    Liquid Biopsy in Rare Cancers: Lessons from Hemangiopericytoma

    Hemangiopericytoma (HPT) is a rare mesenchymal tumor of fibroblastic type and for its rarity is poorly studied. The most common sites of metastatic disease in patients with intracranial HPT are the bone, liver, and lung, suggestive for an hematogenous dissemination; for this reason, we investigated, for the first time, the presence of circulating tumor cells (CTCs) in hemangiopericytoma patient by CellSearch® and SceenCell® devices. Peripheral blood samples were drawn and processed by CellSearch, an EpCAM-dependent device, and ScreenCell®, a device size based. We found nontypical CTCs by CellSearch system and the immunofluorescence analysis performed on CTCs isolate by ScreenCell demonstrated the presence of single CTCs and CTC clusters. The molecular characterization of single CTCs and CTC clusters, using antibodies directed against EpCAM, CD34, cytokeratins (8, 18, and 19), and CD45, showed a great heterogeneity in CTC clusters. We believe that the present study may open a new scenario in the rare tumors: the introduction of the liquid biopsy and the molecular characterization of circulating tumor cells could lead to personalized targeted treatments and also for rare tumors

    Exploring Land Use Scenarios in Metropolitan Areas: Food Balance in a Local Agricultural System by Using a Multi-objective Optimization Model☆

    Abstract The assessment of sustainability of agro-food systems is based on different and several tools. However, economic and policy implications of structural changes, such as land use change, and other modifications, for instance a different orientation or farming technique, could be mainly determined through scenario analysis. Especially in metropolitan regions, where the agricultural sector is threatened by various different pressures, a food supply adequately respondent to the demand is needed. In this sense the paper proposes the utilization of mathematical programming to assess some possible scenarios related to a higher compliance between food supply and demand. The approach is based on a linear programming model that takes into account the production and consumption dimensions in the Milan Metropolitan Area. Five scenarios are simulated, with different levels of sustainability and compliance with demand, demonstrating the potentialities of the regional agro-food system in adapting and adjusting itself to such modifications

    Fabrication and First Full Characterisation of Timing Properties of 3D Diamond Detectors

    Tracking detectors at future high luminosity hadron colliders are expected to be able to stand unprecedented levels of radiation as well as to efficiently reconstruct a huge number of tracks and primary vertices. To face the challenges posed by the radiation damage, new extremely radiation hard materials and sensor designs will be needed, while the track and vertex reconstruction problem can be significantly mitigated by the introduction of detectors with excellent timing capabilities. Indeed, the time coordinate provides extremely powerful information to disentangle overlapping tracks and hits in the harsh hadronic collision environment. Diamond 3D pixel sensors optimised for timing applications provide an appealing solution to the above problems as the 3D geometry enhances the already outstanding radiation hardness and allows to exploit the excellent timing properties of diamond. We report here the first full timing characterisation of 3D diamond sensors fabricated by electrode laser graphitisation in Florence. Results from a 270MeV pion beam test of a first prototype and from tests with a β source on a recently fabricated 55×55μm2 pitch sensor are discussed. First results on sensor simulation are also presented

    Sensitivity of Hydra vulgaris to Nanosilver for Environmental Applications

    Nanosilver applications, including sensing and water treatment, have significantly increased in recent years, although safety for humans and the environment is still under debate. Here, we tested the environmental safety of a novel formulation of silver nanoparticles functionalized with citrate and L-cysteine (AgNPcitLcys) on freshwater cnidarian Hydra vulgaris as an emerging ecotoxicological model for the safety of engineered nanomaterials. AgNPcitLcys behavior was characterized by dynamic light scattering (DLS), while Ag release was measured by inductively coupled plasma mass spectrometry (ICP-MS). H. vulgaris (n = 12) subjects were evaluated for morphological aberration after 96 h of exposure and regeneration ability after 96 h and 7 days of exposure, after which the predatory ability was also assessed. The results show a low dissolution of AgNPcitLcys in Hydra medium (max 0.146% of nominal AgNPcitLcys concentration) and highlight a lack of ecotoxicological effects, both on morphology and regeneration, confirming the protective role of the double coating against AgNP biological effects. Predatory ability evaluation suggests a mild impairment of the entangling capacity or of the functionality of the tentacles, as the number of preys killed but not ingested was higher than the controls in all exposed animals. While their long-term sub-lethal effects still need to be further evaluated on H. vulgaris, AgNPcitLcys appears to be a promising tool for environmental applications, for instance, for water treatment and sensing

    Adult separation anxiety in patients with complicated grief versus healthy control subjects: relationships with lifetime depressive and hypomanic symptoms

    <p>Abstract</p> <p>Background</p> <p>Around 9% to 20% of bereaved individuals experience symptoms of complicated grief (CG) that are associated with significant distress and impairment. A major issue is whether CG represents a distinctive nosographic entity, independent from other mental disorders, particularly major depression (MD), and the role of symptoms of adult separation anxiety. The purpose of this study was to compare the clinical features of patients with CG versus a sample of healthy control subjects, with particular focus on adult separation anxiety and lifetime mood spectrum symptoms.</p> <p>Methods</p> <p>A total of 53 patients with CG and 50 healthy control subjects were consecutively recruited and assessed by means of the Structured Clinical Interview for DSM-IV Axis-I disorders (SCID-I/P), Inventory of Complicated Grief (ICG), Adult Separation Anxiety Questionnaire (ASA-27), Work and Social Adjustment Scale (WSAS) and Mood Spectrum-Self Report (MOODS-SR) lifetime version.</p> <p>Results</p> <p>Patients with CG reported significantly higher scores on the MOODS-SR, ASA-27, and WSAS with respect to healthy control subjects. The scores on the ASA-27 were significantly associated with the MOODS-SR depressive and manic components amongst both patients and healthy control subjects, with a stronger association in the latter.</p> <p>Conclusions</p> <p>A major limitation of the present study is the small sample size that may reduce the generalizability of the results. Moreover, lifetime MOODS-SR does not provide information about the temporal sequence of the manic or depressive symptoms and the loss. The frequent comorbidity with MD and the association with both depressive and manic lifetime symptoms do not support the independence of CG from mood disorders. In our patients, CG is associated with high levels of separation anxiety in adulthood. However, the presence of lifetime mood instability, as measured by the frequent presence of depressive and hypomanic lifetime symptoms, suggests that cyclothymia might represent the common underlying feature characterizing the vulnerability to both adult separation anxiety and CG.</p

    Land consumption is currently one of the major problems affecting green spaces, natural or agricultural. This issue is very important both for agricultural system functionality and, consequently, for its permanence. This paper proposes an analyzing tool of land use changes to support planning policies, in order to highlight the major determinants of land use that affect the permanence of agricultural activity. The case studies are Lodi, Milan and Monza Brianza Provinces, in Lombardy Region.
