An opioid-like system regulating feeding behavior in C. elegans
Neuropeptides are essential for the regulation of appetite. Here we show that neuropeptides could regulate feeding in mutants that lack neurotransmission from the motor neurons that stimulate feeding muscles. We identified nlp-24 by an RNAi screen of 115 neuropeptide genes, testing whether they affected growth. NLP-24 peptides have a conserved YGGXX sequence, similar to mammalian opioid neuropeptides. In addition, morphine and naloxone respectively stimulated and inhibited feeding in starved worms, but not in worms lacking NPR-17, which encodes a protein with sequence similarity to opioid receptors. Opioid agonists activated heterologously expressed NPR-17, as did at least one NLP-24 peptide. Worms lacking the ASI neurons, which express npr-17, did not response to naloxone. Thus, we suggest that Caenorhabditis elegans has an endogenous opioid system that acts through NPR-17, and that opioids regulate feeding via ASI neurons. Together, these results suggestC. elegans may be the first genetically tractable invertebrate opioid model
Becoming Bridge Citizens: Educating for Social Justice in Conflict-Affected Settings
This study draws on empirical data to fine-tune the theoretical concept, ‘bridging civic identity’, which we propose as an educational aim in conflict-affected settings. We analyse interview data from Liberian respondents and North Korean migrants living in South Korea, using a conceptual framework based on the notions of ‘bridge citizens’ and agency. The analysis reveals the following: (1) that a high sense of agency is related to resourcefulness and fortitude, (2) that identifying oneself as a ‘bridge citizen’ is connected to recognising others as such, and (3) that concrete, large-scale aspirations of social justice for the larger community – and therefore ‘imaginativeness’ – are central components of a bridging civic identity. The findings suggest that learners in similar settings ought to be trained in resourcefulness and fortitude, be shown the collective nature of working towards shared goals, and be given encouragement to visualise the just future they desire for their community or nation
Starvation-induced collective behavior in C. elegans
We describe a new type of collective behavior in C. elegans nematodes, aggregation of starved L1 larvae. Shortly after hatching in the absence of food, L1 larvae arrest their development and disperse in search for food. In contrast, after two or more days without food, the worms change their behavior—they start to aggregate. The aggregation requires a small amount of ethanol or acetate in the environment. In the case of ethanol, it has to be metabolized, which requires functional alcohol dehydrogenase sodh-1. The resulting acetate is used in de novo fatty acid synthesis, and some of the newly made fatty acids are then derivatized to glycerophosphoethanolamides and released into the surrounding medium. We examined several otherCaenorhabditis species and found an apparent correlation between propensity of starved L1s to aggregate and density dependence of their survival in starvation. Aggregation locally concentrates worms and may help the larvae to survive long starvation. This work demonstrates how presence of ethanol or acetate, relatively abundant small molecules in the environment, induces collective behavior in C. elegans associated with different survival strategies
Collective autobiographical reflexivity on active and compassionate citizenship in the COVID-19 crisis
Since the outbreak of the coronavirus pandemic, socio-economic inequalities have become exacerbated and COVID-19 related hate crimes have increased. This paper explores how citizenship education might be reimagined in response to this context, with the vision of rebuilding a more equitable and compassionate society. By using a collective autobiographical writing approach, this study documented six different autobiographical reflexivities of citizenship education scholars who were from different parts of the world: China, South Korea, the Philippines, the United States, Nepal and the United Kingdom. It also observed the way the pandemic played out in the location where they were situated during the research as well as how it played out in their countries of origin, and further, how it affected the civic development in each context. The scholars\u27 range of autobiographical expressions resulted in insights for developing a type of citizenship education, namely, education for active and compassionate citizenship

Caenorhabditis elegans as a model system to identify therapeutics for alcohol use disorders
Alcohol use disorders (AUDs) cause serious problems in society and few effective treatments are available. Caenorhabditis elegans (C. elegans) is an excellent invertebrate model to study the neurobiological basis of human behavior with a conserved, fully tractable genome, and a short generation time for fast generation of data at a fraction of the cost of other organisms. C. elegans demonstrate movement toward, and concentration-dependent self-exposure to various psychoactive drugs. The discovery of opioid receptors in C. elegans provided the impetus to test the hypothesis that C. elegans may be used as a medications screen to identify new AUD treatments. We tested the effects of naltrexone, an opioid antagonist and effective treatment for AUDs, on EtOH preference in C. elegans. Six-well agar test plates were prepared with EtOH placed in a target zone on one side and water in the opposite target zone of each well. Worms were treated with naltrexone before EtOH preference testing and then placed in the center of each well. Wild-type worms exhibited a concentration-dependent preference for 50, 70 and 95% EtOH. Naltrexone blocked acute EtOH preference, but had no effect on attraction to food or benzaldehyde in wild-type worms. Npr-17 opioid receptor knockout mutants did not display a preference for EtOH. In contrast, npr-17 opioid receptor rescue mutants exhibited significant EtOH preference behavior, which was attenuated by naltrexone. Chronic EtOH exposure induced treatment resistance and compulsive-like behavior. These data indicate that C. elegans can serve as a model system to identify compounds to treat AUDs
Effect of magnetic order on the superfluid response of single-crystal ErNiBC: A penetration depth study
We report measurements of the in-plane magnetic penetration depth (T) in single crystals of ErNiBC down to 0.1 K using
a tunnel-diode based, self-inductive technique at 21 MHz. We observe four
features: (1) a slight dip in (T) at the Nel
temperature = 6.0 K, (2) a peak at = 2.3 K, where a weak
ferromagnetic component sets in, (3) another maximum at 0.45 K, and (4) a final
broad drop down to 0.1 K. Converting to superfluid density , we see
that the antiferromagnetic order at 6 K only slightly depresses
superconductivity. We seek to explain some of the above features in the context
of antiferromagnetic superconductors, where competition between the
antiferromagnetic molecular field and spin fluctuation scattering determines
increased or decreased pairbreaking. Superfluid density data show only a slight
decrease in pair density in the vicinity of the 2.3 K feature, thus supporting
other evidences against bulk ferromagnetism in this temperature range.Comment: 15 pages, 5 figure
Microtubule distribution in somatic cell nuclear transfer bovine embryos following control of nuclear remodeling type
This study was conducted to evaluate the microtubule distribution following control of nuclear remodeling by treatment of bovine somatic cell nuclear transfer (SCNT) embryos with caffeine or roscovitine. Bovine somatic cells were fused to enucleated oocytes treated with either 5 mM caffeine or 150 µM roscovitine to control the type of nuclear remodeling. The proportion of embryos that underwent premature chromosome condensation (PCC) was increased by caffeine treatment but was reduced by roscovitine treatment (p < 0.05). The microtubule organization was examined by immunostaining β- and γ-tubulins at 15 min, 3 h, and 20 h of fusion using laser scanning confocal microscopy. The γ-tubulin foci inherited from the donor centrosome were observed in most of the SCNT embryos at 15 min of fusion (91.3%) and most of them did not disappear until 3 h after fusion, regardless of treatment (82.9-87.2%). A significantly high proportion of embryos showing an abnormal chromosome or microtubule distribution was observed in the roscovitine-treated group (40.0%, p < 0.05) compared to the caffeine-treated group (22.1%). In conclusion, PCC is a favorable condition for the normal organization of microtubules, and inhibition of PCC can cause abnormal mitotic division of bovine SCNT embryos by causing microtubule dysfunction
