Polymorphisms in NF-kappa B, PXR, LXR, PPAR gamma and risk of inflammatory bowel disease
AIM: To investigate the contribution of polymorphisms in nuclear receptors to risk of inflammatory bowel disease (IBD). METHODS: Genotypes of nuclear factor (NF)-kappa B (NFKB1) NF kappa B -94ins/del (rs28362491); peroxisome proliferator-activated receptor (PPAR)-gamma (PPAR gamma) PPAR gamma Pro12Ala (rs 1801282) and C1431T (rs 3856806); pregnane X receptor (PXR) (NR1I2) PXR A-24381C (rs1523127), C8055T (2276707), and A7635G (rs 6785049); and liver X receptor (LXR) (NR1H2) LXR T-rs1405655-C and T-rs2695121-C were assessed in a Danish case-control study of 327 Crohn's disease patients, 495 ulcerative colitis (UC) patients, and 779 healthy controls. Odds ratio (OR) and 95% CI were estimated by logistic regression models. RESULTS: The PXR A7635G variant, the PPAR gamma Pro12Ala and LXR T-rs2695121-C homozygous variant genotypes were associated with risk of UC (OR: 1.31, 95% CI: 1.03-1.66, P = 0.03, OR: 2.30, 95% CI: 1.04-5.08, P = 0.04, and OR: 1.41, 95% CI: 1.00-1.98, P = 0.05, respectively) compared to the corresponding homozygous wild-type genotypes. Among never smokers, PXR A7635G and the LXR T-rs1405655-C and T-rs2695121-C variant genotypes were associated with risk of IBD (OR: 1.41, 95% CI: 1.05-1.91, P = 0.02, OR: 1.63, 95% CI: 1.21-2.20, P = 0.001, and OR: 2.02, 95% CI: 1.36-2.99, P = 0.0005, respectively) compared to the respective homozygous variant genotypes. PXR A7635G (rs6785049) variant genotype was associated with a higher risk of UC diagnosis before the age of 40 years and with a higher risk of extensive disease (OR: 1.34, 95% CI: 1.03-1.75 and OR: 2.49, 95% CI: 1.24-5.03, respectively). CONCLUSION: Common PXR and LXR polymorphisms may contribute to risk of IBD, especially among never smokers. (C) 2011 Baishideng. All rights reserved
Marker-dependent associations among oxidative stress, growth and survival during early life in a wild mammal
Oxidative stress (OS) is hypothesized to be a key physiological mechanism mediating life-history trade-offs, but evidence from wild populations experiencing natural environmental variation is limited. We tested the hypotheses that increased early life growth rate increases OS, and that increased OS reduces first-winter survival, in wild Soay sheep (Ovis aries) lambs. We measured growth rate and first-winter survival for four consecutive cohorts, and measured two markers of oxidative damage (malondialdehyde (MDA), protein carbonyls (PC)) and two markers of antioxidant (AOX) protection (total AOX capacity (TAC), superoxide dismutase (SOD)) from blood samples. Faster lamb growth was weakly associated with increased MDA, but not associated with variation in the other three markers. Lambs with higher SOD activity were more likely to survive their first winter, as were male but not female lambs with lower PC concentrations. Survival did not vary with MDA or total TAC. Key predictions relating OS to growth and survival were therefore supported in some OS markers, but not others. This suggests that different markers capture different aspects of the complex relationships between individual oxidative state, physiology and fitness, and that overarching hypotheses relating OS to life-history variation cannot be supported or refuted by studying individual markers
The impact of Hurricane Sandy on the shoreface and inner shelf of Fire Island, New York : large bedform migration but limited erosion

offshore the barrier island system of Fire Island, NY using before-and-after surveys involving
swath bathymetry, backscatter and CHIRP acoustic reflection data. As sea level rises over the
long term, the shoreface and inner shelf are eroded as barrier islands migrate landward; large
storms like Sandy are thought to be a primary driver of this largely evolutionary process. The
“before” data were collected in 2011 by the U.S. Geological Survey as part of a long-term
investigation of the Fire Island barrier system. The “after” data were collected in January, 2013,
~two months after the storm. Surprisingly, no widespread erosional event was observed. Rather,
the primary impact of Sandy on the shoreface and inner shelf was to force migration of major
bedforms (sand ridges and sorted bedforms) 10’s of meters WSW alongshore, decreasing in
migration distance with increasing water depth. Although greater in rate, this migratory behavior
is no different than observations made over the 15-year span prior to the 2011 survey.
Stratigraphic observations of buried, offshore-thinning fluvial channels indicate that long-term
erosion of older sediments is focused in water depths ranging from the base of the shoreface
(~13-16 m) to ~21 m on the inner shelf, which is coincident with the range of depth over which
sand ridges and sorted bedforms migrated in response to Sandy. We hypothesize that bedform
migration regulates erosion over these water depths and controls the formation of a widely
observed transgressive ravinement; focusing erosion of older material occurs at the base of the
stoss (upcurrent) flank of the bedforms. Secondary storm impacts include the formation of
ephemeral hummocky bedforms and the deposition of a mud event layer.This work was funded primarily by a rapid response grant from the Jackson

