    In this article, the author conducted preliminary research on public transportation systems in four countries to aid in the development of a Comparative Public Administration course. The four countries (Canada, Mexico, the United Kingdom and South Africa) each offer unique challenges regarding mass transit. Increasing investment in mass public transit is vital to controlling urban pollution, reducing dependency on foreign oil, and reducing traffic congestion. Additionally, the article looks at the use of sabbaticals as a tool for researchers, both as a method for revitalizing professionals in the academic world and as a way to encourage research on subjects which may be less well-funded. The research will be incorporated into the course as an example of the kinds of research the students can do themselves, regardless of area of interest

    Sensitization of renal carcinoma cells to TRAIL-induced apoptosis by rocaglamide and analogs

    Rocaglamide has been reported to sensitize several cell types to TRAIL-induced apoptosis. In recent years, advances in synthetic techniques have led to generation of novel rocaglamide analogs. However, these have not been extensively analyzed as TRAIL sensitizers, particularly in TRAIL-resistant renal cell carcinoma cells. Evaluation of rocaglamide and analogs identified 29 compounds that are able to sensitize TRAIL-resistant ACHN cells to TRAIL-induced, caspase-dependent apoptosis with sub-µM potency which correlated with their potency as protein synthesis inhibitors and with loss of cFLIP protein in the same cells. Rocaglamide alone induced cell cycle arrest, but not apoptosis. Rocaglates averaged 4–5-fold higher potency as TRAIL sensitizers than as protein synthesis inhibitors suggesting a potential window for maximizing TRAIL sensitization while minimizing effects of general protein synthesis inhibition. A wide range of other rocaglate effects (e.g. on JNK or RAF-MEK-ERK signaling, death receptor levels, ROS, ER stress, eIF4E phosphorylation) were assessed, but did not contribute to TRAIL sensitization. Other than a rapid loss of MCL-1, rocaglates had minimal effects on mitochondrial apoptotic pathway proteins. The identification of structurally diverse/mechanistically similar TRAIL sensitizing rocaglates provides insights into both rocaglate structure and function and potential further development for use in RCC-directed combination therapy.This project has been funded in whole or in part with Federal funds from the National Cancer Institute, National Institutes of Health, under Contract No. HHSN261200800001E.     The Phenomenon of Collaboration: A Phenomenologic Study of Collaboration between Family Medicine and Obstetrics and Gynecology Departments at an Academic Medical Center

    Collaboration is essential to manage complex real world problems. We used phenomenologic methods to elaborate a description of collaboration between two departments at an academic medical center who considered their relationship to represent a model of effective collaboration. Key collaborative structures included a shared vision and commitment by leaders, rigorous quality improvement, clear delineation of roles with built-in flexibility, ongoing commitment to formal and informal communication channels and conflict resolution, relationship development grounded in respect and responsiveness, and shared training in a supportive learning environment with legitimate participation fostering skill development. This study reveals the complexity and resources required for collaboration which both explains why collaboration is not as easy to achieve and identifies processes that foster collaboration

    Learning Our Way Through: Collaborative Self-study in an Evolving Professional Development School (PDS) Partnership

    Purpose of study: Professional development schools (PDSs) are essentially learning communities in which all participants increase their knowledge about how teaching and learning works and how best to manage the collaborative enterprise (Goodman, 2002; Horn, 2007; Mantle- Bromley, 2002; Patrizio & Gadja, 2007; Sue, 2002). The National Council for the Accreditation of Teachers (NCATE, 2010), identified PDS as an avenue through which aspiring teachers can be provided the opportunity to integrate theory with practice. In addition, PDS partnerships serve as a vehicle for the professionalization of teachers and systematic examination and evaluation of practice. However, according to the National Association of Professional Development Schools (NAPDS, 2008), many colleges and universities who participate in PDS partnerships do not fully understand the true meaning of PDS. This creates a void between the concept of PDS as originally proposed (Holmes Group, 1990), and the reality of the PDS as it operates in many of the partnerships (Webb-Dempsey, Steel, Shambaugh and Dampsey, 2007). In addition, while interorganizational collaboration is a PDS imperative, it remains complex, multilayered and labor-intensive for both school and university faculty (Patrizio & Gadja, 2007; Rice & Afman, 2002; Su, 2002). This situation calls for a clear understanding of the concept of PDS as well as frequent and systematic review of the goals and objectives of PDS partnerships. The purpose of this study was to better understand one teaching college\u27s collaborative relationship with the partner schools. The study is guided by the following questions. What was the nature of the collaboration between a teaching college and five urban-based PDSs? What were the individual and collective experiences of the faculty as well as other stakeholders in the collaboration? What factors, if any, impacted the collaboration

    Chronic Systemic Immune Dysfunction in African-Americans with Small Vessel-Type Ischemic Stroke

