    Apical transport and folding of prostate-specific membrane antigen occurs independent of glycan processing.

    Prostate-specific membrane antigen (PSMA) is an integral cell-surface membrane glycoprotein that is overexpressed in prostate carcinomas rendering it an appropriate target for antibody-based therapeutic strategies. The biosynthesis of PSMA in transfected COS-1 cells reveals a slow conversion of mannose-rich to complex glycosylated PSMA compatible with slow transport kinetics from the endoplasmic reticulum to the Golgi. Importantly, mannose-rich PSMA persists as a trypsin-sensitive protein throughout its entire life cycle, and only Golgi-located PSMA glycoforms acquire trypsin resistance. This resistance, used here as a tool to examine correct folding, does not depend on the type of glycosylation, because different PSMA glycoforms generated in the presence of inhibitors of carbohydrate processing in the Golgi are also trypsin resistant. The conformational transition of PSMA to a correctly folded molecule is likely to occur in the Golgi and does not implicate ER molecular chaperones, such as BiP. We show here that PSMA is not only heavily N-but also O-glycosylated. The question arising is whether glycans, which do not play a role in folding of PSMA, are implicated in its transport to the cell surface. Neither the cell-surface expression of PSMA nor its efficient apical sorting in polarized Madin-Darby canine kidney cells are influenced by modulators of N- and O-glycosylation. The acquisition of folding determinants in the Golgi, therefore, is an essential prerequisite for protein trafficking and sorting of PSMA and suggests that altered or aberrant glycosylation often occurring during tumorigenesis has no regulatory effect on the cell-surface expression of PSMA

    Genetic dissection of maize phenology using an intraspecific introgression library

    Background: Collections of nearly isogenic lines where each line carries a delimited portion of a donor source genome into a common recipient genetic background are known as introgression libraries and have already shown to be instrumental for the dissection of quantitative traits. By means of marker-assisted backcrossing, we have produced an introgression library using the extremely early-flowering maize (Zea mays L.) variety Gasp\ue9 Flint and the elite line B73 as donor and recipient genotypes, respectively, and utilized this collection to investigate the genetic basis of flowering time and related traits of adaptive and agronomic importance in maize.Results: The collection includes 75 lines with an average Gasp\ue9 Flint introgression length of 43.1 cM. The collection was evaluated for flowering time, internode length, number of ears, number of nodes (phytomeres), number of nodes above the ear, number and proportion of nodes below the ear and plant height. Five QTLs for flowering time were mapped, all corresponding to major QTLs for number of nodes. Three additional QTLs for number of nodes were mapped. Besides flowering time, the QTLs for number of nodes drove phenotypic variation for plant height and number of nodes below and above the top ear, but not for internode length. A number of apparently Mendelian-inherited phenotypes were also observed.Conclusions: While the inheritance of flowering time was dominated by the well-known QTL Vgt1, a number of other important flowering time QTLs were identified and, thanks to the type of plant material here utilized, immediately isogenized and made available for fine mapping. At each flowering time QTL, early flowering correlated with fewer vegetative phytomeres, indicating the latter as a key developmental strategy to adapt the maize crop from the original tropical environment to the northern border of the temperate zone (southern Canada), where Gasp\ue9 Flint was originally cultivated. Because of the trait differences between the two parental genotypes, this collection will serve as a permanent source of nearly isogenic materials for multiple studies of QTL analysis and cloning. \ua9 2011 Salvi et al; licensee BioMed Central Ltd

    Climate-informed environmental inflows to revive a drying lake facing meteorological and anthropogenic droughts

    The rapid shrinkage of Lake Urmia, one of the world\u27s largest saline lakes located in northwestern Iran, is a tragic wake-up call to revisit the principles of water resources management based on the socio-economic and environmental dimensions of sustainable development. The overarching goal of this paper is to set a framework for deriving dynamic, climate-informed environmental inflows for drying lakes considering both meteorological/climatic and anthropogenic conditions. We report on the compounding effects of meteorological drought and unsustainable water resource management that contributed to Lake Urmia\u27s contemporary environmental catastrophe. Using rich datasets of hydrologic attributes, water demands and withdrawals, as well as water management infrastructure (i.e. reservoir capacity and operating policies), we provide a quantitative assessment of the basin\u27s water resources, demonstrating that Lake Urmia reached a tipping point in the early 2000s. The lake level failed to rebound to its designated ecological threshold (1274 m above sea level) during a relatively normal hydro-period immediately after the drought of record (1998–2002). The collapse was caused by a marked overshoot of the basin\u27s hydrologic capacity due to growing anthropogenic drought in the face of extreme climatological stressors. We offer a dynamic environmental inflow plan for different climate conditions (dry, wet and near normal), combined with three representative water withdrawal scenarios. Assuming effective implementation of the proposed 40% reduction in the current water withdrawals, the required environmental inflows range from 2900 million cubic meters per year (mcm yr−1) during dry conditions to 5400 mcm yr−1 during wet periods with the average being 4100 mcm yr−1. Finally, for different environmental inflow scenarios, we estimate the expected recovery time for re-establishing the ecological level of Lake Urmia

