    Study of interaction of nifedipine with haloperidol on conditioned avoidance response and catalepsy in rats

    Background: To study the interaction of calcium channel blocker (Nifedipine) with Antipsychotic drug (Haloperidol) on Conditioned avoidance Response and catalepsy in Rats.Methods: Every group consisted of 10 healthy albino rats of either sex. Different groups received Nifedipine (5, 10 & 20 mg/kg, i.p.), Haloperidol (ED50 -0.2mg/kg for CAR & 0.4mg/kg for catalepsy) alone and combined doses of both drugs. The Antipsychotic effect of drugs was measured by Conditioned avoidance response (CAR) using Cook’s Pole climbing apparatus and Adverse drug effect (Extra pyramidal syndrome) was measured by Catalepsy.Results: 5 mg/kg i.p. of Nifedipine inhibited CAR in 50 % of Rats (compared to control, p<0.001). 10mg/kg i.p. of Nifedipine inhibited CAR in 60% of Rats (p<0.001) & 20 mg/kg i.p. inhibited CAR in 70% of Rats (p<0.001). When Nifedipine (5 mg/kg i.p) was combined with Haloperidol ED50-0.2mg/kg  the CAR was inhibited in 70% of the rats (p<0.01) and after combining Nifedipine (10mg/kg) with Haloperidol ED50-0.2mg/kg the CAR was inhibited in 80% Rats (p<0.001). Nifedipine at the dose of 5 mg/kg and 10 mg/kg (i.p.) did not induce catalepsy in the rats at any testing time interval.  At 20 mg/kg i.p., it produced catalepsy in 2 rats at half hour and in 4 rats at 1 hour and 2 hour testing interval each (p<0.01). In the dose of 5, 10 and 20 mg/kg, pretreatment with Nifedipine significantly increased Haloperidol induced cataleptic scores at all testing intervals (p<0.05).Conclusions: Nifedipine blocked CAR. Its higher doses induced catalepsy and it is synergistic with haloperidol in blockade of CAR and catalepsy

    Studies on intra-specific variations in the diamondback moth, Plutella xylostella (Lepidoptera: Yponomeutidae) under different geographical regions

    Studies on intraspecific variations in insect pests are an important tool for preparation of management strategy in different geographical regions. In this paper we tried to establish that these variations are also found in the diamondback moth (P. xylostella) populations under north Indian conditions with respect to reproductive biology.The pest populations were collected from five different geographical regions viz. Hisar (800feet), Kangra (2200feet), Solan (4200 feet), Theog (7500feet) and Kinnaur (9000feet) and then reared under laboratory conditions at 25±1oC on cauliflower. The life table analysis revealed that the female from the Kangra population laid maximum eggs (332.16 eggs/female) whereas the number of eggs laid by the female from the Hisar, Solan, Theog and Kinnaur population was 189.53, 207, 252 and 270 eggs/female, respectively. The doubling time (DT) was observed to be the lowest for Kangra (3.12 days) and maximum for the Hisar (4.59 days) population, whereas weekly multiplication rate was minimum for the Solan (2.252) and maximum for the kangra (4.73) population. The true generation time was the lowest for the Kangra (18.54 days) and highest for the Hisar (24.38 days) population. The true intrinsic rate of increase (rm) was found to be maximum (0.222 female progeny/female/day) for the Kangra whereas for the Kinnaur, Theog, Solan and Hisar population it was 0.203, 0.202, 0.182 and 0.151 female progeny/female/day thereby indicating that the Kangra population is more prolific as compared to the population from other geographical regions. The results are indicative of geographical variations among different populations of P. xylostella

    A Modified Approch Audio Stagnography Based On Technique LSB Coding

    Information security is becoming very important part of our life now-a-days. Information hiding is the fundamental of information security. Information hiding can be achieved by steganography as well. This paper presents a modified method of audio stegnography. Audio steganography is the technique of hiding secret information in the audio files. nbspWe have presented a high capacity and high stego-signal quality audio steganography scheme based on samples comparison in DWT domain where selected coefficient of a segment are compared with pre determined threshold value T and based on comparison bits are embedded. The strength of our algorithm is depend on the segment size andnbsp their strength are enabled the algorithm to achieve very high embedding capacity for different data type that can reach up to 25%nbsp from the input audio file size with lest of 35 dBnbsp SNR for the output stego signal. Further we have tabulated the result and the conclusion is made on the basis of the obtained results