    The incidence of small vessel-type (lacunar) ischemic strokes is greater in African-Americans compared to whites. The chronic inflammatory changes that result from lacunar stroke are poorly understood. To elucidate these changes, we measured serum inflammatory and thrombotic biomarkers in African-Americans at least 6 weeks post-stroke compared to control individuals. Cases were African-Americans with lacunar stroke (n = 30), and controls were age-matched African-Americans with no history of stroke or other major neurologic disease (n = 37). Blood was obtained \u3e 6 weeks post-stroke and was analyzed for inflammatory biomarkers. Freshly isolated peripheral blood mononuclear cells were stimulated with lipopolysaccharide (LPS) to assess immune responsiveness in a subset of cases (n = 5) and controls (n = 4). After adjustment for covariates, the pro-inflammatory biomarkers, soluble vascular cadherin adhesion molecule-1 (sVCAM-1) and thrombin anti-thrombin (TAT), were independently associated with lacunar stroke. Immune responsiveness to LPS challenge was abnormal in cases compared to controls. African-Americans with lacunar stroke had elevated blood levels of VCAM-1 and TAT and an abnormal response to acute immune challenge \u3e 6 weeks post-stroke, suggesting a chronically compromised systemic inflammatory response

    Social and Environmental Factors Associated with Preschoolers' Non-sedentary Physical Activity

    The two-fold purpose of the investigation was (1) to describe with direct observation data the physical activity behaviors and the accompanying social and environmental events of those behaviors for children in preschools; and (2) to determine which contextual conditions were predictors of moderate-to-vigorous physical activity (MVPA) and non-sedentary physical activity (i.e., light activity + MVPA) for 3-, 4-, and 5-year-old children during their outdoor play. The results indicate that preschoolers' physical activity is characterized as sedentary in nature throughout their preschool day (i.e., 89% sedentary, 8% light activity, 3% MVPA). During outdoor play periods, when children are most likely to be physically active, some contextual and social circumstances better predict their physical activity.     Seasonal patterns of estuarine acidification in seagrass beds of the Snohomish Estuary, WA

    Recent studies have begun to explore physical and biogeochemical mechanisms of carbonate chemistry variability in a variety of coastal habitats, including coral reefs, upwelling margins, and inland seas. To our knowledge, there have been limited mechanistic studies of annual carbonate chemistry variability in nearshore estuarine environments. Here, we present autonomous sensor and grab sample data of carbonate chemistry covering a 10 month period from two subtidal seagrass bed sites in Possession Sound, WA. Simple mass balance stoichiometric models are used to evaluate seasonal drivers of carbonate system parameters in the seagrass beds. Simulations of increasing anthropogenic carbon (Canth) burdens in the habitats reveal seasonal differences in the magnitude of carbonate system responses. The addition of Canth alters the thermodynamic buffer factors (e.g. the Revelle factor) of the carbonate system, decreasing the system’s ability to buffer natural variability in the seagrass habitat on high-frequency (e.g. tidal, diel) and seasonal timescales. As a result, the most harmful carbonate system indices for many estuarine organisms (minimum pHT, minimum Ωarag, and maximum pCO2(s.w.)) change most rapidly with increasing Canth. We highlight how the observed seasonal climatology and non-linear response of the carbonate system to increasing Canth drive the timing of the crossing of established physiological stress thresholds for endemic organisms, as well as thresholds relevant for water quality management. In this system, the relative benefits of the seagrass beds in locally mitigating ocean acidification during the growing season increase with the higher atmospheric CO2 levels predicted toward 2100. Presently however, these mitigating effects are mixed due to intense diel cycling of CO2 driven by community metabolism

    Proximate and fatty acid composition of 40 southeastern U.S. finfish species

    This report describes the proximate compositions (protein, moisture, fat, and ash) and major fatty acid profiles for raw and cooked samples of 40 southeastern finfish species. All samples (fillets) were cooked by a standard procedure in laminated plastic bags to an internal temperature of 70'C (lS8'F). Both summarized compositional data, with means and ranges for each species, and individual sample data including harvest dates and average lengths and weights are presented. When compared with raw samples, cooked samples exhibited an increase in protein content with an accompanying decrease in moisture content. Fat content either remained approximately the same or increased due to moisture loss during cooking. Our results are discussed in reference to compositional data previously published by others on some of the same species. Although additional data are needed to adequately describe the seasonal and geographic variations in the chemical compositions of many of these fish species, the results presented here should be useful to nutritionists, seafood marketers, and consumers.(PDF file contains 28 pages.

    Effects of Climate Change on Water Quality in the Yaquina Estuary, Oregon

    As part of a larger study to examine the effect of climate change (CC) on estuarine resources, we simulated the effect of rising sea level, alterations in river discharge, and increasing atmospheric temperatures on water quality in the Yaquina Estuary. Due to uncertainty in the effects of climate change, initial model simulations were performed for different steady river discharge rates that span the historical range in inflow, and for a range of increases in sea level and atmospheric temperature. Model simulations suggest that in the central portion of the estuary (19 km from mouth), a 60-cm increase in sea level will result in a 2-3 psu change in salinity across a broad range of river discharges. For the oligohaline portion of the estuary, salinity increases associated with a rise in sea level of 60 cm are only apparent at low river discharge rates (\u3c 50 m3 s-1). Simulations suggest that the water temperatures near the mouth of the estuary will decrease due to rising sea level, while water temperatures in upriver portions of the estuary will increase due to rising atmospheric temperatures. We present results which demonstrate how the interaction of changes in river discharge, rising sea level, and atmospheric temperature associated with climate change produce non-linear patterns in the response of estuarine salinity and temperature, which vary with location inside the estuary and season. We also will discuss the importance of presenting results in a manner that incorporates uncertainty in climate projections
