    The effects of goal orientations on knowledge management system usage, knowledge sourcing and learning outcome

    This research investigates individual differences in the context of knowledge management system (KMS) usage, knowledge sourcing and learning outcome. Individual differences are measured for mastery, performance and avoidance goal orientations. Individuals with high mastery goal orientations prefer to master and develop new skills and knowledge. Those with high performance goal orientations prefer to compare their performances with others. Lastly, those with high avoidance goal orientations prefer to avoid a new task if they feel incompetent. User posting and reading activities in a KMS are examined over a 3- month period. Consistent with goal orientation theory, the results show that mastery goal orientation has an effect on reading, knowledge sourcing and learning outcome. In contrast, performance goal orientation has an effect only on posting, but not on reading. To a lesser degree than mastery goal orientation, avoidance goal orientation has an effect on reading, but not posting. These findings have important implications for both knowledge management researchers and practitioners

    The Role of Visualization Tools in Spreadsheet Error Correction from a Cognitive Fit Perspective

    Errors in spreadsheets pose a serious problem for organizations and academics. This has resulted in ongoing efforts to devise measures for reducing errors or efficient ways of correcting them. Visualization tools are often advertised as means for improving spreadsheet error correction performance. This study investigates the role of visualization tools in spreadsheet error correction. For this purpose, this study proposes a framework for classifying activities associated with spreadsheet error correction. The framework is to highlight the activities that are important for correcting different types of spreadsheet errors, and to show how different visualization tools can aid error correction by effectively supporting these activities. By identifying chaining as one of the most important activities from the framework, this study uses cognitive fit theory to examine the effects of a visualization tool that supports chaining on spreadsheet error correction performance. Experimental methodology is used to test the outcome of cognitive fit between the error correction task and the visualization tool. The results of the experiment highlight the importance of cognitive fit between the type of task and the visualization tool for attaining better performance. This study also provides guidelines for designing and developing tools for spreadsheet error correction

    Embodied Persuasion: How Holding Your Smartphone Changes Your Product Perception

    Online shopping through mobile devices has dramatically increased worldwide. This research investigates the role embodied interactions may play in stimulating virtual product experience in mobile commerce settings. Drawing on research on virtual product experience and embodied cognition, we hypothesize that holding a mobile device in hands (vs. putting the mobile device on the table) is more likely to create an illusion that the products being viewed are actually present in the real world and to stimulate imagery consumption experience, leading to higher purchase intention and choice satisfaction. This effect is more salient for desirable products than for undesirable products. We describe an experiment design for testing the hypotheses, report preliminary data analysis results, and discuss the potential theoretical and practical implications of this study

    cAMP/PKA signaling balances respiratory activity with mitochondria dependent apoptosis via transcriptional regulation

    Background Appropriate control of mitochondrial function, morphology and biogenesis are crucial determinants of the general health of eukaryotic cells. It is therefore imperative that we understand the mechanisms that co-ordinate mitochondrial function with environmental signaling systems. The regulation of yeast mitochondrial function in response to nutritional change can be modulated by PKA activity. Unregulated PKA activity can lead to the production of mitochondria that are prone to the production of ROS, and an apoptotic form of cell death. Results We present evidence that mitochondria are sensitive to the level of cAMP/PKA signaling and can respond by modulating levels of respiratory activity or committing to self execution. The inappropriate activation of one of the yeast PKA catalytic subunits, Tpk3p, is sufficient to commit cells to an apoptotic death through transcriptional changes that promote the production of dysfunctional, ROS producing mitochondria. Our data implies that cAMP/PKA regulation of mitochondrial function that promotes apoptosis engages the function of multiple transcription factors, including HAP4, SOK2 and SCO1. Conclusions We propose that in yeast, as is the case in mammalian cells, mitochondrial function and biogenesis are controlled in response to environmental change by the concerted regulation of multiple transcription factors. The visualization of cAMP/TPK3 induced cell death within yeast colonies supports a model that PKA regulation plays a physiological role in coordinating respiratory function and cell death with nutritional status in budding yeast

    The Essentials of Protein Import in the Degenerate Mitochondrion of Entamoeba histolytica