Impaired phosphate transport in SLC34A2 variants in patients with pulmonary alveolar microlithiasis
Background: Variants in SLC34A2 encoding the sodium-dependent phosphate transport protein 2b (NaPi-IIb) cause the rare lung disease pulmonary alveolar microlithiasis (PAM). PAM is characterised by the deposition of calcium-phosphate concretions in the alveoli usually progressing over time. No effective treatment is available. So far, 30 allelic variants in patients have been reported but only a few have been functionally characterised. This study aimed to determine the impact of selected SLC34A2 variants on transporter expression and phosphate uptake in cellular studies.
Methods: Two nonsense variants (c.910A > T and c.1456C > T), one frameshift (c.1328delT), and one in-frame deletion (c.1402_1404delACC) previously reported in patients with PAM were selected for investigation. Wild-type and mutant c-Myc-tagged human NaPi-IIb constructs were expressed in Xenopus laevis oocytes. The transport function was investigated with a 32Pi uptake assay. NaPi-IIb protein expression and localisation were determined with immunoblotting and immunohistochemistry, respectively.
Results: Oocytes injected with the wild-type human NaPi-IIb construct had significant 32Pi transport compared to water-injected oocytes. In addition, the protein had a molecular weight as expected for the glycosylated form, and it was readily detectable in the oocyte membrane. Although the protein from the Thr468del construct was synthesised and expressed in the oocyte membrane, phosphate transport was similar to non-injected control oocytes. All other mutants were non-functional and not expressed in the membrane, consistent with the expected impact of the truncations caused by premature stop codons.
Conclusions: Of four analysed SLC34A2 variants, only the Thr468del showed similar protein expression as the wild-type cotransporter in the oocyte membrane. All mutant transporters were non-functional, supporting that dysfunction of NaPi-IIb underlies the pathology of PAM
Challenging Social Cognition Models of Adherence:Cycles of Discourse, Historical Bodies, and Interactional Order
Attempts to model individual beliefs as a means of predicting how people follow clinical advice have dominated adherence research, but with limited success. In this article, we challenge assumptions underlying this individualistic philosophy and propose an alternative formulation of context and its relationship with individual actions related to illness. Borrowing from Scollon and Scollon’s three elements of social action – “historical body,” “interaction order,” and “discourses in place” – we construct an alternative set of research methods and demonstrate their application with an example of a person talking about asthma management. We argue that talk- or illness-related behavior, both viewed as forms of social action, manifest themselves as an intersection of cycles of discourse, shifting as individuals move through these cycles across time and space. We finish by discussing how these dynamics of social action can be studied and how clinicians might use this understanding when negotiating treatment with patients
NuSTAR Spectroscopy of Multi-Component X-ray Reflection from NGC 1068
We report on observations of NGC1068 with NuSTAR, which provide the best
constraints to date on its ~keV spectral shape. We find no strong
variability over the past two decades, consistent with its Compton-thick AGN
classification. The combined NuSTAR, Chandra, XMM-Newton, and Swift-BAT
spectral dataset offers new insights into the complex reflected emission. The
critical combination of the high signal-to-noise NuSTAR data and a spatial
decomposition with Chandra allow us to break several model degeneracies and
greatly aid physical interpretation. When modeled as a monolithic (i.e., a
single N_H) reflector, none of the common Compton-reflection models are able to
match the neutral fluorescence lines and broad spectral shape of the Compton
reflection. A multi-component reflector with three distinct column densities
(e.g., N_H~1.5e23, 5e24, and 1e25 cm^{-2}) provides a more reasonable fit to
the spectral lines and Compton hump, with near-solar Fe abundances. In this
model, the higher N_H components provide the bulk of the Compton hump flux
while the lower N_H component produces much of the line emission, effectively
decoupling two key features of Compton reflection. We note that ~30% of the
neutral Fe Kalpha line flux arises from >2" (~140 pc), implying that a
significant fraction of the <10 keV reflected component arises from regions
well outside of a parsec-scale torus. These results likely have ramifications
for the interpretation of poorer signal-to-noise observations and/or more

Discordant MZ twins With Cleft Lip and Palate: a model for identifying genes in complex traits.
Monozygotic (MZ) twins may be discordant for complex traits due to differential environmental exposure in utero, epigenetic variability in imprinting, X chromosome inactivation, or stochastic effects. Occasionally MZ twins may be discordant for chromosomal and single gene disorders due to somatic mosaicism. For complex traits, which are due to the interactive effects of multiple genes and environmental factors, the affected twin of a discordant MZ pair offers the possibility for identifying somatic mutations in candidate genes. DNA sequencing of candidate genes in discordant MZ twins can identify those rare etiologic mutational events responsible for the different phenotypes since the confounding effects of common single nucleotide polymorphisms are eliminated, as DNA sequences should be identical in MZ pairs. In this report we describe the extensive DNA sequencing of 18 candidate genes in a sample of MZ and dizygotic (DZ) twins with nonsyndromic cleft lip with or without cleft palate. We were unable to identify any somatic differences in approximately 34 Kb of DNA sequenced in 13 MZ pairs, for a total of approximately 900 Kb of sequence comparisons, supporting the hypothesis that nonetiologic posttwinning mutations are rare. While no etiologic variants were identified in this study, sequence comparisons of discordant MZ twins can serve as a tool for identifying etiologic mutations in clefting and other complex traits