    The genetic control of flowering time has been addressed by many quantitative trait locus (QTL) studies. A survey of the results from 29 independent studies reporting information on 441 QTLs led to the production of a QTL consensus map, which enabled the identification of 59 chromosome regions distributed on all chromosomes and shown to be frequently involved in the genetic control of flowering time and related traits. One of the major QTLs for flowering time, the Vegetative to generative transition 1 (Vgt1) locus , corresponds to an upstream (70 kb) non-coding regulatory element of ZmRap2.7, a repressor of flowering. A transposon (MITE) insertion was identified as a major allelic difference within Vgt1. One of the hypotheses is that Vgt1 might function by modifying ZmRap2.7 chromatin through an epigenetic mechanism. Therefore, the methylation state at Vgt1 was investigated using an approach that combines digestion with McrBc, an endonuclease that acts upon methylated DNA, and quantitative PCR. The analyses were performed on genomic DNA from leaves of six different maize lines at four stages of development. The results showed a trend of reduction of methylation from the first to the last stage with the exception of a short genomic region flanking the MITE insertion, which showed a constant and very dense methylation throughout leaf development and for both alleles. Preliminary results from bisulfite sequencing of a small portion of Vgt1 revealed differential methylation of a single cytosine residue between the two alleles. ZmRap2.7 expression was assayed in the four developmental stages afore mentioned for the six genotypes, in order to establish a link between methylation at Vgt1 and ZmRap2.7 transcription.     Expansion of the maize growing area was central for food security in temperate regions. In addition to the suppression of the short-day requirement for floral induction, it required breeding for a large range of flowering time that compensates the effect of South-North gradients of temperatures. Here we show the role of a novel florigen gene, ZCN12, in the latter adaptation in cooperation with ZCN8. Strong eQTLs of ZCN8 and ZCN12, measured in 327 maize lines, accounted for most of the genetic variance of flowering time in platform and field experiments. ZCN12 had a strong effect on flowering time of transgenic Arabidopsis thaliana plants; a path analysis showed that it directly affected maize flowering time together with ZCN8. The allelic composition at ZCN QTLs showed clear signs of selection by breeders. This suggests that florigens played a central role in ensuring a large range of flowering time, necessary for adaptation to temperate areas

    Hemolytic uremic syndrome (HUS) is a thrombotic microangiopathy with manifestations of hemolytic anemia, thrombocytopenia, and renal impairment. Genetic studies have shown that mutations in complement regulatory proteins predispose to non–Shiga toxin–associated HUS (non-Stx–HUS). We undertook genetic analysis on membrane cofactor protein (MCP), complement factor H (CFH), and factor I (IF) in 156 patients with non-Stx–HUS. Fourteen, 11, and 5 new mutational events were found in MCP, CFH, and IF, respectively. Mutation frequencies were 12.8%, 30.1%, and 4.5% for MCP, CFH, and IF, respectively. MCP mutations resulted in either reduced protein expression or impaired C3b binding capability. MCP-mutated patients had a better prognosis than CFH-mutated and nonmutated patients. In MCP-mutated patients, plasma treatment did not impact the outcome significantly: remission was achieved in around 90% of both plasma-treated and plasma-untreated acute episodes. Kidney transplantation outcome was favorable in patients with MCP mutations, whereas the outcome was poor in patients with CFH and IF mutations due to disease recurrence. This study documents that the presentation, the response to therapy, and the outcome of the disease are influenced by the genotype. Hopefully this will translate into improved management and therapy of patients and will provide the way to design tailored treatments

    Use of Dalbavancin in Skin, Bone and Joint Infections: A Real-Life Experience in an Italian Center

    Dalbavancin is a lipoglycopeptide approved for the treatment of acute bacterial skin and skin structure infections (ABSSSI). The aim of the study was to evaluate the efficacy and safety in all patients who received at least one administration of dalbavancin. Methods: We carried out a retrospective study of the use of dalbavancin in 55 patients at the Azienda Ospedaliera Ospedali Riuniti Umberto I (Ancona, Italy) from February 2017 to May 2020 and compared “on label” and “off label” use of dalbavancin in ABSSSI and non-ABSSSI. Results: A total of 55 patients were included in the study. The median age was 61 years; 51% had ABSSSI; 24% had prosthetic joint infections, and 14% had osteomyelitis. A total of 53% received a single 1500 mg infusion of dalbavancin, and 18% received a second dose 14 days later; 24% of patients received further doses at 14-day intervals. In 91% of cases, patients achieved clinical objectives with dalbavancin: 96% of patients with ABSSSI and 69% of those with prosthetic joint infections. Conclusions: Dalbavancin was shown to have an excellent tolerability profile and to be a highly successful therapeutic approach even in those cases treated “off-label”
