    Effect of organic loading rate on the production of Polyhydroxyalkanoates from sewage sludge

    The aim of this work was to study the effect of organic loading rate on the production of Polyhydroxyalkanoates (PHA) from sewage sludge. Synthesis of PHA using sewage sludge as platform was achieved in this work. Three pilot-scale selection-sequencing batch reactors (S-SBR) were used for obtaining a culture able to accumulate PHA following a strategy of aerobic dynamic feeding (ADF) at different volumetric organic-loading-rate (vOLR): 1.3, 1.8 and 0.8 g COD L-1 d-1 for S-SBR 1, S-SBR 2 and S-SBR 3, respectively. Decreasing the vOLR enhanced the general performance of the process as for organic matter removal (from 99.2% ± 0.3% in S-SBR-3 to 92 ± 2 in S-SBR-2) while the opposite trend was recorded for PHA production (6.0 PHA % w/w in S-SBR-3 vs 13.7 PHA % w/w in S-SBR-2 at the end of the feast phase). Furthermore, indirect and direct emissions, as N2O, were evaluated during the process for the first time. Finally, three accumulation tests were performed achieving 24% w/w

    Zero-temperature generalized phase diagram of the 4d transition metals under pressure

    We use an accurate implementation of density functional theory (DFT) to calculate the zero-temperature generalized phase diagram of the 4dd series of transition metals from Y to Pd as a function of pressure PP and atomic number ZZ. The implementation used is full-potential linearized augmented plane waves (FP-LAPW), and we employ the exchange-correlation functional recently developed by Wu and Cohen. For each element, we obtain the ground-state energy for several crystal structures over a range of volumes, the energy being converged with respect to all technical parameters to within 1\sim 1 meV/atom. The calculated transition pressures for all the elements and all transitions we have found are compared with experiment wherever possible, and we discuss the origin of the significant discrepancies. Agreement with experiment for the zero-temperature equation of state is generally excellent. The generalized phase diagram of the 4dd series shows that the major boundaries slope towards lower ZZ with increasing PP for the early elements, as expected from the pressure induced transfer of electrons from spsp states to dd states, but are almost independent of PP for the later elements. Our results for Mo indicate a transition from bcc to fcc, rather than the bcc-hcp transition expected from spsp-dd transfer.Comment: 28 pages and 10 figures. Submitted to Phys. Rev.

    Thallium under extreme compression

    We present a combined theoretical and experimental study of the high-pressure behavior of thallium. X-ray diffraction experiments have been carried out at room temperature up to 125 GPa using diamond-anvil cells, nearly doubling the pressure range of previous experiments. We have confirmed the hcp-fcc transition at 3.5 GPa and determined that the fcc structure remains stable up to the highest pressure attained in the experiments. In addition, HP-HT experiments have been performed up to 8 GPa and 700 K by using a combination of x-ray diffraction and a resistively heated diamond-anvil cell. Information on the phase boundaries is obtained, as well as crystallographic information on the HT bcc phase. The equation of state for different phases is reported. Ab initio calculations have also been carried out considering several potential high-pressure structures. They are consistent with the experimental results and predict that, among the structures considered in the calculations, the fcc structure of thallium is stable up to 4.3 TPa. Calculations also predict the post-fcc phase to have a close-packed orthorhombic structure above 4.3 TPa.Comment: 29 pages, 14 figure

    Las bacterias pertenecientes al género Pseudomonas constituyen un modelo de estudio de la interacción microorganismo-planta. Pseudomonas syringae es una especie que produce enfermedades en la parte aérea de algunas plantas. Por otro lado, Pseudomonas chlororaphis interacciona con la raíz y ha demostrado ser un agente de control biológico. Ambas especies forman biopelículas y, a pesar de su diferente estilo de vida, análisis in silico han puesto de manifiesto la existencia de regiones genómicas tanto comunes como diferenciales que parecen estar implicadas en la biosíntesis de matriz extracelular. En este trabajo se han construido mutantes de estas regiones y analizado su implicación en la formación de biopelícula y aspectos relacionados.Universidad de Málaga.     A Psl-like polysaccharide has a key role in the biofilm architecture of two plant-associated Pseudomonas

    Comunicación congresoThe Psl polysaccharide has only been studied in Pseudomonas aeruginosa, and its role in bacteria that interact with plants is still unknown. On the one hand, P. syringae is a model of study of plant-pathogen interactions. On the other hand, P. chlororaphis is a biocontrol agent of plant-fungal diseases, as the white root rot caused by Rosellinia necatrix. In silico analysis have let us identify in both species a genomic region which is ortholog to the Psl-encoding region of P. aeruginosa PAO1. We constructed mutants of this region in both strains and analysed its phenotype in biofilm formation, both in static microwell plates and dynamic flow-cell chamber experiments. The results revealed a different and an important role of this polysaccharide in the biofilm architecture of both strains. Furthermore, in P. syringae this Psl-like polysaccharide has a very important role in swarming motility. All these striking phenotypes led us further study the implication of this polysaccharide in the lifestyles of both models of plant-bacteria interaction.Universidad de Málaga.     Extensive Monte Carlo simulations of helium-4 monolayer films adsorbed on weak substrates have been carried out, aimed at ascertaining the possible occurrence of a quasi-two-dimensional supersolid phase. Only crystalline films not registered with underlying substrates are considered. Numerical results yield strong evidence that helium-4 will not form a supersolid film on {any} substrate strong enough to stabilize a crystalline layer. On weaker substrates, continuous growth of a liquid film takes place

