Cardiomyopathy Following Latrodectus Envenomation
Latrodectus envenomations are common throughout the United States and the world. While many envenomations can result in catecholamine release with resultant hypertension and tachycardia, myocarditis is very rare. Transitioning to rural practice together: a rural fellowship model (in 6 Ps).
PURPOSE: Maintaining a robust healthcare workforce in underserved rural communities continues to be a challenge. To better meet healthcare needs in rural areas, training programs must develop innovative ways to foster transition to, and integration into, these communities. Mountain Area Health Education Center designed and implemented a 12-month post-residency Rural Fellowship program to enhance placement, transition, and retention in rural North Carolina. Utilizing a '6 Ps' framework, the program targeted physicians and pharmacists completing residency with the purpose of recruiting and supporting their transition into the first year of rural practice.
METHOD: To better understand Rural Fellows' experiences and the immediate impact of their Fellowship year, we conducted a semi-structured interview using a narrative technique and evaluated retention rates over time. Interviews with the eight participants, which included Fellowship alumni and current Fellows, demonstrated the impact and influence of the key curricular '6 Ps' framework.
RESULTS: An early retention rate of 100% and a long-term retention rate of 87%, combined with expressed clarity of curricular knowledge, skills, and attitudes related to the '6 Ps', demonstrate the potential and effectiveness of this Rural Fellowship model. Participants indicated the Rural Fellowship experience supports the transition to rural practice communities and expands their clinical skills.
CONCLUSION: The Rural Fellowship program demonstrates an effective model to support early career healthcare providers as they begin practice in rural communities in western North Carolina through academic opportunities, personal growth, and professional development. Implementation of this model has demonstrated the success of a rural retention model over a 6-year period. This model has the potential to target an array of clinical providers and disciplines. We started with family medicine and have expanded to psychiatry, obstetrics, pharmacy, and nursing. This study demonstrated that this model supports clinical providers during the critical transition period from residency to practice. Targeting the most important stage of one's medical training, the commencement of professional practice, this is a scalable model for other rural-based health professions education sites where rural recruitment and retention remain a problem
Probing the Security of DNA Origami
DNA origami is a method used to program the self-assembly of nanoscale computational systems. The contribution of this paper is to probe the security of a canonical DNA origami that is often reused. The paper describes the techniques we used to design probes whose input targets the origami systems’ integrity. Experimental results show that the probes successfully caused up to a third of the systems to fail. Moreover, our approach identified a single, small input whose addition was able to cause the failure of a quarter of the systems. Extending such probes to other DNA origami systems may aid in understanding how to enhance their reliability
Potential of herbariomics for studying repetitive DNA in angiosperms
Repetitive DNA has an important role in angiosperm genomes and is relevant to our understanding of genome size variation, polyploidisation and genome dynamics more broadly. Much recent work has harnessed the power of high-throughput sequencing (HTS) technologies to advance the study of repetitive DNA in flowering plants. Herbarium collections provide a useful historical perspective on genome diversity through time, but their value for the study of repetitive DNA has not yet been explored. We propose that herbarium DNA may prove as useful for studies of repetitive DNA content as it has for reconstructed organellar genomes and low-copy nuclear sequence data. Here we present a case study in the tobacco genus (Nicotiana; Solanaceae), showing that herbarium specimens can provide accurate estimates of the repetitive content of angiosperm genomes by direct comparison with recently-collected material. We show a strong correlation between the abundance of repeat clusters, e.g., different types of transposable elements and satellite DNA, in herbarium collections versus recent material for four sets of Nicotiana taxa. These results suggest that herbarium specimen genome sequencing (herbariomics) holds promise for both repeat discovery and analyses that aim to investigate the role of repetitive DNAs in genomic evolution, particularly genome size evolution and/or contributions of repeats to the regulation of gene space
In the Round: The Future of Food
In collaboration with PopTech, the Center for Complexity (CfC) hosted a two day convening in March 2019 to discuss the future of food, conducted in-the-round. Conversations-in-the-round offer an experimental and collaborative forum where a variety of perspectives collide to reimagine how we should approach navigating complex challenges.
30 professionals, community leaders, ag-tech startups, policymakers, local activists, global aid administrators, and many other important voices in the food industry were in attendance. We did not set out to create solutions, rather we devoted our time to exploration — a process that allowed us to assess, discover, reality-check, and strategically plan the next phase of the journey. A publication was created to capture the discussions, workshops, proposals, deign principles and actionable next steps.
