A Systematic Study on Energy Dependence of Quasi-Periodic Oscillation Frequency in GRS 1915+105
Systematically studying all the RXTE/PCA observations for GRS 1915+105 before
November 2010, we have discovered three additional patterns in the relation
between Quasi-Periodic Oscillation (QPO) frequency and photon energy, extending
earlier outcomes reported by Qu et al. (2010). We have confirmed that as QPO
frequency increases, the relation evolves from the negative correlation to
positive one. The newly discovered patterns provide new constraints on the QPO
models
New insights into the genetic etiology of Alzheimer's disease and related dementias
Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele
Multiancestry analysis of the HLA locus in Alzheimer’s and Parkinson’s diseases uncovers a shared adaptive immune response mediated by HLA-DRB1*04 subtypes
Across multiancestry groups, we analyzed Human Leukocyte Antigen (HLA) associations in over 176,000 individuals with Parkinson’s disease (PD) and Alzheimer’s disease (AD) versus controls. We demonstrate that the two diseases share the same protective association at the HLA locus. HLA-specific fine-mapping showed that hierarchical protective effects of HLA-DRB1*04 subtypes best accounted for the association, strongest with HLA-DRB1*04:04 and HLA-DRB1*04:07, and intermediary with HLA-DRB1*04:01 and HLA-DRB1*04:03. The same signal was associated with decreased neurofibrillary tangles in postmortem brains and was associated with reduced tau levels in cerebrospinal fluid and to a lower extent with increased Aβ42. Protective HLA-DRB1*04 subtypes strongly bound the aggregation-prone tau PHF6 sequence, however only when acetylated at a lysine (K311), a common posttranslational modification central to tau aggregation. An HLA-DRB1*04-mediated adaptive immune response decreases PD and AD risks, potentially by acting against tau, offering the possibility of therapeutic avenues
Evaluation of the DayCent model to predict carbon fluxes in French crop sites [résumé]
EGU.European Geosciences Union General Assembly, Vienne, AUT, 23-/04/2017 - 28/04/2017Croplands in temperate regions are an important component of the carbon balance and can act as a sink or a source of carbon, depending on pedoclimatic conditions and management practices. Therefore the evaluation of carbon fluxes in croplands by modelling approach is relevant in the context of global change. This study was part of the Comete-Global project funded by the multi-Partner call FACCE JPI. Carbon fluxes, net ecosystem exchange (NEE), leaf area index (LAI), biomass, and grain production were simulated at the site level in three French crop experiments from the CarboEurope project. Several crops were studied, like winter wheat, rapeseed, barley, maize, and sunflower. Daily NEE was measured with eddy covariance and could be partitioned between gross primary production (GPP) and total ecosystem respiration (TER). Measurements were compared to DayCent simulations, a process-based model predicting plant production and soil organic matter turnover at daily time step. We compared two versions of the model: the original one with a simplified plant module and a newer version that simulates LAI. Input data for modelling were soil properties, climate, and management practices. Simulations of grain yields and biomass production were acceptable when using optimized crop parameters. Simulation of NEE was also acceptable. GPP predictions were improved with the newer version of the model, eliminating temporal shifts that could be observed with the original model. TER was underestimated by the model. Predicted NEE was more sensitive to soil tillage and nitrogen applications than measured NEE. DayCent was therefore a relevant tool to predict carbon fluxes in French crops at the site level. The introduction of LAI in the model improved its performance
Outbreaks, gene flow and effective population size in the migratory locust, Locusta migratoria : a regional-scale comparative survey
The potential effect of population outbreaks on within and between genetic variation of populations in pest species has rarely been assessed. In this study, we compare patterns of genetic variation in different sets of historically frequently outbreaking and rarely outbreaking populations of an agricultural pest of major importance, the migratory locust, Locusta migratoria. We analyse genetic variation within and between 24 populations at 14 microsatellites in Western Europe, where only ancient and low-intensity outbreaks have been reported (non-outbreaking populations), and in Madagascar and Northern China, where frequent and intense outbreak events have been recorded over the last century (outbreaking populations). Our comparative survey shows that (i) the long-term effective population size is similar in outbreaking and non-outbreaking populations, as evidenced by similar estimates of genetic diversity, and (ii) gene flow is substantially larger among outbreaking populations than among non-outbreaking populations, as evidenced by a fourfold to 30-fold difference in F-ST values. Genetic variation for parental effects on the propensity to gregarise in Locusta migratoria - art. no. 37
