    Improved GNSS-R bi-static altimetry and independent DEMs of Greenland and Antarctica from TechDemoSat-1

    Improved Digital Elevation Models (DEMs) of the Antarctic and Greenland Ice Sheets are presented, derived from Global Navigation Satellite Systems-Reflectometry (GNSS-R). This builds on a previous study (Cartwright et al., 2018) using GNSS-R to derive an Antarctic DEM but uses improved processing and an additional 13 months of measurements, totalling 46 months of data from the UK TechDemoSat-1 satellite. A median bias of under 10 m and root-mean-square (RMS) errors of under 53 m for the Antarctic and 166 m for Greenland are obtained, as compared to existing DEMs. The results represent, compared to the earlier study, a halving of the median bias to 9 m, an improvement in coverage of 18 %, and a four times higher spatial resolution (now gridded at 25 km). In addition, these are the first published satellite altimetry measurements of the region surrounding the South Pole. Comparisons south of 88° S yield RMS errors of less than 33 m when compared to NASA’s Operation IceBridge measurements. Differences between DEMs are explored and the future potential for ice sheet monitoring by this technique is noted

    Multiview geometry is the study of two-dimensional images of three-dimensional scenes, a foundational subject in computer vision. We determine a universal Groebner basis for the multiview ideal of n generic cameras. As the cameras move, the multiview varieties vary in a family of dimension 11n-15. This family is the distinguished component of a multigraded Hilbert scheme with a unique Borel-fixed point. We present a combinatorial study of ideals lying on that Hilbert scheme.Comment: 26 page

    Muddles and puzzles: Metaphor use associated with disease progression in Primary Progressive Aphasia

    Background: Primary Progressive Aphasia describes a language-led dementia and its variants. There is little research exploring the experiences of living with this disease. Metaphor, words that represent something else, have been studied extensively in health-related narratives to gain a more intimate insight into health experiences. // Aims: This study explored the metaphors used spontaneously by people with PPA, their care partners (family), and speech and language therapists/pathologists (SLT/Ps) providing support along the continuum of care. // Methods & Procedures: This study examined two previously collected data sets comprising naturalistic talk where metaphors were not the specific focus, the first from focus groups conducted with people with PPA and their families and the second from focus groups conducted with SLT/Ps working with people with PPA. Transcribed data were analysed for metaphor use through an iterative narrative approach. // Outcomes & Results: In all, 237 examples of metaphorical language were identified in the data, with 14 metaphors from people with PPA, 116 from the families and 106 from SLT/Ps. Different metaphors were used by participants to describe their experiences depending on which variant of PPA they were living with, and people also described their disease differently over time. SLT/Ps also used metaphors, however, their language reflected the structured, professional perspective of delivering speech and language therapy services. // Conclusions & Implications: SLT/Ps should listen for and recognise the metaphorical language used by people with PPA and their families to ensure therapeutic alignment, see beyond the PPA to recognise the individual’s needs, and provide person-centred and empathic support

    Episodic, compression-driven fluid venting in layered sedimentary basins

    Fluid venting phenomena are prevalent in sedimentary basins globally. Offshore, these localised fluid-expulsion events are archived in the geologic record via the resulting pockmarks at the sea-floor. Venting is widely interpreted to occur via hydraulic fracturing, which requires near-lithostatic pore pressures for initiation. One common driver for these extreme pressures is horizontal tectonic compression, which pressurises the entire sedimentary column over a wide region. Fluid expulsion leads to a sudden, local relief of this pressure, which then gradually recharges through continued compression, leading to episodic venting. Pressure recharge will also occur through pressure diffusion from neighboring regions that remain pressurised, but the combined role of compression and pressure diffusion in episodic venting has not previously been considered. Here, we develop a novel poroelastic model for episodic, compression-driven venting. We show that compression and pressure diffusion together set the resulting venting period. We derive a simple analytical expression for this venting period, demonstrating that pressure diffusion can significantly reduce the venting period associated with a given rate of compression and allowing this rate of compression to be inferred from observations of episodic venting. Our results indicate that pressure diffusion is a major contributor to episodic fluid venting in mudstone-dominated basins

    Episodic fluid venting from sedimentary basins fuelled by pressurised mudstones

    Subsurface sandstone reservoirs sealed by overlying, low-permeability layers provide capacity for long-term sequestration of anthropogenic waste. Leakage can occur if reservoir pressures rise sufficiently to fracture the seal. Such pressures can be generated within the reservoir by vigorous injection of waste or, over thousands of years, by natural processes. In either case, the precise role of intercalated mudstones in the long-term evolution of reservoir pressure remains unclear; these layers have variously been viewed as seals, as pressure sinks or as pressure sources. Here, we use the geological record of episodic fluid venting in the Levant Basin to provide striking evidence for the pressure-source hypothesis. We use a Bayesian framework to combine recently published venting data, which record critical subsurface pressures since ∼\sim2 Ma, with a stochastic model of pressure evolution to infer a pressure-recharge rate of ∼\sim30 MPa/Myr. To explain this large rate, we quantify and compare a range of candidate mechanisms. We find that poroelastic pressure diffusion from mudstones provides the most plausible explanation for these observations, amplifying the ∼\sim1 MPa/Myr recharge caused by tectonic compression. Since pressurised mudstones are ubiquitous in sedimentary basins, pressure diffusion from mudstones is likely to promote seal failure globally

