    Integrated nontargeted and targeted metabolomics analyses amino acids metabolism in infantile hemangioma

    Infantile hemangioma (IH) is the most common benign tumor in children. However, the exact pathogenesis of IH remains unclear. Integrated nontargeted and targeted metabolic analyses were performed to obtain insight into the possible pathogenic mechanism of IH. The results of nontargeted metabolic analysis showed that 216 and 128 differential metabolites (DMs) were identified between hemangioma-derived endothelial cells (HemECs) and HUVECs in positive-ion and negative-ion models, respectively. In both models, these DMs were predominantly enriched in pathways related to amino acid metabolism, including aminoacyl-tRNA biosynthesis and arginine and proline metabolism. Then, targeted metabolic analysis of amino acids was further performed to further clarify HemEC metabolism. A total of 22 amino acid metabolites were identified, among which only 16 metabolites, including glutamine, arginine and asparagine, were significantly differentially expressed between HemECs and HUVECs. These significant amino acids were significantly enriched in 10 metabolic pathways, including ‘alanine, aspartate and glutamate metabolism’, ‘arginine biosynthesis’, ‘arginine and proline metabolism’, and ‘glycine, serine and threonine metabolism’. The results of our study revealed that amino acid metabolism is involved in IH. Key differential amino acid metabolites, including glutamine, asparagine and arginine, may play an important role in regulating HemEC metabolism

    LiveVV: Human-Centered Live Volumetric Video Streaming System

    Volumetric video has emerged as a prominent medium within the realm of eXtended Reality (XR) with the advancements in computer graphics and depth capture hardware. Users can fully immersive themselves in volumetric video with the ability to switch their viewport in six degree-of-freedom (DOF), including three rotational dimensions (yaw, pitch, roll) and three translational dimensions (X, Y, Z). Different from traditional 2D videos that are composed of pixel matrices, volumetric videos employ point clouds, meshes, or voxels to represent a volumetric scene, resulting in significantly larger data sizes. While previous works have successfully achieved volumetric video streaming in video-on-demand scenarios, the live streaming of volumetric video remains an unresolved challenge due to the limited network bandwidth and stringent latency constraints. In this paper, we for the first time propose a holistic live volumetric video streaming system, LiveVV, which achieves multi-view capture, scene segmentation \& reuse, adaptive transmission, and rendering. LiveVV contains multiple lightweight volumetric video capture modules that are capable of being deployed without prior preparation. To reduce bandwidth consumption, LiveVV processes static and dynamic volumetric content separately by reusing static data with low disparity and decimating data with low visual saliency. Besides, to deal with network fluctuation, LiveVV integrates a volumetric video adaptive bitrate streaming algorithm (VABR) to enable fluent playback with the maximum quality of experience. Extensive real-world experiment shows that LiveVV can achieve live volumetric video streaming at a frame rate of 24 fps with a latency of less than 350ms

The results showed that PCR amplification yield and specificity were significantly promoted by the addition of EcoExoIIIM.     The eukaryotic GINS, Cdc45, and minichromosome maintenance proteins form an essential complex that moves with the DNA replication fork. The GINS protein complex has also been reported to associate with DNA polymerase. In archaea, the third domain of life, DNA polymerase D (PolD) is essential for DNA replication, and the genes encoding PolDs exist only in the genomes of archaea. The archaeal GAN (GINS-associated nuclease) is believed to be a homolog of the eukaryotic Cdc45. In this study, we found that the Thermococcus sp. 4557 DP1 (small subunit of PolD) interacted with GINS15 in vitro, and the 3’-5’ exonuclease activity of DP1 was inhibited by GINS15. We also demonstrated that the GAN, GINS15, and DP1 proteins interact to form a complex adapting a GAN-GINS15-DP1 order.     Genetic Studies of Leptin Concentrations Implicate Leptin in the Regulation of Early Adiposity.

