    Contributions of IFN-gamma and granulysin to the clearance of Plasmodium yoelii blood stage

    P. vivax-infected Retics (iRetics) express human leukocyte antigen class I (HLA-I), are recognized by CD8+ T cells and killed by granulysin (GNLY) and granzymes. However, how Plasmodium infection induces MHC-I expression on Retics is unknown. In addition, whether GNLY helps control Plasmodium infection in vivo has not been studied. Here, we examine these questions using rodent infection with the P. yoelii 17XNL strain, which has tropism for Retics. Infection with P. yoelii caused extramedullary erythropoiesis, reticulocytosis and expansion of CD8+CD44+CD62L- IFN-gamma-producing T cells that form immune synapses with iRetics. We now provide evidence that MHC-I expression by iRetic is dependent on IFN-gamma-induced transcription of IRF-1, MHC-I and beta2-microglobulin (beta2-m) in erythroblasts. Consistently, CTLs from infected wild type (WT) mice formed immune synapses with iRetics in an IFN-gamma- and MHC-I-dependent manner. When challenged with P. yoelii 17XNL, WT mice cleared parasitemia and survived, while IFN-gamma KO mice remained parasitemic and all died. beta2-m KO mice that do not express MHC-I and have virtually no CD8+ T cells had prolonged parasitemia, and 80% survived. Because mice do not express GNLY, GNLY-transgenic mice can be used to assess the in vivo importance of GNLY. Parasite clearance was accelerated in GNLY-transgenic mice and depletion of CD8+ T cells ablated the GNLY-mediated resistance to P. yoelii. Altogether, our results indicate that in addition to previously described mechanisms, IFN-gamma promotes host resistance to the Retic-tropic P. yoelii 17XNL strain by promoting MHC-I expression on iRetics that become targets for CD8+ cytotoxic T lymphocytes and GNLY

    Infestação de Enneothrips flavens Moulton e produtividade de cultivares de amendoim

    Because of damages caused, general occurrence in crops, and high population levels, the most important peanut pest in Brazil is the thrips Enneothrips flavens Moulton (Thysanoptera: Thripidae). A field experiment was carried out during the 2001/2002 growing season, in Campinas and Pindorama, State of São Paulo, Brazil, to evaluate the influence of E. flavens on yield of six peanut cultivars. Treatments comprised three cultivars of the upright type and three of the runner type, with and without chemical control of the insect. Thrips infestation was evaluated by insect countings on young leaflets. Yield and yield reduction percentage due to the lack of thrips control were estimated. The lack of insect control resulted in yield reductions varying from 19.5 to 62.7%, depending on the level of infestation, cultivars, and location. Runner cultivar IAC Caiapo, presented the lowest yield reductions in both locations (19.5 and 28.7%), even when exposed to thrips infestation for a longer period because of its longer cycle. This fact characterizes 'IAC Caiapo' as resistant to E. flavens.Em função dos danos causados e ocorrência generalizada na cultura, a praga mais importante do amendoim no Brasil é Enneothrips flavens Moulton (Thysanoptera: Thripidae). Com o objetivo de avaliar a influência do ataque de E. flavens na produtividade de seis cultivares de amendoim, foi implantado um experimento em campo, no ano agrícola 2001/2002, em duas localidades do Estado de São Paulo: Campinas e Pindorama. As cultivares de amendoim avaliadas foram três de porte vegetativo ereto e três rasteiros, com controle e ausência total de controle químico da praga. Para a avaliação da infestação de tripes, foram realizadas amostragens a partir dos 29 dias após o plantio, a cada duas semanas, quando foram coletados 10 folíolos fechados por subparcela. Ao final do ciclo, avaliou-se o peso em casca das parcelas e determinou-se o índice de redução na produção provocado pela ausência de controle do tripes nessas cultivares. A ausência de controle do tripes provocou reduções de produção entre 19,5 a 62,7%, dependendo do nível de infestação, da cultivar utilizada e do local de plantio. Embora exposto a um maior período de infestação devido a seu ciclo mais longo, 'IAC Caiapó' apresentou a menor perda produtiva quando não houve controle da praga (19,5 a 28,7%, dependendo da localidade plantada), demonstrando possuir resistência a esse inseto

    The evaluation of the antibacterial capacity of tea tree functionalized microcapsules in textiles

