    Pd/Cu Site Interchange and Non-Fermi-Liquid Behavior in UCu_4Pd

    X-ray-absorption fine-structure measurements of the local structure in UCu_4Pd are described which indicate a probable lattice-disorder origin for non-Fermi-liquid behavior in this material. Short Pd-Cu distances are observed, consistent with 24 +/- 3% of the Pd atoms occupying nominally Cu sites. A "Kondo disorder" model, based on the effect on the local Kondo temperature T_K of this interchange and some additional bond-length disorder, agrees quantitatively with previous experimental susceptibility data, and therefore also with specific heat and magnetic resonance experiments.Comment: 4 pages, 3 PostScript figures, to be published in PR

    Transition Analysis for the HIFiRE-5 Vehicle

    The Hypersonic International Flight Research and Experimentation (HIFiRE) 5 flight experiment by Air Force Research Laboratories and Australian Defense Science and Technology Organization is designed to provide in-flight boundary-layer transition data for a canonical 3D configuration at hypersonic Mach numbers. This paper outlines the progress, to date, on boundary layer stability analysis for the HIFiRE-5 flight configuration, as well as for selected test conditions from the wind tunnel experiments supporting the flight test. At flow conditions corresponding to the end of the test window, rather large values of linear amplification factor are predicted for both second mode (N>40) and crossflow (N>20) instabilities, strongly supporting the feasibility of first in-flight measurements of natural transition on a fully three-dimensional hypersonic configuration. Additional results highlight the rich mixture of instability mechanisms relevant to a large segment of the flight trajectory, as well as the effects of angle of attack and yaw angle on the predicted transition fronts for ground facility experiments at Mach 6

    Ubiquitinylation of α-Synuclein by Carboxyl Terminus Hsp70-Interacting Protein (CHIP) Is Regulated by Bcl-2-Associated Athanogene 5 (BAG5)

    Parkinson's disease (PD) is a common neurodegenerative condition in which abnormalities in protein homeostasis, or proteostasis, may lead to accumulation of the protein α-synuclein (α-syn). Mutations within or multiplications of the gene encoding α-syn are known to cause genetic forms of PD and polymorphisms in the gene are recently established risk factors for idiopathic PD. α-syn is a major component of Lewy bodies, the intracellular proteinaceous inclusions which are pathological hallmarks of most forms of PD. Recent evidence demonstrates that α-syn can self associate into soluble oligomeric species and implicates these α-syn oligomers in cell death. We have previously shown that carboxyl terminus of Hsp70-interacting protein (CHIP), a co-chaperone molecule with E3 ubiquitin ligase activity, may reduce the levels of toxic α-syn oligomers. Here we demonstrate that α-syn is ubiquitinylated by CHIP both in vitro and in cells. We find that the products from ubiquitinylation by CHIP include both monoubiquitinylated and polyubiquitinylated forms of α-syn. We also demonstrate that CHIP and α-syn exist within a protein complex with the co-chaperone bcl-2-associated athanogene 5 (BAG5) in brain. The interaction of CHIP with BAG5 is mediated by Hsp70 which binds to the tetratricopeptide repeat domain of CHIP and the BAG domains of BAG5. The Hsp70-mediated association of BAG5 with CHIP results in inhibition of CHIP E3 ubiquitin ligase activity and subsequently reduces α-syn ubiquitinylation. Furthermore, we use a luciferase-based protein-fragment complementation assay of α-syn oligomerization to investigate regulation of α-syn oligomers by CHIP in living cells. We demonstrate that BAG5 mitigates the ability of CHIP to reduce α-syn oligomerization and that non-ubiquitinylated α-syn has an increased propensity for oligomerization. Thus, our results identify CHIP as an E3 ubiquitin ligase of α-syn and suggest a novel function for BAG5 as a modulator of CHIP E3 ubiquitin ligase activity with implications for CHIP-mediated regulation of α-syn oligomerization

