    The living aortic valve: From molecules to function.

    The aortic valve lies in a unique hemodynamic environment, one characterized by a range of stresses (shear stress, bending forces, loading forces and strain) that vary in intensity and direction throughout the cardiac cycle. Yet, despite its changing environment, the aortic valve opens and closes over 100,000 times a day and, in the majority of human beings, will function normally over a lifespan of 70-90 years. Until relatively recently heart valves were considered passive structures that play no active role in the functioning of a valve, or in the maintenance of its integrity and durability. However, through clinical experience and basic research the aortic valve can now be characterized as a living, dynamic organ with the capacity to adapt to its complex mechanical and biomechanical environment through active and passive communication between its constituent parts. The clinical relevance of a living valve substitute in patients requiring aortic valve replacement has been confirmed. This highlights the importance of using tissue engineering to develop heart valve substitutes containing living cells which have the ability to assume the complex functioning of the native valve

    The Luminous Convolution Model as an alternative to dark matter in spiral galaxies

    The Luminous Convolution Model (LCM) demonstrates that it is possible to predict the rotation curves of spiral galaxies directly from estimates of the luminous matter. We consider two frame-dependent effects on the light observed from other galaxies: relative velocity and relative curvature. With one free parameter, we predict the rotation curves of twenty-three (23) galaxies represented in forty-two (42) data sets. Relative curvature effects rely upon knowledge of both the gravitational potential from luminous mass of the emitting galaxy and the receiving galaxy, and so each emitter galaxy is compared to four (4) different Milky Way luminous mass models. On average in this sample, the LCM is more successful than either dark matter or modified gravity models in fitting the observed rotation curve data.     Identifying Planetary Biosignature Impostors: Spectral Features of CO and O4 Resulting from Abiotic O2/O3 Production

    O2 and O3 have been long considered the most robust individual biosignature gases in a planetary atmosphere, yet multiple mechanisms that may produce them in the absence of life have been described. However, these abiotic planetary mechanisms modify the environment in potentially identifiable ways. Here we briefly discuss two of the most detectable spectral discriminants for abiotic O2/O3: CO and O4. We produce the first explicit self-consistent simulations of these spectral discriminants as they may be seen by JWST. If JWST-NIRISS and/or NIRSpec observe CO (2.35, 4.6 um) in conjunction with CO2 (1.6, 2.0, 4.3 um) in the transmission spectrum of a terrestrial planet it could indicate robust CO2 photolysis and suggest that a future detection of O2 or O3 might not be biogenic. Strong O4 bands seen in transmission at 1.06 and 1.27 um could be diagnostic of a post-runaway O2-dominated atmosphere from massive H-escape. We find that for these false positive scenarios, CO at 2.35 um, CO2 at 2.0 and 4.3 um, and O4 at 1.27 um are all stronger features in transmission than O2/O3 and could be detected with SNRs \gtrsim 3 for an Earth-size planet orbiting a nearby M dwarf star with as few as 10 transits, assuming photon-limited noise. O4 bands could also be sought in UV/VIS/NIR reflected light (at 0.345, 0.36, 0.38, 0.445, 0.475, 0.53, 0.57, 0.63, 1.06, and 1.27 um) by a next generation direct-imaging telescope such as LUVOIR/HDST or HabEx and would indicate an oxygen atmosphere too massive to be biologically produced.Comment: 7 pages, 4 figures, accepted to the Astrophysical Journal Letter

    Assessment of the onset of action of afoxolaner against existing adult flea (Ctenocephalides felis) infestations on dogs

    AbstractThe speed of kill of afoxolaner against experimental infestations by Ctenocephalides felis was evaluated after oral administration of afoxolaner in a soft chew (NEXGARD®) at a dose to achieve 2.5mg/kg bodyweight. Forty beagles were allocated to two treatment groups. Dogs in Treatment Group 1 were untreated controls. Dogs in Treatment Group 2 were treated on Day-0 with afoxolaner, according to their pre-treatment bodyweight. All dogs were infested with approximately 100 C. felis on Day-1. Live fleas were counted upon removal at 5 time points after treatment (i.e., 2, 4, 8, 12 and 24h after treatment). For each time point, counts were performed on 4 dogs from each of the treated and the untreated groups. Early curative flea killing efficacy was evaluated with respect to the untreated control group. The afoxolaner treated group had significantly fewer fleas than the untreated control group at 8, 12, and 24h (p<0.001). The percent efficacies of orally administered afoxolaner were 15.0%, 87.8%, 99.5%, 100.0%, and 100.0% at 2, 4, 8, 12, and 24h, respectively. In this study, afoxolaner began killing fleas by 2h after treatment with increasing efficacy at subsequent time points and had >99.5% efficacy at 8, 12, and 24h after treatment demonstrating an early onset of action

    Effects of The Novel Endocannabinoid Uptake Inhibitor, LY2183240, on Fear-Potentiated Startle and Alcohol-Seeking Behaviors in Mice Selectively Bred for High Alcohol Preference

