Periodically shellfish accumulate harm¬ful levels of a variety of algal toxins, including domoic acid, yessotoxins, pectenotoxins and brevetoxins. When this occurs, regulatory authorities may impose harvesting closures which have a consequential economic impact on both farmers and staff employed to harvest and market shellfish products

    A Cholecystokinin B Receptor-Specific Aptamer Does Not Activate Receptor Signaling

    Targeted nanoparticles which deliver effective doses of chemotherapeutic drugs directly to pancreatic tumors could improve treatment efficacy without the toxicities associated with systemic drug administration. One protein on tumor cells that can be targeted by nanoparticles is a G-protein coupled cell surface receptor, the cholecystokinin B receptor (CCKBR). Previously, we had shown that attaching the CCKBR ligand gastrin to the surface of nanoparticles can enhance their up-take by tumors. The drawback of using gastrin is that it can also activate the receptor, causing tumor cell growth. This study shows that a DNA aptamer that binds to the CCKBR and enhances nanoparticle up-take by tumors does not activate this receptor. PANC-1 cells, a cultured human pancreatic cancer cell line, were treated for 24 h with CCKBR aptamer 1153. Cell lysates were run on Bis-Tris gels, transferred to membranes, blocked in 5% BSA and incubated overnight with primary antibodies, including antibodies directly against phosphorylated-Akt (Ser473), total Akt, and beta-actin, a protein loading control. Although the CCKBR aptamer 1153 is internalized by pancreatic cancer cells in a receptor-mediated fashion, it does not stimulate cell proliferation. Because of this, we anticipate that it will not activate CCKBR signaling. If aptamer 1153 does not activate downstream receptor signaling, our future work will test whether the aptamer could be used to specifically direct drug-containing nanoparticles to tumors, making chemotherapy treatments for pancreatic cancer patients more effective with fewer off-target effects and toxicity

    A Comparison of EAST Shock-Tube Radiation Measurements with a New Air Radiation Model

    This paper presents a comparison between the recent EAST shock tube radiation measurements (Grinstead et al., AIAA 2008-1244) and the HARA radiation model. The equilibrium and nonequilibrium radiation measurements are studied for conditions relevant to lunar-return shock-layers; specifically shock velocities ranging from 9 to 11 kilometers per second at initial pressures of 0.1 and 0.3 Torr. The simulated shock-tube flow is assumed one-dimensional and is calculated using the LAURA code, while a detailed nonequilibrium radiation prediction is obtained in an uncoupled manner from the HARA code. The measured and predicted intensities are separated into several spectral ranges to isolate significant spectral features, mainly strong atomic line multiplets. The equations and physical data required for the prediction of these strong atomic lines are reviewed and their uncertainties identified. The 700-1020 nm wavelength range, which accounts for roughly 30% of the radiative flux to a peak-heating lunar return shock-layer, is studied in detail and the measurements and predictions are shown to agree within 15% in equilibrium. The plus or minus 1.5% uncertainty on the measured shock velocity is shown to cause up to a plus or minus 30% difference in the predicted radiation. This band of predictions contains the measured values in almost all cases. For the highly nonequilibrium 0.1 Torr cases, the nonequilibrium radiation peaks are under-predicted by about half. This under-prediction is considered acceptable when compared to the order-of-magnitude over-prediction obtained using a Boltzmann population of electronic states. The reasonable comparison in the nonequilibrium regions provides validation for both the non-Boltzmann modeling in HARA and the thermochemical nonequilibrium modeling in LAURA. The N2 (+)(1-) and N2(2+) molecular band systems are studied in the 290 480 nm wavelength range for both equilibrium and nonequilibrium regimes. The non-Boltzmann rate models for these systems, which have significant uncertainties, are tuned to improve the comparison with measurements

    Dynamic Shadow Removal from Front Projection Displays

    A technique and system for detecting a radiometric variation/artifacts of a front-projected dynamic display region under observation by at least one camera. The display is comprised of one or more images projected from one or more of a plurality of projectors; the system is preferably calibrated by using a projective relationship. A predicted image of the display region by the camera is constructed using frame-buffer information from each projector contributing to the display, which has been geometrically transformed for the camera and its relative image intensity adjusted. A detectable difference between a predicted image and the display region under observation causes corrective adjustment of the image being projected from at least one projector. The corrective adjustment may be achieved by way of pixel-wise approach (an alpha-mask is constructed from delta pixels/images), or bounding region approach (difference/bounding region is sized to include the area of the display affected by the radiometric variation).     The Combination of Organization and Emotion: An Immediate Free Recall Task

    Both organization (e.g., semantic-relatedness) and emotional valence have been found to lead to better memory as seen through the superior recall of semantically-related and emotionally-valenced lists. However, research suggests that these two factors may not only lack an additive effect when combined, but may result in worse recall. Based on this research, we hypothesized emotion would hinder recall of a semantically-related list by impairing the relational processing that usually benefits recall of semantically-related material. This hypothesis was supported, as it was found that emotional valence resulted in reduced recall in a semantically-related list. This finding may have important implications for fields like education and advertising, where the use of emotional stimuli within a message could cause a a person to miss its bigger picture

    Classroom belonging, self-efficacy, and perceived instrumentality: Influences on academic engagement and achievement.

    Self-efficacy, perceived instrumentality, belonging, achievement goals, cognitive engagement strategies, and enjoyment were measured using five questionnaires distributed and completed in the participant's English class. Achievement was defined as the percentage grade that each student earned at the conclusion of the spring semester. Correlations, regressions, and path models were used to analyze the data.The goal of the present study is to examine student perceptions of belonging, self-efficacy, and perceived instrumentality in terms of their unique and collective contribution to understanding student reported mastery goals, meaningful cognitive engagement strategies and achievement. Two hundred and forty-nine secondary students ranging in age from 14 to 19 years old from three high schools participated in the present study.Results indicated that there was a correlational relationship between belonging, self-efficacy, and perceived instrumentality. Belonging also explained additional variance beyond self-efficacy and perceived instrumentality when accounting for mastery goals, but not meaningful cognitive strategy use or achievement. Theoretical implications of the findings are discussed, as are limitations and implications for future research