The outcomes of this first convening will guide what comes next. Our aim is to initiate a series of projects conducted by the CfC and our partners to advance the future of food according to shared design principles. Collaboration for conservation: assessing countrywide carnivore occupancy dynamics from sparse data
Aim: Assessing the distribution and persistence of species across their range is a crucial component of wildlife conservation. It demands data at adequate spatial scales and over extended periods of time, which may only be obtained through collaborative efforts, and the development of methods that integrate heterogeneous datasets. We aimed to combine existing data on large carnivores to evaluate population dynamics and improve knowledge on their distribution nationwide.
Location: Botswana.
Methods: Between 2010 and 2016, we collated data on African wild dog, cheetah, leopard, brown and spotted hyaena and lion gathered with different survey methods by independent researchers across Botswana. We used a multi-species, multi-method dynamic occupancy model to analyse factors influencing occupancy, persistence and colonization, while accounting for imperfect detection. Lastly, we used the gained knowledge to predict the probability of occurrence of each species countrywide.
Results: Wildlife areas and communal rangelands had similar occupancy probabilities for most species. Large carnivore occupancy was low in commercial farming areas and where livestock density was high, except for brown hyaena. Lion occupancy was negatively associated with human density; lion and spotted hyaena occupancy was high where rainfall was high, while the opposite applied to brown hyaena. Lion and leopard occupancy remained constant countrywide over the study period. African wild dog and cheetah occupancy declined over time in the south and north, respectively, whereas both hyaena species expanded their ranges. Countrywide predictions identified the highest occupancy for leopards and lowest for the two hyaena species.
Main Conclusions: We highlight the necessity of data sharing and propose a generalizable analytical method that addresses the challenges of heterogeneous data common in ecology. Our approach, which enables a comprehensive multi-species assessment at large spatial and temporal scales, supports the development of data-driven conservation guidelines and the implementation of evidence-based management strategies nationally and internationally
D1 Dopamine Receptor Signaling Is Modulated by the R7 RGS Protein EAT-16 and the R7 Binding Protein RSBP-1 in Caenoerhabditis elegans Motor Neurons
Dopamine signaling modulates voluntary movement and reward-driven behaviors by acting through G protein-coupled receptors in striatal neurons, and defects in dopamine signaling underlie Parkinson's disease and drug addiction. Despite the importance of understanding how dopamine modifies the activity of striatal neurons to control basal ganglia output, the molecular mechanisms that control dopamine signaling remain largely unclear. Dopamine signaling also controls locomotion behavior in Caenorhabditis elegans. To better understand how dopamine acts in the brain we performed a large-scale dsRNA interference screen in C. elegans for genes required for endogenous dopamine signaling and identified six genes (eat-16, rsbp-1, unc-43, flp-1, grk-1, and cat-1) required for dopamine-mediated behavior. We then used a combination of mutant analysis and cell-specific transgenic rescue experiments to investigate the functional interaction between the proteins encoded by two of these genes, eat-16 and rsbp-1, within single cell types and to examine their role in the modulation of dopamine receptor signaling. We found that EAT-16 and RSBP-1 act together to modulate dopamine signaling and that while they are coexpressed with both D1-like and D2-like dopamine receptors, they do not modulate D2 receptor signaling. Instead, EAT-16 and RSBP-1 act together to selectively inhibit D1 dopamine receptor signaling in cholinergic motor neurons to modulate locomotion behavior

Genetic Discontinuities among Populations of \u3ci\u3eCleistes\u3c/i\u3e (Orchidaceae, Vanilloideae) in North America
Genetic patterns were examined in five populations of North American Cleistes with three sources of molecular data: amplified fragment length polymorphisms (AFLP), DNA sequencing, and plastid microsatellites. Populations of C. bifaria were sampled in four areas of the south-eastern US: the coastal plains of Florida and North Carolina and the mountains of North Carolina and West Virginia. A population of C. divaricata sympatric with the North Carolina coastal plain C. bifaria was also sampled. Analysis of the three types of molecular data resulted in the same relationships among the five sampled populations. The coastal plain population of C. divaricata was consistently grouped with the C. bifaria populations from the mountains of West Virginia and North Carolina, and the two coastal plain populations of C. bifaria formed a separate group, results not supporting the existing concepts of species relationships. For future studies, greater sampling of C. divaricata populations and more detailed morphological and phenological studies are recommended for better characterization of the diversity within North American Cleistes