    Persistence of neutralizing antibodies to SARS-CoV-2 in first wave infected individuals at ten months post-infection: The UnIRSA cohort study

    Longitudinal mapping of antibody-based SARS-CoV-2 immunity is critical for public health control of the pandemic and vaccine development. We performed a longitudinal analysis of the antibody-based immune response in a cohort of 100 COVID-19 individuals who were infected during the first wave of infection in northern Italy. The SARS-CoV-2 humoral response was tested using the COVID-SeroIndex, Kantaro Quantitative SARS-CoV-2 IgG Antibody RUO Kit (R&amp;D Systems, Bio-Techne, Minneapolis, USA) and pseudotype-based neutralizing antibody assay. Using sequential serum samples collected from 100 COVID-19 recovered individuals from northern Italy—mostly with mild disease—at 2 and 10 months after their first positive PCR test, we show that 93% of them seroconverted at 2 months, with a geometric mean (GeoMean) half-maximal neutralization titer (NT50) of 387.9. Among the 35 unvaccinated subjects retested at 10 months, 7 resulted seronegative, with an 80% drop in seropositivity, while 28 showed decreased anti-receptor binding domain (RBD) and anti-spike (S) IgG titers, with a GeoMean NT50 neutralization titer dropping to 163.5. As an NT50 &gt; 100 is known to confer protection from SARS-CoV-2 re-infection, our data show that the neutralizing activity elicited by the natural infection has lasted for at least 10 months in a large fraction of subjects

    Klebsiella pneumoniae is able to trigger epithelial-mesenchymal transition process in cultured airway epithelial cells

    The ability of some bacterial pathogens to activate Epithelial-Mesenchymal Transition normally is a consequence of the persistence of a local chronic inflammatory response or depends on a direct interaction of the pathogens with the host epithelial cells. In this study we monitored the abilities of the K. pneumoniae to activate the expression of genes related to EMT-like processes and the occurrence of phenotypic changes in airway epithelial cells during the early steps of cell infection. We describe changes in the production of intracellular reactive oxygen species and increased HIF-1α mRNA expression in cells exposed to K. pneumoniae infection. We also describe the upregulation of a set of transcription factors implicated in the EMT processes, such as Twist, Snail and ZEB, indicating that the morphological changes of epithelial cells already appreciable after few hours from the K. pneumoniae infection are tightly regulated by the activation of transcriptional pathways, driving epithelial cells to EMT. These effects appear to be effectively counteracted by resveratrol, an antioxidant that is able to exert a sustained scavenging of the intracellular ROS. This is the first report indicating that strains of K. pneumoniae may promote EMT-like programs through direct interaction with epithelial cells without the involvement of inflammatory cells

    Virological, immunological and pathological findings of transplacentally transmitted bluetongue virus serotype 1 in IFNAR1-blocked mice during early and mid gestation

    © 2020, The Author(s). Transplacental transmission (TPT) of wild-type Indian BTV-1 had never been experimentally proved. This study was first time investigated TPT of Indian BTV-1 (isolated from aborted and stillborn goat fetal spleens). The sequential pathology, virological and immune cell kinetics (CD4+, CD8+ T-lymphocytes and NK cells in spleen and PBMCs), and apoptosis in IFNAR1-blocked pregnant mice during early (infected on 1 GD) and mid (infected on 8 GD) gestation have been studied. There was higher rate of TPT during mid stage (71.43%) than early (57.14%) stage. In early stage reduced implantation sites, early embryonic deaths, abortions, and necro-haemorrhagic lesions had observed. Mid stage, congenital defects and neurological lesions in foetuses like haemorrhages, diffuse cerebral edema, necrotizing encephalitis and decreased bone size (Alizarin red staining) were noticed. BTV-1 antigen was first time demonstrable in cells of mesometrium, decidua of embryos, placenta, uterus, ovary, and brain of foetuses by immunohistochemistry and quantified by real-time qRT-PCR. BTV-inoculated mice were seroconverted by 7 and 5 dpi, and reached peak levels by 15 and 9 dpi in early and mid gestation, respectively. CD4+ and CD8+ cells were significantly decreased (increased ratio) on 7 dpi but subsequently increased on 15 dpi in early gestation. In mid gestation, increased CD8+ cells (decreased ratio) were observed. Apoptotic cells in PBMCs and tissues increased during peak viral load. This first time TPT of wild-type Indian BTV-1 deserves to be reported for implementation of control strategies. This model will be very suitable for further research into mechanisms of TPT, overwintering, and vaccination strategies