    Fragmentation of a Circular Disc by Impact on a Frictionless Plate

    The break-up of a two-dimensional circular disc by normal and oblique impact on a hard frictionless plate is investigated by molecular dynamics simulations. The disc is composed of numerous unbreakable randomly shaped convex polygons connected together by simple elastic beams that break when bent or stretched beyond a certain limit. It is found that for both normal and oblique impacts the crack patterns are the same and depend solely on the normal component of the impact velocity. Analysing the pattern of breakage, amount of damage, fragment masses and velocities, we show the existence of a critical velocity which separates two regimes of the impact process: below the critical point only a damage cone is formed at the impact site (damage), cleaving of the particle occurs at the critical point, while above the critical velocity the disc breaks into several pieces (fragmentation). In the limit of very high impact velocities the disc suffers complete disintegration (shattering) into many small fragments. In agreement with experimental results, fragment masses are found to follow the Gates-Gaudin-Schuhmann distribution (power law) with an exponent independent of the velocity and angle of impact. The velocity distribution of fragments exhibit an interesting anomalous scaling behavior when changing the impact velocity and the size of the disc.Comment: submitted to J.     Branching Ratio and CP Violation of B to pi pi Decays in Perturbative QCD Approach

    We calculate the branching ratios and CP asymmetries for B^0 to pi^+pi^-, B^+ to pi^+pi^0 and B^0 to pi^0pi^0 decays, in a perturbative QCD approach. In this approach, we calculate non-factorizable and annihilation type contributions, in addition to the usual factorizable contributions. We found that the annihilation diagram contributions are not very small as previous argument. Our result is in agreement with the measured branching ratio of B to pi^+pi^- by CLEO collaboration. With a non-negligible contribution from annihilation diagrams and a large strong phase, we predict a large direct CP asymmetry in B^0 to pi^+pi^-, and pi^0pi^0, which can be tested by the current running B factories.Comment: Latex, 28 pages including 11 figures; added contents and figures, corrected typo

    The CIPAZ study protocol: an open label randomised controlled trial of azithromycin versus ciprofloxacin for the treatment of children hospitalised with dysentery in Ho Chi Minh City, Vietnam

    Background: Diarrhoeal disease remains a common cause of illness and death in children <5 years of age. Faecal-oral infection by Shigella spp. causing bacillary dysentery is a leading cause of moderate-to-severe diarrhoea, particularly in low and middle-income countries. In Southeast Asia, S. sonnei predominates and infections are frequently resistant to first-line treatment with the fluoroquinolone, ciprofloxacin. While resistance to all antimicrobials is increasing, there may be theoretical and clinical benefits to prioritizing treatment of bacillary dysentery with the azalide, azithromycin. In this study we aim to measure the efficacy of treatment with azithromycin compared with ciprofloxacin, the current standard of care, for the treatment of children with bacillary dysentery. Methods and analysis: We will perform a multicentre, open-label, randomized controlled trial of two therapeutic options for the antimicrobial treatment of children hospitalised with dysentery. Children (6–60 months of age) presenting with symptoms and signs of dysentery at Children’s Hospital 2 in Ho Chi Minh City will be randomised (1:1) to treatment with either oral ciprofloxacin (15mg/kg/twice daily for 3 days, standard-of-care) or oral azithromycin (10mg/kg/daily for 3 days). The primary endpoint will be the proportion of treatment failure (defined by clinical and microbiological parameters) by day 28 (+3 days) and will be compared between study arms by logistic regression modelling using treatment allocation as the main variable. Ethics and dissemination: The study protocol (version 1.2 dated 27th December 2018) has been approved by the Oxford Tropical Research Ethics Committee (47–18) and the ethical review boards of Children's Hospital 2 (1341/NĐ2-CĐT). The study has also been approved by the Vietnamese Ministry of Health (5044/QĐ-BYT). Trial registration: Clinicaltrials.gov: NCT03854929 (February 26th 2019)