    Leptin influences food intake by informing the brain about the status of body fat stores. Rare LEP mutations associated with congenital leptin deficiency cause severe early-onset obesity that can be mitigated by administering leptin. However, the role of genetic regulation of leptin in polygenic obesity remains poorly understood. We performed an exome-based analysis in up to 57,232 individuals of diverse ancestries to identify genetic variants that influence adiposity-adjusted leptin concentrations. We identify five novel variants, including four missense variants, in LEP, ZNF800, KLHL31, and ACTL9, and one intergenic variant near KLF14. The missense variant Val94Met (rs17151919) in LEP was common in individuals of African ancestry only, and its association with lower leptin concentrations was specific to this ancestry (P = 2 × 10-16, n = 3,901). Using in vitro analyses, we show that the Met94 allele decreases leptin secretion. We also show that the Met94 allele is associated with higher BMI in young African-ancestry children but not in adults, suggesting that leptin regulates early adiposity

    Fabrication of thick anodic films on 304-type stainless steel for binder-free supercapacitors

    We report the fabrication of thick nanoporous oxide films on stainless steel (SS) foils via long-term constant-potential anodization at room temperature. The anodization potential of 50 V was employed for 1, 2 and 3 h, respectively to form the anodic films with different thickness. The effects of anodic film thickness on supercapacitance behavior have been further investigated as binder-free electrodes. It is found that the anodic film thickness increases linearly with the anodization time while the electrochemical behaviors of anodic film were not proportionally enhanced with the thickness. Furthermore, the formed anodic layers formed at 50 V/3 h exhibits the highest area capacitance ~ 215 mF cm-2 at 1 mA cm-2, and the capacitance still retained 58.9% of its initial value after 1000 cycles

    Sirolimus in the treatment of kaposiform lymphangiomatosis

    Abstract Background Kaposiform lymphangiomatosis (KLA), which is a new subtype of generalized lymphatic anomaly, is a rare disease with a poor prognosis. Currently, there is no standard treatment due to the poor understanding of KLA. Sirolimus, which is an inhibitor of mammalian target of rapamycin, has been shown to have promising potential in the treatment of complicated vascular anomalies. The aim of this study was to introduce the use of sirolimus for the treatment of KLA and to highlight the challenges of managing this refractory disease. Results We reported seven patients with KLA who received sirolimus therapy in our center. Combined with previously reported cases, 58.3% achieved a partial response, 25.0% had stable disease, and 16.7% experienced disease progression. No severe sirolimus-related adverse events occurred during treatment. Conclusions This study suggests that sirolimus is currently an option for the treatment of KLA, and it is hoped that more specific therapies will be developed in the future. Rapid advances in basic science and clinical practice may facilitate the development of important new treatments for KLA

    Difference expansion based reversible data hiding using two embedding directions

    Current difference-expansion (DE) embedding techniques perform one layer embedding in a difference image. They do not turn to the next difference image for another layer embedding unless the current difference image has no expandable differences left. The obvious disadvantage of these techniques is that image quality may have been severely degraded even before the later layer embedding begins because the previous layer embedding has used up all expandable differences, including those with large magnitude. Based on integer Haar wavelet transform, we propose a new DE embedding algorithm, which utilizes the horizontal as well as vertical difference images for data hiding. We introduce a dynamical expandable difference search and selection mechanism. This mechanism gives even chances to small differences in two difference images and effectively avoids the situation that the largest differences in the first difference image are used up while there is almost no chance to embed in small differences of the second difference image. We also present an improved histogram-based difference selection and shifting scheme, which refines our algorithm and makes it resilient to different types of images. Compared with current algorithms, the proposed algorithm often has better embedding capacity versus image quality performance. The advantage of our algorithm is more obvious near the embedding rate of 0.5 bpp

    Learning curve of laparoscopic Kasai portoenterostomy for biliary atresia: report of 100 cases

    Abstract Background Laparoscopic Kasai portoenterostomy (LKPE) is performed for biliary atresia (BA). As LKPE is a technically demanding operation, a learning curve should be defined to guide training. The aim of this study was to identify the learning curve of LKPE for BA. Methods Metrics of perioperative safety and efficiency for 100 cases of LKPE were evaluated. Outcomes were followed to 67.2 ± 12.1 months. Cumulative sum (CUSUM) analysis was used to identify inflexion point corresponding to the learning curve. Outcome measures included operative time (ORT), rate of clearance of jaundice (CJ) and survival with native liver (SNL). Results Between May 2009 and May 2013, 100 consecutive patients with BA underwent LKPE. The rate of conversion from LKPE to open Kasai portoenterostomy (OKPE), intraoperative transfusion and any perioperative complications was 11, 26 and 16%, respectively. There was no perioperative mortality. The CUSUM analysis revealed a learning curve of 50 for LKPE. Precipitous ORT reductions from an initial mean operative time of 316.3 min that was observed in the first 50 to 232.2 min of the late 50 cases (P 0.05). Conclusions In this experience, improved perioperative and postoperative parameters for LKPE were observed in the last 50 patients when compared with the first 50 patients. The dedicated training is likely to contribute to significantly shorter learning curves in future adopters