Background: Environmental parental effects can have important ecological and evolutionary consequences, yet little is known about genetic variation among populations in the plastic responses of offspring phenotypes to parental environmental conditions. This type of variation may lead to rapid phenotypic divergence among populations and facilitate speciation. With respect to density- dependent phenotypic plasticity, locust species ( Orthoptera: family Acrididae), exhibit spectacular developmental and behavioural shifts in response to population density, called phase change. Given the significance of phase change in locust outbreaks and control, its triggering processes have been widely investigated. Whereas crowding within the lifetime of both offspring and parents has emerged as a primary causal factor of phase change, less is known about intraspecific genetic variation in the expression of phase change, and in particular in response to the parental environment. We conducted a laboratory experiment that explicitly controlled for the environmental effects of parental rearing density. This design enabled us to compare the parental effects on offspring expression of phase- related traits between two naturally- occurring, genetically distinct populations of Locusta migratoria that differed in their historical patterns of high population density outbreak events. Results: We found that locusts from a historically outbreaking population of L. migratoria expressed parentally- inherited density- dependent phase changes to a greater degree than those from a historically non- outbreaking population. Conclusion: Because locusts from both populations were raised in a common environment during our experiment, a genetically- based process must be responsible for the observed variation in the propensity to express phase change. This result emphasizes the importance of genetic factors in the expression of phase traits and calls for further investigations on density- dependent parental effects in locust phase change. More population samples with different outbreak histories need to be analyzed to demonstrate that differences in propensity to gregarise evolve because of different outbreak histories

Fluconazole prophylaxis prevents intra-abdominal candidiasis in high-risk surgical patients.
OBJECTIVE: To evaluate the efficacy and safety of intravenous fluconazole for the prevention of intra-abdominal Candida infections in high-risk surgical patients. DESIGN: Randomized, prospective, double-blind, placebo-controlled study. SETTING: Two university-affiliated hospitals in Switzerland. PATIENTS: Forty-nine surgical patients with recurrent gastrointestinal perforations or anastomotic leakages. INTERVENTIONS: Prophylaxis with intravenous fluconazole (400 mg per day) or placebo continued until resolution of the underlying surgical condition. MEASUREMENTS AND MAIN RESULTS: Patients were evaluated daily, and specimens for culture were obtained three times per week during prophylaxis. The primary study end points were the frequency of and the time to intra-abdominal Candida infections. Secondary end points were the frequency of candidiasis (intra-abdominal and extra-abdominal) and the emergence or persistence of Candida colonization. Among patients who were not colonized at study entry, Candida was isolated from surveillance cultures during prophylaxis in 15% of the patients in the fluconazole group and in 62% of the patients in the placebo group (relative risk, 0.25; 95% confidence interval, 0.07 to 0.96; p = .04). Candida peritonitis occurred in one of 23 patients (4%) who received fluconazole and in seven of 20 patients (35%) who received placebo (relative risk, 0.12; 95% confidence interval, 0.02 to 0.93; p = .02). In addition, one catheter-related Candida albicans sepsis occurred in a fluconazole-treated patient. Thus, overall, candidiasis developed in two fluconazole patients and seven placebo patients (relative risk, 0.25; 95% confidence interval, 0.06 to 1.06; p = .06). C. albicans accounted for 87% of the Candida species isolated before or during prophylaxis, and all C. albicans strains were susceptible to fluconazole. Fluconazole was well tolerated, and adverse events occurred at similar frequencies in both treatment groups. CONCLUSIONS: Fluconazole prophylaxis prevents colonization and invasive intra-abdominal Candida infections in high-risk surgical patients
