    Development of a one-step embryonic stem cell-based assay for the screening of sprouting angiogenesis

    BACKGROUND: Angiogenesis assays are important tools for the identification of regulatory molecules and the potential development of therapeutic strategies to modulate neovascularization. Although numerous in vitro angiogenesis models have been developed in the past, they exhibit limitations since they do not recapitulate the entire angiogenic process or correspond to multi-step procedures that are not easy to use. Convenient, reliable, easily quantifiable and physiologically relevant assays are still needed for pharmacological screenings of angiogenesis. RESULTS: Here, we have optimized an angiogenesis model based on ES cell differentiation for screening experiments. We have established conditions leading to angiogenic sprouting of embryoid bodies during ES cell differentiation in type I three-dimensional collagen gels. Immunostaining experiments carried out during these cultures showed the formation of numerous buds comprising CD31 positive cells, after 11 days of culture of ES cells. Moreover, this one-step model has been validated in response to activators and inhibitors of angiogenesis. Sprouting was specifically stimulated in the presence of VEGF and FGF2. Alternatively, endothelial sprouting induced by angiogenic activators was inhibited by angiogenesis inhibitors such as angiostatin, TGFÎČ and PF4. Sprouting angiogenesis can be easily quantified by image analysis after immunostaining of endothelial cells with CD31 pan-endothelial marker. CONCLUSION: Taken together, these data clearly validate that this one-step ES differentiation model constitutes a simple and versatile angiogenesis system that should facilitate, in future investigations, the screening of both activators and inhibitors of angiogenesis

    Abstract-Edge detection is a fundamental issue in image analysis. Due to the presence of speckle, which can be modeled as a strong, multiplicative noise, edge detection in synthetic aperture radar (SAR) images is extremely difficult, and edge detectors developed for optical images are inefficient. Several robust operators have been developed for the detection of isolated step edges in speckled images. We propose a new step-edge detector for SAR images, which is optimal in the minimum mean square error (MSSE) sense under a stochastic multiedge model. It computes a normalized ratio of exponentially weighted averages (ROEWA) on opposite sides of the central pixel. This is done in the horizontal and vertical direction, and the magnitude of the two components yields an edge strength map. Thresholding of the edge strength map by a modified version of the watershed algorithm and region merging to eliminate false edges complete an efficient segmentation scheme. Experimental results obtained from simulated SAR images as well as ERS-1 data are presented.     Triphenylamine/Tetracyanobutadiene-Based π-Conjugated Push–Pull Molecules End-Capped with Arene Platforms:Synthesis, Photophysics, and Photovoltaic Response

    π-Conjugated push–pull molecules based on triphenylamine and 1,1,4,4-tetracyanobuta-1,3-diene (TCBD) have been functionalized with different terminal arene units. In solution, these highly TCBD-twisted systems showed a strong internal charge transfer band in the visible spectrum and no detectable photoluminescence (PL). Photophysical and theoretical investigations revealed very short singlet excited state deactivation time of ≈10 ps resulting from significant conformational changes of the TCBD-arene moiety upon photoexcitation, opening a pathway for non-radiative decay. The PL was recovered in vacuum-processed films or when the molecules were dispersed in a PMMA matrix leading to a significant increase of the excited state deactivation time. As shown by cyclic voltammetry, these molecules can act as electron donors compared to C 60. Hence, vacuum-processed planar heterojunction organic solar cells were fabricated leading to a maximum power conversion efficiency of ca. 1.9 % which decreases with the increase of the arene size

    Speaker detection in the wild: Lessons learned from JSALT 2019

    Submitted to ICASSP 2020This paper presents the problems and solutions addressed at the JSALT workshop when using a single microphone for speaker detection in adverse scenarios. The main focus was to tackle a wide range of conditions that go from meetings to wild speech. We describe the research threads we explored and a set of modules that was successful for these scenarios. The ultimate goal was to explore speaker detection; but our first finding was that an effective diarization improves detection, and not having a diarization stage impoverishes the performance. All the different configurations of our research agree on this fact and follow a main backbone that includes diarization as a previous stage. With this backbone, we analyzed the following problems: voice activity detection, how to deal with noisy signals, domain mismatch, how to improve the clustering; and the overall impact of previous stages in the final speaker detection. In this paper, we show partial results for speaker diarizarion to have a better understanding of the problem and we present the final results for speaker detection
