    Vortex matter freezing in Bi2_{2}Sr2_{2}CaCu2_{2}O8_{8} samples with a very dense distribution of columnar defects

    We show that the dynamical freezing of vortex structures nucleated at diluted densities in Bi2_{2}Sr2_{2}CaCu2_{2}O8_{8} samples with a dense distribution of columnar defects, B102BΦB \sim 10^{-2} B_{\Phi} with BΦ=5B_{\Phi}=5\,kG, results in configurations with liquid-like correlations. We propose a freezing model considering a relaxation dynamics dominated by double-kink excitations driven by the local stresses obtained directly from experimental images. With this model we estimate the relaxation barrier and the freezing temperature. We argue that the low-field frozen vortex structures nucleated in a dense distribution of columnar defects thus correspond to an out-of-equilibrium non-entangled liquid with strongly reduced mobility rather than to a snapshot of a metastable state with divergent activation barriers as for instance expected for the Bose-glass phase at equilibrium.Comment: 12 pages, 7 figure

    Phenotypic and molecular characterization of Phaseolus vulgaris plants from non-cryopreserved and cryopreserved seeds

    The objective of this work was to evaluate if cryostorage of Phaseolus vulgaris L. seeds induced variations in regenerated plants at the phenotypic and molecular levels. A series of agricultural traits was measured on plants grown from control, non-cryopreserved and cryopreserved seeds, and the genetic stability of plants of the second generation was analysed at selected microsatellite loci. The phenotype of the second generation plants was evaluated as well. No statistically significant phenotypic differences were observed for the parameters measured, neither in the first nor in the second generations. Averaging both treatments, about 76% of the seeds had germinated 10 days after sowing. At harvest we recorded plants with about 73 cm in height, 13 stem internodes, 25 fruits, 103 grains and 4 grains per fruit. One hundred seeds weighted about 26 g. The genetic analyses performed on the second generation plants using six nuclear Simple Sequences Repeats (SSR) markers revealed no changes in microsatellite length between control and cryopreserved samples, implying that there was no effect of seed liquid nitrogen exposure on genome integrity. The phenotypic and molecular results reported here confirm that cryostorage is an efficient and reliable technique to conserve P. vulgaris seeds and regenerate true-to-type plants

    Transcriptional Regulator CNOT3 Defines an Aggressive Colorectal Cancer Subtype.

    Cancer cells exhibit dramatic alterations of chromatin organization at cis-regulatory elements, but the molecular basis, extent, and impact of these alterations are still being unraveled. Here, we identify extensive genome-wide modification of sites bearing the active histone mark H3K4me2 in primary human colorectal cancers, as compared with corresponding benign precursor adenomas. Modification of certain colorectal cancer sites highlighted the activity of the transcription factor CNOT3, which is known to control self-renewal of embryonic stem cells (ESC). In primary colorectal cancer cells, we observed a scattered pattern of CNOT3 expression, as might be expected for a tumor-initiating cell marker. Colorectal cancer cells exhibited nuclear and cytoplasmic expression of CNOT3, suggesting possible roles in both transcription and mRNA stability. We found that CNOT3 was bound primarily to genes whose expression was affected by CNOT3 loss, and also at sites modulated in certain types of colorectal cancers. These target genes were implicated in ESC and cancer self-renewal and fell into two distinct groups: those dependent on CNOT3 and MYC for optimal transcription and those repressed by CNOT3 binding and promoter hypermethylation. Silencing CNOT3 in colorectal cancer cells resulted in replication arrest. In clinical specimens, early-stage tumors that included >5% CNOT3(+) cells exhibited a correlation to worse clinical outcomes compared with tumors with little to no CNOT3 expression. Together, our findings implicate CNOT3 in the coordination of colonic epithelial cell self-renewal, suggesting this factor as a new biomarker for molecular and prognostic classification of early-stage colorectal cancer. Las Acciones desarrolladas y Productos alcanzados se enmarcan en 5 Ejes: a) Investigación sobre el Diseño y Promoción de sistemas para el calentamiento solar de agua; b) Desarrollo de 2 Prototipos Didácticos a escala; c) Experimentación: del Prototipo 2 pues el 1 está en construcción; d) Evaluación y Monitoreo de lo realizado; e) Transferencia: en zonas rurales. La evaluación pudo corroborar: a) el eficaz funcionamiento del Prototipo 2 -en períodos breves, elevó la temperatura del agua entre 60° y 70°C- sobrepasando los niveles requeridos, p.ej. para higiene personal (45-55°C); b) el bajo costo de ambos prototipos en relación a un sistema convencional (calefón a leña).The objectives are: a) to promote the quality of life of the low-income population, and in general, through the development and dissemination strategies and devices that leverage the solar energy; b) to demonstrate its convenience and efficiency for water heating; c) to give tools to the sector for the comfort, hygiene and environmental care requirements; d) promote the development of human resources committed to the social and ecological reality; and e) to provide alternative solutions in the field of Habitat.     Comparative study of lipid and fatty acid composition in different tissues of wild and cultured female broodstock of greater amberjack (Seriola dumerili)

