    Polymeric coatings for photostability enhancement of poly(p-phenylene vinylene) derivative films

    Poly(p-phenylene vinylene) (PPV) derivatives are an important class of conjugated polymers, known for their applications as electroluminescent materials for light-emitting devices and sensors. These derivatives are highly susceptible to photodegradation by the combined action of oxygen and light. Here, the use of various commercial polymers as protective coatings against the photodegradation of PPV derivatives was explored. Cast films of two similar PPV derivatives, poly[(2-methoxy-5-n-hexyloxy)-p-phenylene vinylene] and poly[2-methoxy-5-(2-ethylhexyloxy)-p-phenylene vinylene], were submitted to photodegradation by exposure to white light under atmospheric conditions in order to verify if the type of side chain (linear or branched) had an effect on the photodegradation. No significant differences in the photodegradation behaviour between the two polymers were noticed. The following commercial polymers were tested as protective coatings for the PPV derivative cast films: 99 and 80% hydrolysed poly(vinyl alcohol) (PVA) and starch. The best results were achieved using coatings of 99% hydrolysed PVA, which increased about 700 times the time necessary for complete degradation of the PPV derivative films. The results show the effectiveness of this coating in minimizing and, possibly, controlling the effects of the photodegradation of PPV derivative films, which can be useful in many applications, e.g. oxygen sensors.FAPESPCNPqInstituto Multidisciplinar de Materiais Poliméricos (IMMP)/MCT

    Neutrino mixing and large CP violation in B physics

    We show that in see-saw models of neutrino mass a la SUSY SO(10), the observed large mixing in atmospheric neutrinos naturally leads to large b-s transitions. If the associated new CP phase turns out to be large, this SUSY contributions can drastically affect the CP violation in some of the B decay channels yielding the beta and gamma angles of the unitarity triangle. They can even produce sizeable CP asymmetries in some decay modes which are not CP violating in the standard model context. Hence the observed large neutrino mixing makes observations of low energy SUSY effect in some CP violating decay channels potentially promising in spite of the agreement between the Standard Model and data in K and B physics so far.Comment: References adde

    This article describes a solution that allows people with visual impairments who also have the inability to use hands, to have access to the Dosvox system, a system that enables the interaction between the visually impaired. In this solution, the Dosvox, whose operation is originally performed solely by keyboard, now can be operated via a gyroscope coupled to the user's head, whose movement is translated into a series of keystrokes, chosen to provide the full operation of the menus Dosvox which correspond to a high percentage of interaction with this system. The solution identified several problems to be solved, they were treated by means of algoritms and techniques presented in the article.Este artigo descreve uma solução que permite à pessoas com deficiência visual, que também têm impossibilidade de utilizar as mãos, possam ter acesso ao sistema Dosvox, um sistema que possibilita a interação entre deficiente visual e o computador. O Dosvox é uma das tecnologias assistivas mais utilizadas no Brasil, por deficientes visuais. Nesta solução, o Dosvox, cuja operação é originalmente realizada unicamente pelo teclado, passa a poder ser operado através de um giroscópio, acoplado à cabeça do usuário, cujo movimento é traduzido numa série de acionamentos de teclas, escolhidas para proporcionar a operação plena dos menus do Dosvox, que correspondem a um alto percentual da interação com este sistema.     Functional Inactivation of CXC Chemokine Receptor 4–mediated Responses through SOCS3 Up-regulation

    Hematopoietic cell growth, differentiation, and chemotactic responses require coordinated action between cytokines and chemokines. Cytokines promote receptor oligomerization, followed by Janus kinase (JAK) kinase activation, signal transducers and transactivators of transcription (STAT) nuclear translocation, and transcription of cytokine-responsive genes. These include genes that encode a family of negative regulators of cytokine signaling, the suppressors of cytokine signaling (SOCS) proteins. After binding their specific receptors, chemokines trigger receptor dimerization and activate the JAK/STAT pathway. We show that SOCS3 overexpression or up-regulation, stimulated by a cytokine such as growth hormone, impairs the response to CXCL12, measured by Ca2+ flux and chemotaxis in vitro and in vivo. This effect is mediated by SOCS3 binding to the CXC chemokine receptor 4 receptor, blocking JAK/STAT and Gαi pathways, without interfering with cell surface chemokine receptor expression. The data provide clear evidence for signaling cross-talk between cytokine and chemokine responses in building a functional immune system