    Rationale Alcohol-use disorders often occur together with anxiety disorders in humans which may be partly due to common inherited genetic factors. Evidence suggests that the endocannabinoid system (ECS) is a promising therapeutic target for the treatment of individuals with anxiety and/or alcohol-use disorders. Objectives The present study assessed the effects of a novel endocannabinoid uptake inhibitor, LY2183240, on anxiety- and alcohol-seeking behaviors in a unique animal model that may represent increased genetic risk to develop co-morbid anxiety and alcohol-use disorders in humans. Mice selectively bred for high alcohol preference (HAP) show greater fear-potentiated startle (FPS) than mice selectively bred for low alcohol preference (LAP). We examined the effects of LY2183240 on the expression of FPS in HAP and LAP mice and on alcohol-induced conditioned place preference (CPP) and limited-access alcohol drinking behavior in HAP mice. Results Repeated administration of LY2183240 (30 mg/kg) reduced the expression of FPS in HAP but not LAP mice when given prior to a second FPS test 48 hrs after fear conditioning. Both the 10 and 30 mg/kg doses of LY2183240 enhanced the expression of alcohol-induced CPP and this effect persisted in the absence of the drug. LY2183240 did not alter limited-access alcohol drinking behavior, unconditioned startle responding, or locomotor activity. Conclusions These findings suggest that ECS modulation influences both conditioned fear and conditioned alcohol reward behavior. LY2183240 may be an effective pharmacotherapy for individuals with anxiety disorders, such as post-traumatic stress disorder, but may not be appropriate for individuals with co-morbid anxiety and alcohol-use disorders

    Rehabilitation following first-time patellar dislocation: a randomised controlled trial of purported vastus medialis obliquus muscle versus general quadriceps strengthening exercises.

    Purpose: Define whether distal vastus medialis (VM) muscle strengthening improves functional outcomes compared to general quadriceps muscles strengthening following first-time patellar dislocation (FTPD). Methods: Fifty patients post-FTPD were randomised to either a general quadriceps exercise or rehabilitation programme (n=25) or to a specific-VM exercise and rehabilitation regime (n=25). Primary outcome was the Lysholm Knee Score, secondary outcomes included the Tegner Level of Activity Scale, the Norwich Patellar Instability (NPI) Score, and isometric knee extensions strength at various knee flexion ranges of motion. Outcomes were assessed at baseline, six weeks, six months and 12 months. Results: There were statistically significant differences in functional outcome and activity levels through the Lysholm Knee Score and Tegner Level of Activity Scale at 12 months in the general quadriceps exercise group compared to the VM group (p=0.05; 95% CI: -14.0 to 0.0/p=0.04; 95% CI: -3.0 to 0.0). This did not reach a clinically important difference. There was no statistically significant difference between the groups for the NPI Score and isometric strength at any follow-up interval. The trial experienced substantial participant attrition (52% at 12 months). Conclusions: Whilst there was a statistical difference in Lysholm Knee Score and Tegner Level of Activity Score between general quadriceps and VM exercise groups at 12 months, this may not have necessarily been clinically important. This trial highlights that the recruitment and retention of participants from this population is a challenge and should be considered during the design of future trials in this populatio

    Exploring the Reality of Hog Raisers in Cebu Amidst the African Swine Fever (ASF) Outbreak

    Hog farming is a significant industry in the Philippines, providing 60% of the country's animal meat consumption. However, the African Swine Fever (ASF) outbreak has caused its collapse. In effect, the hog raisers stopped their operation and sought alternatives to earn money for the family's needs. As a result, a phenomenological study investigated the hog raisers' lived experiences and how they cope with the existing phenomenon. Six key informants were selected based on inclusion criteria and purposive sampling for one-on-one interviews conducted by researchers. Using Creswell's approach, (1) fear of the virus, (2) frustration from the border controls and lockdowns, (3) broken promises of financial assistance from the Department of Agriculture, and (4) incurred substantial financial loss were themes concerning the lived experiences of the key informants. Also, the key informants shared how they coped with the outbreak, including (1) shifting to a farming business, (2) working in a construction site, and (3) making hollow blocks. An effective approach to combat the ASF outbreak in Cebu necessitates a comprehensive strategy that draws on insights from key informants. It is imperative to establish systems of support that consider the financial, emotional, and psychological needs of those affected. Collaborative efforts with local communities, researchers, and government organizations are also crucial to devising sustainable plans for managing and preventing ASF in the long run. This study sheds light on the obstacles encountered by the hog industry in Cebu and underscores the significance of implementing coordinated measures to mitigate the effects of ASF

    Carving out OPE space and precise O(2) model critical exponents

    We develop new tools for isolating CFTs using the numerical bootstrap. A “cutting surface” algorithm for scanning OPE coefficients makes it possible to find islands in high-dimensional spaces. Together with recent progress in large-scale semidefinite programming, this enables bootstrap studies of much larger systems of correlation functions than was previously practical. We apply these methods to correlation functions of charge-0, 1, and 2 scalars in the 3d O(2) model, computing new precise values for scaling dimensions and OPE coefficients in this theory. Our new determinations of scaling dimensions are consistent with and improve upon existing Monte Carlo simulations, sharpening the existing decades-old 8σ discrepancy between theory and experiment