    The aim of this study was to compare the total lipid (TL) content, the lipid class (LC) composition and their associated fatty acids from muscle, liver and ovary of wild and cultured mature females of greater amberjack (Seriola dumerili), in order to obtain information to formulate a more suitable diet for this species broodstock. TL content in muscle and liver was higher in cultured fish than in wild fish, mainly due to TG accumulation, while the ovary TL content was higher in wild fish. Regarding to fatty acids profile, the percentage of 18:1n-9 in TL and TG was lower in ovaries and muscle of cultured fish than in wild ones. Cultured fish displayed lower proportion of arachidonic acid (20:4n-6, ARA) and higher proportions of 18:2n-6 and eicosapentaenoic acid (20:5n-3, EPA) than wild specimens for all tissues in TL and LC. In contrast, differences in the proportion of docosahexaenoic acid (22:6n-3, DHA) between both groups were found only in some tissues and in some LC, being in those cases higher in wild fish. In consequence, cultured fish presented a lower DHA/EPA ratio and a higher EPA/ARA ratio with respect to wild fish. These results suggest that 18:1n-9, 18:2n-6 and essential fatty acids (EFA), especially EPA and ARA, are not supplied in the appropriate proportions in the diet of cultured fish and could negatively affect their reproductive performance

    Phenotypic and genotypic characterization of imipenem-resistant Pseudomonas aeruginosa isolated in Buenos Aires

    En el presente estudio, que tuvo por objeto analizar los mecanismos involucrados en la resistencia a carbapenemes, se incluyeron 129 aislamientos de Pseudomonas aeruginosa recuperados durante el año 2006 en el Hospital “Eva Perón” de la Provincia de Buenos Aires. La caracterización fenotípica y genotípica de la resistencia permitió reconocer la presencia de metalo-β-lactamasas (MBL) en el 14% de esos aislamientos. En todos ellos se identificó la presencia de la enzima IMP-13; sin embargo, algunos aislamientos resultaron sensibles a carbapenemes de acuerdo a los puntos de corte establecidos por el CLSI e incluso a las sugerencias de la Subcomisión de Antimicrobianos de SADEBAC, AAM. El ensayo de detección fenotípica de MBL de sinergia con doble disco resultó útil en este estudio. Sólo aquellos aislamientos productores de IMP-13 que a su vez presentaron alteraciones en las proteínas de membrana externa resultaron completamente resistentes a imipenem. Los aislamientos productores de MBL correspondieron a varios tipos clonales, lo cual sugiere no sólo la diseminación de una cepa resistente, sino también la diseminación horizontal de este mecanismo de resistencia entre clones diferentesrom 129 P. aeruginosa isolated at a health care centre located in Buenos Aires (Hospital “Eva Perón”), 14% produced IMP-13. Although 18 isolates were metallo-β-lactamases (MBL) producers, only those isolates that displayed altered outer membrane protein profiles correlated with the resistant category according to CLSI or even Subcomisión de Antimicrobianos, SADEBAC, AAM. Phenotypic screening of metallo-β-lactamases proved to be appropriate for detecting MBL producing isolates. IMP-13 producing isolates corresponded to at least five different clonal types, which not only suggests the dissemination of the resistant strain but also of the resistant marker.Fil: Cejas, D..