    Solidification of small para-H2 clusters at zero temperature

    We have determined the ground-state energies of para-H2_2 clusters at zero temperature using the diffusion Monte Carlo method. The liquid or solid character of each cluster is investigated by restricting the phase through the use of proper importance sampling. Our results show inhomogeneous crystallization of clusters, with alternating behavior between liquid and solid phases up to N=55. From there on, all clusters are solid. The ground-state energies in the range N=13--75 are established and the stable phase of each cluster is determined. In spite of the small differences observed between the energy of liquid and solid clusters, the corresponding density profiles are significantly different, feature that can help to solve ambiguities in the determination of the specific phase of H2_2 clusters.Comment: 17 pages, accepted for publication in J.     Effects of Chronic Atrial Fibrillation on Active and Passive Force Generation in Human Atrial Myofibrils

    Rationale: Chronic atrial fibrillation (cAF) is associated with atrial contractile dysfunction. Sarcomere remodeling may contribute to this contractile disorder. Objective: Here, we use single atrial myofibrils and fast solution switching techniques to directly investigate the impact of cAF on myofilament mechanical function eliminating changes induced by the arrhythmia in atrial myocytes membranes and extracellular components. Remodeling of sarcomere proteins potentially related to the observed mechanical changes is also investigated. Methods and Results: Myofibrils were isolated from atrial samples of 15 patients in sinus rhythm and 16 patients with cAF. Active tension changes following fast increase and decrease in [Ca2+] and the sarcomere length\u2013passive tension relation were determined in the 2 groups of myofibrils. Compared to sinus rhythm myofibrils, cAF myofibrils showed (1) a reduction in maximum tension and in the rates of tension activation and relaxation; (2) an increase in myofilament Ca2+ sensitivity; (3) a reduction in myofibril passive tension. The slow \u3b2-myosin heavy chain isoform and the more compliant titin isoform N2BA were up regulated in cAF myofibrils. Phosphorylation of multiple myofilament proteins was increased in cAF as compared to sinus rhythm atrial myocardium. Conclusions: Alterations in active and passive tension generation at the sarcomere level, explained by translational and post-translational changes of multiple myofilament proteins, are part of the contractile dysfunction of human cAF and may contribute to the self-perpetuation of the arrhythmia and the development of atrial dilatation

    Trypanocidal Activity of Smallanthus sonchifolius. Identification of Active Sesquiterpene Lactones by Bioassay-Guided Fractionation.

    In order to find novel plant-derived biologically active compounds against Trypanosoma cruzi, we isolated, from the organic extract of Smallanthus sonchifolius, the sesquiterpene lactones enhydrin (1), uvedalin (2), and polymatin B (3) by bioassay-guided fractionation technique. These compounds showed a significant trypanocidal activity against the epimastigote forms of the parasite with IC50 values of 0.84 μM (1), 1.09 μM (2), and 4.90 μM (3). After a 24 h treatment with 10 μg/mL of enhydrin or uvedalin, parasites were not able to recover their replication rate. Compounds 1 and 2 showed IC50 values of 33.4 μM and 25.0 μM against T. cruzi trypomastigotes, while polymatin B was not active. When the three compounds were tested against the intracellular forms of T. cruzi, they were able to inhibit the amastigote replication with IC50 of 5.17 μM, 3.34 μM, and 9.02 μM for 1, 2, and 3, respectively. The cytotoxicity of the compounds was evaluated in Vero cells obtaining CC50 values of 46.5 μM (1), 46.8 μM (2), and 147.3 μM (3) and the selectivity index calculated. According to these results, enhydrin and uvedalin might have potentials as agents against Chagas disease and could serve as lead molecules to develop new drugs.Fil: Frank, Fernanda Maria.     Spatial and temporal variability of carbonaceous aerosols: assessing the impact of biomass burning in the urban environment

    Biomass burning (BB) is a significant source of atmospheric particles in many parts of the world. Whereas many studies have demonstrated the importance of BB emissions in central and northern Europe, especially in rural areas, its impact in urban air quality of southern European countries has been sparsely investigated. In this study, highly time resolved multi-wavelength absorption coefficients together with levoglucosan (BB tracer) mass concentrations were combined to apportion carbonaceous aerosol sources. The Aethalometer model takes advantage of the different spectral behaviour of BB and fossil fuel (FF) combustion aerosols. The model was found to be more sensitive to the assumed value of the aerosol Ångström exponent (AAE) for FF (AAEff) than to the AAE for BB (AAEbb). As result of various sensitivity tests the model was optimized with AAEff = 1.1 and AAEbb = 2. The Aethalometer model and levoglucosan tracer estimates were in good agreement. The Aethalometer model was further applied to data from three sites in Granada urban area to evaluate the spatial variation of CMff and CMbb (carbonaceous matter from FF or BB origin, respectively) concentrations within the city. The results showed that CMbb was lower in the city centre while it has an unexpected profound impact on the CM levels measured in the suburbs (about 40%). Analysis of BB tracers with respect to wind speed suggested that BB was dominated by sources outside the city, to the west in a rural area. Distinguishing whether it corresponds to agricultural waste burning or with biomass burning for domestic heating was not possible. This study also shows that although traffic restrictions measures contribute to reduce carbonaceous concentrations, the extent of the reduction is very local.