    The textile industry develops products that go beyond aesthetic concerns. Manufacturers functionalize these new textile products to provide them with new technical properties, to meet and even exceed user needs. The technical properties are diverse, from ultraviolet protection to antimicrobial or self-cleaning properties. The growth of microorganisms on textiles can result in unpleasant odors, stains or even accelerate the wear and tear of textile products. Microcapsules may contain natural or synthetic components in their core, capable of providing the desired properties to textiles (Figure 1). In addition, a shell protects the core and its design allows a gradual release of the main component. In this work, microcapsules were dyed to prevent the appearance of stains. This research tested the antibacterial capacity of microcapsules functionalized with tea tree essential oil on polyester fabric (PES). The fabric was tested with dyed and undyed microcapsules to ensure no dye interference.     Although metacyclic and blood trypomastigotes are completely functional in relation to parasite-host interaction and/or target cell invasion, they differ in the molecules present on the surface. Thus, aspects related to the variability that the forms of T. cruzi interacts with host cells may lead to fundamental implications on the immune response against this parasite and, consequently, the clinical evolution of Chagas disease. We have shown that BT infected mice presented higher levels of parasitemia during all the acute phase of infection. Moreover, the infection with either MT or BT forms resulted in increased levels of total leukocytes, monocytes and lymphocytes, specifically later for MT and earlier for BT. The infection with BT forms presented earlier production of proinflammatory cytokine TNF-α and later of IFN-γ by both T cells subpopulations. This event was accompanied by an early cardiac inflammation with an exacerbation of this process at the end of the acute phase. On the other hand, infection with MT forms result in an early production of IFN-γ, with subsequent control in the production of this cytokine by IL-10, which provided to these animals an immunomodulatory profile in the end of the acute phase. These results are in agreement with what was found for cardiac inflammation where animals infected with MT forms showed intense cardiac inflammation later at infection, with a decrease in the same at the end of this phase. In summary, our findings emphasize the importance of taking into account the inoculums source of T. cruzi, since vectorial or transfusional routes of T. cruzi infection may trigger distinct parasite-host interactions during the acute phase that may influence relevant biological aspects of chronic Chagas disease

They are the closest genetically to humans of any of the models applied in biomedical research; therefore, many authors have published scientific work regarding these animals and infectious diseases, including tuberculosis, AIDS, and tropical diseases. Among these, Chagas disease has caught the attention of many researchers all over the world. Recent studies have demonstrated great similarities with the human pathology, including cardiomyopathy and exacerbated pro-inflammatory response. Besides being genetically close to humans, NHP have a great probability to be naturally infected by Trypanosoma cruzi, which turns them into more interesting models to study Chagas disease mechanisms

    Physiology and Pathology of Infectious Diseases: The Autoimmune Hypothesis of Chagas Disease

    Infectious pathologies are a group of diseases that contribute with great impact on public health worldwide. Among the various diseases, some have a higher epidemiological importance, since their morbidity and mortality are very significant. In addition to the usual immune response, mounted against noxious agents, there is still the concept of infection-induced autoimmunity. Autoimmune diseases are defined as illnesses in which the evolution from benign to pathogenic autoimmunity takes place. However, proving a disease to be of autoimmune etiology is not a simple task. It is well known that both genetic influences and environmental factors trigger autoimmune disorders. However, some theories are still under great discussion. One of the most intriguing self-induced disorders is the hypothesis of autoimmunity during Chagas disease. Since the mid-1970s, the Chagas autoimmunity hypothesis has been considered an important contributor to the complex immune response developed by the host and triggered by Trypanosoma cruzi. New ideas and findings have strengthened this hypothesis, which has been reported in a series of publications from different groups around the world.     The objective of this work was to determine the suitability of Setaria viridis as a model plant in studies to validate candidate genes for cold tolerance by evaluating the response of two of its accessions to different durations of abrupt or gradual cold stress in the vegetative and reproductive stages. Plants of accessions A10.1 and Ast-1, cultivated at 25°C, were subjected to the following cold stress treatments: gradual reduction in temperature from 25 to 0°C, 5°C at a time, every 24 hours in a same chamber; or abrupt reduction in temperature, by transferring plants from a chamber at 25°C to another at 0°C. Plants were kept at 0°C for 3, 5, or 10 days, after which temperature was increased back again to 25°C; a control group remained at 25ºC. Low temperatures – reduced abruptly or gradually – caused a decrease in the gas exchange rates and shoot and root biomass of the plants, besides damage to their photochemical apparatus; the longer the cold lasted, the more pronounced the effect was. Regardless of stress duration, plants recovered and completed their life cycle. The studied accessions are tolerant to cold and, therefore, are not suitable as a model plant in studies to validate candidate genes for cold tolerance.O objetivo deste trabalho foi determinar a adequação de Setaria viridis como planta modelo em estudos de validação de genes candidatos à tolerância ao frio, ao avaliar a resposta de dois de seus acessos a diferentes durações de estresse por frio abrupto ou gradual, nas fases vegetativa e reprodutiva. Plantas dos acessos A10.1 e Ast-1, cultivadas a 25°C, foram submetidas aos seguintes tratamentos de estresse por frio: redução gradual da temperatura de 25 a 0°C, 5°C de cada vez, a cada 24 horas, em única câmera; ou redução abrupta da temperatura, pela transferência das plantas de uma câmera a 25°C para outra a 0°C. As plantas foram mantidas a 0°C por 3, 5, ou 10 dias, após os quais a temperatura foi aumentada novamente para 25°C; um grupo controle permaneceu a 25ºC. As baixas temperaturas – reduzidas gradual ou abruptamente – causaram redução nas taxas de troca gasosa e na biomassa da parte aérea e da raiz das plantas, além de prejuízos ao seu aparato fotoquímico; quanto mais o frio durou, mais pronunciado o efeito foi. Independentemente da duração do estresse, as plantas se recuperaram e completaram seu ciclo de vida. Os acessos estudados são tolerantes ao frio, e, portanto, não são adequados como planta modelo em estudos para validação de genes candidatos de tolerância ao frio

