    The absorption and uptake of recombinant human follicle-stimulating hormone through vaginal subcutaneous injections - a pharmacokinetic study

    <p>Abstract</p> <p>Background</p> <p>Follicle stimulating hormone (FSH) has been routinely used for ovulation induction. Because of rapid clearance of the hormone, FSH is commonly administered by daily intramuscular or subcutaneous injections in in-vitro fertilization (IVF). To reduce the number of visits to the clinic, an intermittent vaginal injection of rhFSH every 3 days employing the concepts of mesotherapy and uterine first-pass effect was invented and has successfully been applied in women receiving IVF treatment. This study was designed to monitor the pharmacokinetic pattern of rhFSH administered vaginally.</p> <p>Methods</p> <p>Twelve healthy women with regular ovulatory cycles were recruited. All volunteers received gonadotrophin-releasing hormone agonist to suppress pituitary function and were assigned to receive single dose recombinant human FSH (rhFSH, Puregon 300) either using conventional abdominal subcutaneous injection or vaginal subcutaneous injection in a randomized cross-over study. Serum samples were collected at pre- scheduled time intervals after injections of rhFSH to determine immunoreactive FSH levels. Pharmacokinetic parameters characterizing rate [maximal plasma concentrations (Cmax) and time of maximal plasma concentrations (tmax)] and extent [area under the plasma concentration-time curve (AUC) and clearance] of absorption of rhFSH were compared.</p> <p>Results</p> <p>Vaginal injection of rhFSH was well tolerated and no drug-related adverse reaction was noted. Our analysis revealed that tmax was significantly earlier (mean 6.67 versus 13.33 hours) and Cmax was significantly higher (mean 17.77 versus 13.96 IU/L) in vaginal versus abdominal injections. The AUC<sub>0-∞</sub> was 1640 versus 1134 IU·hour/L in vaginal and abdominal injections, respectively. Smaller plasma elimination rate constant (0.011 versus 0.016 hour-1), longer mean residence time (106.58 versus 70.47 hours), and slower total body clearance (292.2 versus 400.1 mL/hour) were also found in vaginal injection.</p> <p>Conclusion</p> <p>The vaginal injection mode elicited a rapid and highly extended absorption of rhFSH injected compared to conventional abdominal injection. These data indicate that the rate and extent of FSH absorption from the injection site can vary depending on the route of the FSH administration.</p

    Anti-inflammatory and anti-coagulatory activities of caffeic acid and ellagic acid in cardiac tissue of diabetic mice

    <p>Abstract</p> <p>Background</p> <p>Caffeic acid (CA) and ellagic acid (EA) are phenolic acids naturally occurring in many plant foods. Cardiac protective effects of these compounds against dyslipidemia, hypercoagulability, oxidative stress and inflammation in diabetic mice were examined.</p> <p>Methods</p> <p>Diabetic mice were divided into three groups (15 mice per group): diabetic mice with normal diet, 2% CA treatment, or 2% EA treatment. One group of non-diabetic mice with normal diet was used for comparison. After 12 weeks supplement, mice were sacrificed, and the variation of biomarkers for hypercoagulability, oxidative stress and inflammation in cardiac tissue of diabetic mice were measured.</p> <p>Results</p> <p>The intake of CA or EA significantly increased cardiac content of these compounds, alleviated body weight loss, elevated plasma insulin and decreased plasma glucose levels in diabetic mice (<it>p </it>< 0.05). These treatments also significantly enhanced plasma antithrombin-III and protein C activities (<it>p </it>< 0.05); and decreased triglyceride content in cardiac tissue and plasma (<it>p </it>< 0.05), in which the hypolipidemic effects of EA were significantly greater than that of CA (<it>p </it>< 0.05). CA or EA significantly lowered cardiac levels of malondialdehyde, reactive oxygen species, interleukin (IL)-beta, IL-6, tumor necrosis factor (TNF)-alpha and monocyte chemoattractant protein (MCP)-1 (<it>p </it>< 0.05); and retained cardiac activity of glutathione peroxidase (GPX), superoxide dismutase (SOD) and catalase (<it>p </it>< 0.05). These compounds also significantly up-regulated cardiac mRNA expression of GPX1, SOD and catalase; and down-regulated IL-1beta, IL-6, TNF-alpha and MCP-1 mRNA expression in diabetic mice (<it>p </it>< 0.05).</p> <p>Conclusion</p> <p>These results support that CA and EA could provide triglyceride-lowering, anti-coagulatory, anti-oxidative, and anti-inflammatory protection in cardiac tissue of diabetic mice. Thus, the supplement of these agents might be helpful for the prevention or attenuation of diabetic cardiomyopathy.</p