    Inhibition of cyclin-dependent kinase 9 downregulates cytokine production without detrimentally affecting human monocyte-derived macrophage viability

    Cyclin-dependent kinase (CDK) inhibitor drugs (CDKi), such as R-roscovitine and AT7519, induce neutrophil apoptosis in vitro and enhance the resolution of inflammation in a number of in vivo models. This class of compounds are potential novel therapeutic agents that could promote the resolution of acute and chronic inflammatory conditions where neutrophil activation contributes to tissue damage and aberrant tissue repair. In this study we investigated CDKi effects on macrophage pro-inflammatory mediator production and viability. Treatment of human monocyte-derived macrophages (MDMs) with the CDKi AT7519 and R-roscovitine at concentrations that induce neutrophil apoptosis had no significant effect on control or LPS-activated MDM apoptosis and viability, and did not detrimentally affect MDM efferocytosis of apoptotic cells. In addition, enhanced efferocytosis, induced by the glucocorticoid dexamethasone, was also unaffected after a short time treatment with R-roscovitine. Macrophage cytokine responses to inflammatory stimuli are also of importance during inflammation and resolution. As a key target of CDKi, CDK9, is involved in protein transcription via the RNA polymerase II complex, we investigated the effect of CDKi drugs on cytokine production. Our data show that treatment with AT7519 significantly downregulated expression and release of key MDM cytokines IL-6, TNF, IL-10 and IL-1β, as well as markers of pro-inflammatory macrophage polarisation. R-Roscovitine was also able to downregulate inflammatory cytokine protein secretion from MDMs. Using siRNA transfection, we demonstrate that genetic knock-down of CDK9 replicates these findings, reducing expression and release of pro-inflammatory cytokines. Furthermore, overexpression of CDK9 in THP-1 cells can promote a pro-inflammatory phenotype in these cells, suggesting that CDK9 plays an important role in the inflammatory phenotype of macrophages. Overall, this study demonstrates that pharmacological and genetic targeting of CDK9 inhibits an inflammatory phenotype in human MDMs. As such these data indicate that CDK9 may be key to therapeutically targeting pro-inflammatory macrophage functions during chronic inflammation

    Dilemmas and solutions- experiences of a national Family Medicine applied knowledge licensing test during a pandemic

    ABSTRACT: Background: The unprecedented COVID-19 pandemic brought significant challenges to all of medicine, including primary care training and examinations. The MRCGP AKT is high-stakes licensing 200-item MCQ for UK trainee family physicians and is part of an assessment tripos that, up to the onset of the pandemic, included a Clinical Skills Assessment using Simulated Patients and workplace based assessment. The AKT is blueprinted onto a curriculum content specification and computer delivered three times a year at test centres across the UK. It tests the knowledge base underpinning independent general practice within the context of the UK National Health Service. We report on the challenges and dilemmas faced during the pandemic, decisions taken, and lessons learned. Rapid exam changes needed to be made, and communicated effectively to candidates, whilst maintaining standards and fairness to candidates. Summary of Work: Challenges included lockdown travel restrictions, reduced capacity, social distancing and shielding candidates being unable to leave home. The April 2020 AKT was cancelled and prioritisation measures implemented to ensure candidates at the end of their training could enter the (stressed) workforce. We engaged with a wide range of stakeholders, carefully looked at remote testing, made contingency plans prioritised for those unable to sit exams and changed exam regulations to ensure fairness to candidates. In this emergency, we delivered a previously published exam which some candidates were unaware they had sat previously, and assessed how these candidates performed. We compared cohort performance before and during the pandemic. Summary of Results: We summarise why we did not remote test, how we obtained key worker status, and adapted contingency plans. Analysis of candidates who had previously sat the same exam showed they performed less well. Despite wide-ranging changes in training and workplace experience, there was no significant difference in cohort performance overall pre-and peri-pandemic. Discussion and Conclusions: COVID-19 constraints changed trainees clinical exposure, restricted training and supervisor support. However, exam preparedness did not appear adversely affected when measured by overall pass rates. Unexpectedly, candidates who sat an identical exam did not benefit from previous exposure. Take-home Messages: Involving stakeholders in key decisions and regular communications are essential. Test security and standards were not compromised