    Infection with Plasmodium vivax results in strong activation of monocytes, which are important components of both the systemic inflammatory response and parasite control. The overall goal of this study was to define the role of monocytes during P. vivax malaria. Here, we demonstrate that P. vivax-infected patients display significant increase in circulating monocytes, which were defined as CD14(+)CD16- (classical), CD14(+)CD16(+) (inflammatory), and CD14loCD16(+) (patrolling) cells. While the classical and inflammatory monocytes were found to be the primary source of pro-inflammatory cytokines, the CD16(+) cells, in particular the CD14(+)CD16(+) monocytes, expressed the highest levels of activation markers, which included chemokine receptors and adhesion molecules. Morphologically, CD14(+) were distinguished from CD14lo monocytes by displaying larger and more active mitochondria. CD14(+)CD16(+) monocytes were more efficient in phagocytizing P. vivax-infected reticulocytes, which induced them to produce high levels of intracellular TNF-alpha and reactive oxygen species. Importantly, antibodies specific for ICAM-1, PECAM-1 or LFA-1 efficiently blocked the phagocytosis of infected reticulocytes by monocytes. Hence, our results provide key information on the mechanism by which CD14(+)CD16(+) cells control parasite burden, supporting the hypothesis that they play a role in resistance to P. vivax infection

    Phage resistance of acid lactic bacteria isolated from Coalho cheese industries

    Este trabalho teve como objetivos isolar bacteriófagos de amostras de leite, soro e queijo de Coalho e avaliar a resistência de cepas de Lactobacillus paracasei, pertencentes à Coleção de Micro-organismos de Interesse para a Agroindústria Tropical da Embrapa Agroindústria Tropical, aos fagos isolados. Posteriormente, a resistência destas cepas a fagos específicos para L. paracasei, da Coleção do Instituto de Lactología Industrial - INLAIN (Santa Fe, Argentina), também foi avaliada. As amostras para isolamento dos fagos foram obtidas em quatro unidades de processamento de queijo de Coalho, sendo duas artesanais e duas industriais, localizadas no Estado do Ceará. Para o isolamento dos bacteriófagos, foi empregado o teste de lise celular (spot), enquanto que a resistência das culturas aos fagos foi avaliada pelos testes de capacidade de produção de ácido e avaliação da turbidez. As cepas avaliadas foram resistentes aos bacteriófagos provenientes das unidades de processamento de queijo de Coalho e aos bacteriófagos da Coleção do INLAIN. Os resultados obtidos indicaram que as culturas láticas testadas, resistentes aos bacteriófagos, podem ser utilizadas na composição de fermento lático destinado à elaboração de queijo de Coalho, a partir de leite pasteurizado.The objectives of this research were to isolate bacteriophages from milk samples, whey and Coalho cheese and to evaluate the resistance of strains of Lactobacillus paracasei from the Collection of Microorganisms of Interest for the Tropical Agroindustry, belonging to Embrapa Tropical Agroindustry, to isolate phages. The strains resistance to specific L. paracasei phages from the collection of the Instituto de Lactolog a Industrial - INLAIN (Santa Fe, Argentina) was also evaluated. Samples for phage isolation were from four Coalho cheese processing units, two artisanal and two industrial, localized in the state of Ceara. Spot test was employed for bacteriophages isolation, while the culture phage resistance was evaluated by the acid production and turbidity tests. The strains were resistant both to phages isolated from Coalho cheese processing units and phages from collection of INLAIN. The results showed that lactic cultures evaluated here were resistant to bacteriophages and they can be used in the composition of lactic cultures specifically for Coalho cheese manufacture from pasteurized milk
