    Background: Environmental factors have been associated with transmission and survival of influenza viruses but no studies have ever explored the role of environmental factors on severity of influenza infection.Methods: We applied a Poisson regression model to the mortality data of two Chinese metropolitan cities located within the subtropical zone, to calculate the influenza associated excess mortality risks during the periods with different levels of temperature and humidity.Results: The results showed that high absolute humidity (measured by vapor pressure) was significantly (p < 0.05) associated with increased risks of all-cause and cardiorespiratory deaths, but not with increased risks of pneumonia and influenza deaths. The association between absolute humidity and mortality risks was found consistent among the two cities. An increasing pattern of influenza associated mortality risks was also found across the strata of low to high relative humidity, but the results were less consistent for temperature.Conclusions: These findings highlight the need for people with chronic cardiovascular and respiratory diseases to take extra caution against influenza during hot and humid days in the subtropics and tropics. © 2011 Yang et al; licensee BioMed Central Ltd.published_or_final_versio

    Using combination of lifting wavelet and multiclass SVM based on global optimization class strategy for fault pattern identification

    This paper presents a new method based on lifting wavelet for obtaining a fast multiclass SVM classification based on global optimization class strategy for fault diagnosis of roller bearing. Decision making was performed in two stages: feature extraction by computing the lifting wavelet coefficients and classification using the multiclass SVM classifiers trained on the extracted features. Experiments demonstrate that in comparison to discrete wavelet transform the lifting wavelet feature extraction can speed up the identification phase as well as achieve higher accuracy of multiclass SVM that is based on global optimization class strategy. Experimental results also reveal that the proposed multiclass SVM of global optimization is better than strategy of one against one and DAGSVM

    Levothyroxine Exhibits Inhibitory Effect towards UDP-Glucuronosyltransferase (UGT) 1A6-Mediated 4-Methylumbelliferone (4-MU) Glucuronidation

    Levothyroxine, a synthetic form of thyroid hormone, has been clinically used to treat thyroid hormone deficiency. UDP-glucuronosyltransferases (UGTs) are important phase II drug-metabolizing enzymes, and UGTs-inhibition based drug-drug interaction has been widely reported. The aim of the present study is to investigate the inhibitory effect of levothyroxine towards UGT1A6-catalyzed 4-methylumbelliferone (4-MU) glucuronidation. The results showed that levothyroxine inhibited UGT1A6-catalyzed 4-MU glucuronidation reaction in a dose-dependent manner. Furthermore, Lineweaver-Burk and Dixon plots showed the inhibition of UGT1A6 by levothyroxine was best fit to competitive inhibition, and the inhibition kinetic parameter (Ki ) was determined to be 15.3 μM. Given that UGT1A6 is one of the most important UGT isoforms catalyzing the glucuronidation reaction of many important clinical drugs, including aspirin and serotonin, inhibition of UGT1A6 activity by levothyroxine will significantly influence the pharmacokinetic behaviour of these drugs.     Similarities and Differences in the Fermiology of Kagome Metals AV3_{3}Sb5_{5} (A=K, Rb, Cs) Revealed by Shubnikov-de Haas Oscillations

    Materials with AV3_3Sb5_5 (A=K, Rb, Cs) stoichiometry are recently discovered kagome superconductors with the electronic structure featuring a Dirac band, van Hove singularities and flat bands. These systems undergo anomalous charge-density-wave (CDW) transitions at TCDWT_{\rm CDW}~80-100 K, resulting in the reconstruction of the Fermi surface from the pristine phase. Although comprehensive investigations of the electronic structure via quantum oscillations (QOs) have been performed on the sister compounds CsV3_3Sb5_5 and RbV3_3Sb5_5, a detailed QO study of KV3_3Sb5_5 is so far absent. Here, we report the Shubnikov-de Haas QO study in KV3_3Sb5_5. We resolve a large number of new frequencies with the highest frequency of 2202 T (occupying ~54% of the Brillouin zone area in the kxk_x-kyk_y plane). The Lifshitz-Kosevich analysis further gives relatively small cyclotron effective masses, and the angular dependence study reveals the two-dimensional nature of the frequencies with a sufficient signal-to-noise ratio. Finally, we compare the QO spectra for all three AV3_3Sb5_5 compounds collected under the same conditions, enabling us to point out the similarities and differences across these systems. Our results fill in the gap of the QO study in KV3_3Sb5_5 and provide valuable data to understand the band structure of all three members of AV3_3Sb5_5.Comment: 8 pages, 4 figure

    The CTNNBIP1-CLSTN1 fusion transcript regulates human neocortical development

    Fusion transcripts or RNAs have been found in both disordered and healthy human tissues and cells; however, their physiological functions in the brain development remain unknown. In the analysis of deposited RNA-sequence libraries covering early to middle embryonic stages, we identify 1,055 fusion transcripts present in the developing neocortex. Interestingly, 98 fusion transcripts exhibit distinct expression patterns in various neural progenitors (NPs) or neurons. We focus on CTNNBIP1-CLSTN1 (CTCL), which is enriched in outer radial glial cells that contribute to cortex expansion during human evolution. Intriguingly, downregulation of CTCL in cultured human cerebral organoids causes marked reduction in NPs and precocious neuronal differentiation, leading to impairment of organoid growth. Furthermore, the expression of CTCL fine-tunes Wnt/β-catenin signaling that controls cortex patterning. Together, this work provides evidence indicating important roles of fusion transcript in human brain development and evolution

    Transcription phase, protein characteristics of DEV UL45 and prokaryotic expression, antibody preparation of the UL45 des-transmembrane domain

    <p>Abstract</p> <p>Background</p> <p>Some UL45 gene function of Herpesvirus was reported. While there was no any report of the duck enteritis virus (DEV) UL45 protein as yet.</p> <p>Results</p> <p>The UL45 gene and des-transmembrane domain of UL45 (named UL45Δ gene, 295-675bp of UL45) of DEV were amplified by PCR and subcloned into the prokaryotic expression vector pET-32a(+). The constructed recombinant plasmids were transformed into the host strain BL21(DE3) PLysS and induced by IPTG. SDS-PAGE analysis showed the UL45 gene couldn't express while UL45Δ gene was highly expressed. His Purify Kit or salting-out could purify the protein effectively. Using the purified protein to immunize New-Zealand rabbits and produce polyclonal antibody. The agar diffusion reaction showed the titer of antibody was 1:32. Western blot analysis indicated the purified rabbit anti-UL45Δ IgG had a high level of specificity and the UL45 gene was a part of DEV genome. The transcription phase study of UL45 gene showed that expression of UL45 mRNA was at a low level from 0 to 18 h post-infection (pi), then accumulated quickly at 24 h pi and peaked at 42 h pi. It can be detected till 72 h pi. Besides, western blot analysis of purified virion and different viral ingredients showed that the UL45 protein resided in the purified virion and the viral envelope.</p> <p>Conclusions</p> <p>The rabbit anti-UL45Δ IgG was produced successfully and it can serve as a good tool for penetrating studies of the function of DEV UL45 protein. The transcription phase and protein characteristics analysis indicated that DEV UL45 gene was a late gene and UL45 protein may be a viral envelope protein.</p