    Objective: The metalloprotease ADAM17 (also called TACE) plays fundamental roles in homeostasis by shedding key signaling molecules from the cell surface. Although its importance for the immune system and epithelial tissues is well-documented, little is known about the role of ADAM17 in metabolic homeostasis. The purpose of this study was to determine the impact of ADAM17 expression, specifically in adipose tissues, on metabolic homeostasis.Methods: We used histopathology, molecular, proteomic, transcriptomic, in vivo integrative physiological and ex vivo biochemical approaches to determine the impact of adipose tissue-specific deletion of ADAM17 upon adipocyte and whole organism metabolic physiology.Results: ADAM17adipoq-creD/D mice exhibited a hypermetabolic phenotype characterized by elevated energy consumption and increased levels of adipocyte thermogenic gene expression. On a high fat diet, these mice were more thermogenic, while exhibiting elevated expression levels of genes associated with lipid oxidation and lipolysis. This hypermetabolic phenotype protected mutant mice from obesogenic challenge, limiting weight gain, hepatosteatosis and insulin resistance. Activation of beta-adrenoceptors by the neurotransmitter norepinephrine, a key regulator of adipocyte physiology, triggered the shedding of ADAM17 substrates, and regulated ADAM17 expression at the mRNA and protein levels, hence identifying a functional connection between thermogenic licensing and the regulation of ADAM17. Proteomic studies identified Semaphorin 4B (SEMA4B), as a novel ADAM17-shed adipokine, whose expression is regulated by physiological thermogenic cues, that acts to inhibit adipocyte differentiation and dampen thermogenic responses in adipocytes. Transcriptomic data showed that cleaved SEMA4B acts in an autocrine manner in brown adipocytes to repress the expression of genes involved in adipogenesis, thermogenesis, and lipid uptake, storage and catabolism.Conclusions: Our findings identify a novel ADAM17-dependent axis, regulated by beta-adrenoceptors and mediated by the ADAM17-cleaved form of SEMA4B, that modulates energy balance in adipocytes by inhibiting adipocyte differentiation, thermogenesis and lipid catabolism.(c) 2023 The Authors.     Antiproliferative Activity Of Flavonoids From Croton Sphaerogynus Baill. (euphorbiaceae).

    Croton sphaerogynus is a shrub from the Atlantic Rain Forest in southeastern Brazil. A lyophilized crude EtOH extract from leaves of C. sphaerogynus, obtained by maceration at room temperature (seven days), was suspended in methanol and partitioned with hexane. The purified MeOH phase was fractionated over Sephadex LH-20 yielding five fractions (F1-F5) containing flavonoids, as characterized by HPLC-DAD and HPLC-MS analyses. The antiproliferative activity of the crude EtOH extract, MeOH and hexane phases, and fractions F1-F5 was evaluated on in vitro cell lines NCI-H460 (nonsmall cell lung), MCF-7 (breast cancer), and U251 (glioma). The MeOH phase showed activity (mean log GI50 0.54) higher than the hexane phase and EtOH extract (mean log GI50 1.13 and 1.19, resp.). F1 exhibited activity against NCI-H460 (nonsmall cell lung) (GI50 1.2 μg/mL), which could be accounted for the presence of flavonoids and/or diterpenes. F4 showed moderate activity (mean log GI50 1.05), while F5 showed weak activity (mean log GI50 1.36). It is suggested that the antiproliferative activity of the crude EtOH extract and MeOH phase is accounted for a synergistic combination of flavonoids and diterpenes.201521280