    Several essential biochemical processes are situated in mitochondria. The metabolic transformation of mitochondria in distinct lineages of eukaryotes created proteomes ranging from thousands of proteins to what appear to be a much simpler scenario. In the case of Entamoeba histolytica, tiny mitochondria known as mitosomes have undergone extreme reduction. Only recently a single complete metabolic pathway of sulfate activation has been identified in these organelles. The E. histolytica mitosomes do not produce ATP needed for the sulfate activation pathway and for three molecular chaperones, Cpn60, Cpn10 and mtHsp70. The already characterized ADP/ATP carrier would thus be essential to provide cytosolic ATP for these processes, but how the equilibrium of inorganic phosphate could be maintained was unknown. Finally, how the mitosomal proteins are translocated to the mitosomes had remained unclear. We used a hidden Markov model (HMM) based search of the E. histolytica genome sequence to discover candidate (i) mitosomal phosphate carrier complementing the activity of the ADP/ATP carrier and (ii) membrane-located components of the protein import machinery that includes the outer membrane translocation channel Tom40 and membrane assembly protein Sam50. Using in vitro and in vivo systems we show that E. histolytica contains a minimalist set up of the core import components in order to accommodate a handful of mitosomal proteins. The anaerobic and parasitic lifestyle of E. histolytica has produced one of the simplest known mitochondrial compartments of all eukaryotes. Comparisons with mitochondria of another amoeba, Dictystelium discoideum, emphasize just how dramatic the reduction of the protein import apparatus was after the loss of archetypal mitochondrial functions in the mitosomes of E. histolytica

    The distinctive gastric fluid proteome in gastric cancer reveals a multi-biomarker diagnostic profile

    <p>Abstract</p> <p>Background</p> <p>Overall gastric cancer survival remains poor mainly because there are no reliable methods for identifying highly curable early stage disease. Multi-protein profiling of gastric fluids, obtained from the anatomic site of pathology, could reveal diagnostic proteomic fingerprints.</p> <p>Methods</p> <p>Protein profiles were generated from gastric fluid samples of 19 gastric cancer and 36 benign gastritides patients undergoing elective, clinically-indicated gastroscopy using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry on multiple ProteinChip arrays. Proteomic features were compared by significance analysis of microarray algorithm and two-way hierarchical clustering. A second blinded sample set (24 gastric cancers and 29 clinically benign gastritides) was used for validation.</p> <p>Results</p> <p>By significance analysyis of microarray, 60 proteomic features were up-regulated and 46 were down-regulated in gastric cancer samples (<it>p </it>< 0.01). Multimarker clustering showed two distinctive proteomic profiles independent of age and ethnicity. Eighteen of 19 cancer samples clustered together (sensitivity 95%) while 27/36 of non-cancer samples clustered in a second group. Nine non-cancer samples that clustered with cancer samples included 5 pre-malignant lesions (1 adenomatous polyp and 4 intestinal metaplasia). Validation using a second sample set showed the sensitivity and specificity to be 88% and 93%, respectively. Positive predictive value of the combined data was 0.80. Selected peptide sequencing identified pepsinogen C and pepsin A activation peptide as significantly down-regulated and alpha-defensin as significantly up-regulated.</p> <p>Conclusion</p> <p>This simple and reproducible multimarker proteomic assay could supplement clinical gastroscopic evaluation of symptomatic patients to enhance diagnostic accuracy for gastric cancer and pre-malignant lesions.</p

    Dysregulated Recruitment of the Histone Methyltransferase EZH2 to the Class II Transactivator (CIITA) Promoter IV in Breast Cancer Cells

    One mechanism frequently utilized by tumor cells to escape immune system recognition and elimination is suppression of cell surface expression of Major Histocompatibility Class II (MHC II) molecules. Expression of MHC II is regulated primarily at the level of transcription by the Class II Transactivator, CIITA, and decreased CIITA expression is observed in multiple tumor types. We investigate here contributions of epigenetic modifications to transcriptional silencing of CIITA in variants of the human breast cancer cell line MDA MB 435. Significant increases in histone H3 lysine 27 trimethylation upon IFN-γ stimulation correlate with reductions in transcription factor recruitment to the interferon-γ inducible CIITA promoter, CIITApIV, and with significantly increased CIITApIV occupancy by the histone methyltransferase enhancer of zeste homolog 2 (EZH2). Most compelling is evidence that decreased expression of EZH2 in MDA MB 435 variants results in significant increases in CIITA and HLA-DRA mRNA expression, even in the absence of interferon-γ stimulation, as well as increased cell surface expression of MHC II. Together, these data add mechanistic insight to prior observations of increased EZH2 expression and decreased CIITA expression in multiple tumor types