    Hydrodynamical Simulations of the Barred Spiral Galaxy NGC 1097

    NGC 1097 is a nearby barred spiral galaxy believed to be interacting with the elliptical galaxy NGC 1097A located to its northwest. It hosts a Seyfert 1 nucleus surrounded by a circumnuclear starburst ring. Two straight dust lanes connected to the ring extend almost continuously out to the bar. The other ends of the dust lanes attach to two main spiral arms. To provide a physical understanding of its structural and kinematical properties, two-dimensional hydrodynamical simulations have been carried out. Numerical calculations reveal that many features of the gas morphology and kinematics can be reproduced provided that the gas flow is governed by a gravitational potential associated with a slowly rotating strong bar. By including the self-gravity of the gas disk in our calculation, we have found the starburst ring to be gravitationally unstable which is consistent with the observation in \citet{hsieh11}. Our simulations show that the gas inflow rate is 0.17 M_\sun yr−1^{-1} into the region within the starburst ring even after its formation, leading to the coexistence of both a nuclear ring and a circumnuclear disk.Comment: 32 pages, 14 figures, 1 table, accepted for publication in the Ap

    Face recognition using nonparametric-weighted Fisherfaces

    This study presents an appearance-based face recognition scheme called the nonparametric-weighted Fisherfaces (NW-Fisherfaces). Pixels in a facial image are considered as coordinates in a high-dimensional space and are transformed into a face subspace for analysis by using nonparametric-weighted feature extraction (NWFE). According to previous studies of hyperspectral image classification, NWFE is a powerful tool for extracting hyperspectral image features. The Fisherfaces method maximizes the ratio of between-class scatter to that of within-class scatter. In this study, the proposed NW-Fisherfaces weighted the between-class scatter to emphasize the boundary structure of the transformed face subspace and, therefore, enhances the separability for different persons' face. The proposed NW-Fisherfaces was compared with Orthogonal Laplacianfaces, Eigenfaces, Fisherfaces, direct linear discriminant analysis, and null space linear discriminant analysis methods for tests on five facial databases.     A new broadband wide-view circular polarizer is proposed for high transmittance multi-domain vertical-alignment liquid crystal displays (MVA LCDs). This configuration only requires one biaxial plate in the conventional circular polarizer. The optimal film parameters are obtained analytically through spherical trigonometric method on the Poincare sphere and through computer-aided parameter search method.     Isolated pancreatic metastasis from rectal cancer: a case report and review of literature

    Isolated pancreatic metastases from a non-pancreatic primary malignancy are very rare. Studies have shown that resection of metastases is of proven benefit in some types of tumors. We report a case of 76-year-old Taiwanese woman with rectal adenocarcinoma treated with neoadjuvant chemoradiotherapy and abdominoperineal resection 2 years ago presenting with an asymptomatic mass at the pancreatic tail on a routine follow up abdominal computed tomography scan. The patient underwent distal pancreatectomy and splenectomy under the preoperative impression of a primary pancreatic malignancy. Histological examination of the surgical specimen showed metastatic adenocarcinoma. Immunohistochemical studies confirmed the diagnosis of pancreatic metastasis from rectal adenocarcinoma. Postoperative chemotherapy in the form of oral capecitabine was given. The patient is alive and disease free 12 months after the surgery. In a patient presenting with a pancreatic mass with history of a non-pancreatic malignancy, a differential diagnosis of pancreatic metastasis should be considered. Surgical resection of a solitary pancreatic mass is justified not only to get the definitive diagnosis but also to improve the survival

    The latency between audio and video streams of a device is usually measured using stock test videos. Although the use of stock test videos eases analysis, the test video differs materially from real-world videos, which tend to be far more diverse in content and encoding schemes, resulting in laborious experimental setup and inaccurate synchronization. This disclosure describes techniques to measure the latency between the audio and video streams of a given device using arbitrary, real-world, audio-visual footage (test video). Characteristic video and audio frames and their differences in timestamps (characteristic durations) are identified within the test video. The test video is played by the device-under-test while being recorded by a high-precision video camera. Characteristic durations of the recorded footage are determined. The differences in characteristic durations between the test and the recorded videos are statistically analyzed to determine the AV asynchrony of the device-under-test

    Crystallographic,vibrational modes and optical properties data of α-DIPABcrystal

    The Crystallographic data of the α-DIPAB sample was measured using powder X-ray diffraction (PXRD). The crystal structure was also optimized using density functional based method. The calculations were performed both including vander Waals (vdW) interactions and excluding them to quantify the effects of vdW interaction on the crystal formation. The vibrational modes of DIPAB crystal corresponding to the Bromine deficient samples are calculated and presented. These show the origin of drastic change in dielectric response in Br deficient samples as compared to the ideal stoichiometric DIPAB crystal (Alsaad et al. 2018) [4]. Optical properties of an ideal α-DIPAB were calculated using GGA and HSE06 hybrid functional methods implemented in VASP package. We mainly calculated the real and imaginary parts of the frequency-dependent linear dielectric function, as well as the related quantities such as the absorption, reflectivity, energy-loss function, and refractive index of α-DIPAB in the energy window of (0–12) eV
