    Perceptions of council member– department head interactions in local government

    In this article, we use data collected from municipal council members and department heads in Michigan municipalities with over 10 000 residents to determine how, and why, they view the quality of their interactions with one another. Building theories of small group dynamics and political control of bureaucracy, we test several hypotheses and conclude that council members and department heads hold divergent views of their interactions with one another and that their views are determined by government form and community characteristics. We conclude with simple steps that local government officials and administrators can take to improve their small group dynamics and governing performance

    Organisational Coaching Outcomes: A comparison of a practitioner survey and key findings from the literature

    This paper compares the measurements of organisational outcomes from executive and leadership coaching based on existing studies and research that has been conducted throughout the literature. An overview of executive and leadership coaching is provided and a case study measuring executive and leadership coaching effectiveness is reported. The survey methodology used in this case study was not originally intended for an academic research design, rather it was a customer satisfaction survey. However, the results can provide insights into the value, ROI and impact of executive and leadership coaching

    Publishing Interactive Articles: Integrating Journals And Biological Databases

    In collaboration with the journal GENETICS, we've developed and launched a pipeline by which interactive full-text HTML/PDF journal articles are published with named entities linked to corresponding resource pages in "WormBase":http://www.wormbase.org/ (WB). Our interactive articles allow a reader to click on over ten different data type objects (gene, protein, transgene, etc.) and be directed to the relevant webpage. This seamless connection from the article to summaries of data types promotes a deeper level of understanding for the naïve reader, and incisive evaluation for the sophisticated reader. Further, this collaboration allows us to identify and collect information before the publication of the article. The pipeline uses automated recognition scripts to identify entities that already exist in the database and a self-reporting form we created at WB that is sent to the author by GENETICS for submitting entities that do not already exist in our database. We include a manual quality control step to make sure ambiguous links are corrected, and that all new entities have been reported and linked properly. The automated entity recognition scripts allows us to potentially link any object found in a database as well as to expand this pipeline to other databases. We have already adapted this pipeline for linking _Saccharomyces cerevisiae_ GENETICS articles to the "Saccharomyces Genome Database":http://www.yeastgenome.org/ (SGD) and are currently expanding this pipeline for linking genes in _Drosophila_ articles to "FlyBase":http://flybase.org/. By integrating journals and databases, we are integrating the major modes of communication in the biological sciences, which will undoubtedly increase the pace of discovery.

    CHK1 inhibition is synthetically lethal with loss of B-family DNA polymerase function in human lung and colorectal cancer cells

    Checkpoint kinase 1 (CHK1) is a key mediator of the DNA damage response that regulates cell cycle progression, DNA damage repair and DNA replication. Smallmolecule CHK1 inhibitors sensitise cancer cells to genotoxic agents and have shown single agent preclinical activity in cancers with high levels of replication stress. However, the underlying genetic determinants of CHK1 inhibitor sensitivity remain unclear. We used the developmental clinical drug SRA737 in an unbiased largescale siRNA screen to identify novel mediators of CHK1 inhibitor sensitivity and uncover potential combination therapies and biomarkers for patient selection. We identified members of the B-family of DNA polymerases (POLA1, POLE and POLE2) whose silencing sensitised the human A549 non small cell lung cancer (NSCLC) and SW620 colorectal cancer cell lines to SRA737. B-family polymerases were validated using multiple siRNAs in a panel of NSCLC and colorectal cancer cell lines. Replication stress, DNA damage and apoptosis were increased in human cancer cells following depletion of the B-family DNA polymerases combined with SRA737 treatment. Moreover, pharmacological blockade of B-family DNA polymerases using aphidicolin or CD437 combined with CHK1 inhibitors led to synergistic inhibition of cancer cell proliferation. Furthermore, low levels of POLA1, POLE and POLE2 protein expression in NSCLC and colorectal cancer cells correlated with single agent CHK1 inhibitor sensitivity and may constitute biomarkers of this phenotype. These findings provide a potential basis for combining CHK1 and B-family polymerase inhibitors in cancer therapy

    All-Sky Monitoring of Variable Sources with Fermi GBM

    Using the Gamma ray Burst Monitor (GBM) on Fermi, we monitor the transient hard X-ray/soft gamma ray sky. The twelve GBM NaI detectors span 8 keV to 1 MeV, while the two BGO detectors span 150 keV to 40 MeV. We use the Earth occultation technique to monitor a number of sources, including X-ray binaries, AGN, and solar flaring activity. Our monitoring reveals predictable and unpredictable phenomena such as transient outbursts and state changes. With GBM we also track the pulsed flux and spin frequency of accretion powered pulsars using epoch-folding techniques. Searches for quasi-periodic oscillations and X-ray bursts are also possible with GBM all-sky monitoring. Highlights from the Earth Occultation and Pulsar projects will be presented including our recent surprising discovery of variations in the total flux from the Crab. Inclusion of an all-sky monitor is crucial for a successful future X-ray timing mission
